Enzymatic coupling of specific peptides at nonspecific ligation sites: effect of Asp189Glu mutation in trypsin on substrate mimetic-mediated reactions

Two main drawbacks seriously restrict the synthetic value of proteases as reagents in peptide fragment coupling: (i) native proteolytic activity and, thus, risk of undesired peptide cleavage; (ii) limited enzyme specificities restricting the amino acid residues between which a peptide bond can be formed. While the latter can be overcome by the use of substrate mimetics achieving peptide bond formation at nonspecific ligation sites, the risk of proteolytic cleavage still remains and hinders the wide acceptance of this powerful strategy for peptide coupling. This paper reports on the effect of the trypsin point mutant Asp189Glu on substrate mimetic-mediated reactions. The effect of this mutation on the steady-state hydrolysis of substrate mimetics of the 4-guanidinophenyl ester type and on trypsin-specific Lys- and Arg-containing peptides was investigated. The results were confirmed by enzymatic coupling reactions using substrate mimetics as the acyl donor and specific amino acid-containing peptides as the acyl acceptor. The competition assay verifies the predicted shift in substrate preference from Lys and Arg to the substrate mimetics and, thus, from cleavage to synthesis of peptide bonds. The combination of results obtained qualifies the trypsin mutant D189E as the first substrate mimetic-specific peptide ligase.     

Distance-Balanced Graphs

Distance-balanced graphs are introduced as graphs in which every edge uv has the following property The number of vertices closer to u than to v is equal to the number of vertices closer to v than to u. Basic properties of these graphs are obtained. The new concept is connected with symmetry conditions in graphs and local operations on graphs are studied with respect to it. Distance-balanced Cartesian and lexicographic products of graphs are also characterized. Several open problems are posed along the way. Received August 31, 2005     

MRI description of knee medial collateral ligament abnormalities in the absence of trauma: edema related to osteoarthritis and medial meniscal tears

A retrospective case series regarding the knees of 12 adult patients with MRI abnormalities of the medial collateral ligament (MCL), but without clinical history of trauma to the MCL, were collected and compared with six knee MR images from patients with clinical traumatic injuries to their MCLs. The MR images were studied for the extent of edema of the MCL, as well as other associated findings. Edema of the MCL on MRI could be found in three distinct categories of patients: (a) those with trauma to the MCL, which was an expected finding; (b) those without trauma but with medial compartment osteoarthritis; and (c) those without trauma but with degenerative medial meniscal tears. The clinical significance, if any, of the edema found in MCLs without trauma remains unclear. Atraumatic MCL edema may serve as a marker for medial knee compartment osteoarthritis or for a degenerative medial meniscal tear and should not be confused with traumatically induced MCL injuries, although this distinction can often be difficult to make based on MRI findings alone.     

Cell enumeration and visualisation by transmission electron microscopy of Lactobacillus rhamnosus treated with cinnamon (Cinnamomum zeylanicum B.) essential oil

The use of essential oils (EOs) in functional foods containing probiotic microorganisms must consider the antimicrobial activity of these oils against beneficial bacteria such as Lactobacillus rhamnosus. This study aimed to evaluate the sensitivity of L. rhamnosus cultures treated with cinnamon EO through viable cell counts and visualisation by transmission electron microscopy. Cinnamon EO at a concentration of 0.04% had a bacteriostatic activity after 2?h of incubation. Although slight alterations were detected in the cell structure, this concentration was considered to be bactericidal, since it led to a significant reduction in cell numbers after 24?h. On the other hand, cinnamon EO at a 1.00% concentration decreased cell counts by 3 log units after 2?h incubation and no viable cell count was detected after 24?h. Transmission electron microscopy indicated that cells treated with 1.00% cinnamon EO were severely damaged and presented cell membrane disruption and cytoplasmic leakage.     

On Graphs Having Maximal Independent Sets of Exactly t Distinct Cardinalities

For a given positive integer t we consider graphs having maximal independent sets of precisely t distinct cardinalities and restrict our attention to those that have no vertices of degree one. In the situation when t is four or larger and the length of the shortest cycle is at least 6t ? 6, we completely characterize such graphs.
