Micellar electrokinetic capillary chromatography and reverse-phase liquid chromatography methods were developed in order to perform robustness testing to determine the caffeine content in beverages. Both methods were fully validated and two different robustness approaches were applied. One-variable-at-a-time (OVAT) approach at eleven levels (0; ±1; ±2; ±3; ±4; ±5 units) was carried out and compared with multi-variable-at-a-time (MVAT) approach at three levels (±1 unit per investigated parameter). Four analysts in two laboratories on two capillary electrophoresis and two RPLC equipments have tested the samples. Robustness was statistically analyzed using peak area, migration or retention time, symmetry, and resolution of caffeine and sulfacetamide as internal standard, and presented as R.S.D. values. The RPLC method was found to be more sensitive than the MEKC method. Both methods showed acceptable robustness level for OVAT approach, whilst MEKC method was more robust when the determination of real samples coming from different beverages was based on the MVAT approach.  相似文献

Diketopiperazines (DKPs) corresponding to cyclic dipeptides have been reported to exhibit antimicrobial, antitumor, antimutagenic and antiviral properties. These compounds are commonly isolated from microorganisms and sponges and from a variety of tissues and body fluids. In this work, we used electrospray ionization tandem mass spectrometry (ESI-MS/MS) to investigate the fragmentation of a series of DKPs previously isolated from Aspergillus fumigatus, which exhibit the same structural core. Loss of CO directly from the protonated molecule was found to be a fragmentation process common to all the compounds analyzed. However, our results revealed a series of ions that are diagnostic for the substituents at C(4) and C(9). In order to rationalize the differences in the fragmentation pathways of substituted and nonsubstituted DKPs, the relative Gibbs energies (DeltaG) of the product ions and intermediate ions were estimated using the B3LYP/6-31 + + G(d,p) model. The data reported here can be used for the structural elucidation of DKPs from low sample amounts, as an alternative to NMR.  相似文献

The magnetic anisotropy of epitaxial Fe films with thicknesses in the range of 2-142 monolayers (ML) grown on {4×2} reconstructed InAs(0 0 1) was investigated by in situ ferromagnetic resonance. The easy magnetization direction was found to be parallel to the -direction for Fe films below 4 ML, while it rotates by 45° toward the -direction. It is observed that both surface-interface and volume contribution to the perpendicular anisotropy favor an easy axis perpendicular to the film plane. The cubic surface-interface anisotropy is relatively large with easy axes along -directions in contrast to the volume contribution which favors easy axes along the -directions. The volume contribution is found to be larger than the Fe bulk cubic anisotropy. A thickness independent uniaxial anisotropy has been found in films with a thickness of 2 up to 142 ML.  相似文献

A class of two-parameter eigenvalue problems involving generally nonselfadjoint and unbounded operators is studied. A basis for the root subspace at a geometrically simple eigenvalue of Fredholm type is computed in terms of the underlying two-parameter system. Comparison with Faierman's work on two-parameter boundary value problems of Sturm-Liouville type is given as an application.

Liquid chromatographic method and micellar electrokinetic chromatographic method were optimized for determination of atrazine, desethylatrazine, desisopropylatrazine, hydroxyatrazine and their polar degradation products in solutions with humic acid without previous sample preparation step. Reversed-phase HPLC method was satisfactory in terms of repeatability and detection limits, which were ± 1.7–12.5% (RSD) and 0.1–0.5 mg L?1, respectively. However, the most polar products could not be separated from the front peak pertaining to humic acid. With MEKC, excellent separation of both chloro and hydroxy degradation products and parent compounds was achieved in a single analysis, and possible interferences of humic acid were successfully avoided by its retention at the anode. Drawbacks were detection limits, estimated to be 2–4 mg L?1, and RSD of the migration times was 20% compared to 0.5% with HPLC method. HPLC method was used to monitor degradation of atrazine and its first degradation products in the presence of humic acids, and MEKC was used for confirmation purposes.  相似文献

We provide analytical and numerical evidence that the classical mixing systems, which lack exponential sensitivity on initial conditions, exhibit universal decay of the Loschmidt echo which turns out to be a function of a single scaled time variable delta(2/5)t, where delta is the strength of perturbation. Proteomic analysis of human vitreous humor (VH) may elucidate the pathogenesis of retinal ocular diseases and may provide information for the development of potential therapeutic targets due to its pivotal location near lens and retina. The discovery of whole VH proteome involves a complex analysis of thousands of proteins simultaneously. Therefore, in proteomic studies the protein fractionation is important for reducing sample complexity, facilitating the access to the low‐abundant proteins, and recognizing them as biotargets for clinical research. Although several separation methods have been used, gel‐based proteomics are the most popular and versatile ones applied for global protein separation. However, chromatographic methods and its combination with other separation techniques are now beginning to be used as promising set‐ups for VH protein identification. This review attempts to offer an overview of the techniques currently used with VH, exploring its methodological demands, exposing its advantages, and helping the reader to plan future experiences. Moreover, this review shows the relevance of VH proteomic analysis as a tool for the study of the mechanisms underlying some ocular diseases and for the development of new therapeutic approaches.  相似文献

Grapes are rich in primary and secondary metabolites. Among the secondary metabolites, polyphenolic compounds are the most abundant in grape berries. Besides their important impacts on grape and wine quality, this class of compounds has beneficial effects on human health. Due to their antioxidant activity, polyphenols and phenolic acids can act as anti-inflammatory and anticancerogenic agents, and can modulate the immune system. In grape berries, polyphenols and phenolic acids can be located in the pericarp and seeds, but distribution differs considerably among these tissues. Although some classes of polyphenols and phenolic acids are under strict genetic control, the final content is highly influenced by environmental factors, such as climate, soil, vineyard, and management. This review aims to present the main classes of polyphenolic compounds and phenolic acids in different berry tissues and grape varieties and special emphasis on their beneficial effect on human health.  相似文献
