Mitochondrial metabolism particularly oxidative phosphorylation is greatly influenced by thyroid hormones. Earlier studies have described neuromuscular symptoms as well as impaired muscle metabolism in hypothyroid and hyperthyroid patients. In this study, we intend to look in to the muscle bioenergetics including phosphocreatine recovery kinetics based oxidative metabolism in thyroid dysfunction using in vivo 31P nuclear magnetic resonance spectroscopy (MRS). 31P MRS was carried out at resting state on 32 hypothyroid, 10 hyperthyroid patients and 25 control subjects. Nine out of 32 hypothyroid patients and 17 out of 25 control subjects under went exercise protocol for oxidative metabolism study and performed plantar flexion exercise while lying supine in 1.5 T magnetic resonance scanner using custom built exercise device. MRS measurements of inorganic phosphate (Pi), phosphocreatine (PCr), phosphodiesters (PDE) and adenosine triphosphate (ATP) of the calf muscle were acquired during rest, exercise and recovery phase. PCr recovery rate constant (kPCr) and oxidative capacity were calculated by monoexponential fit of PCr versus time (t) at the beginning of recovery. During resting condition in hypothyroid patients, PCr/Pi ratio was reduced whereas PDE/ATP and Pi/ATP were increased. However, in case of hyperthyroidism, an increased PCr/Pi ratio and reduced PDE/ATP and Pi/ATP were observed. The results confirmed differential energy status of the muscle due to increased or decreased levels of thyroid hormone. Our results also demonstrate reduced oxidative metabolism in hypothyroid patients based on PCr recovery kinetics. PCr recovery kinetics study after exercise revealed decreased PCr recovery rate constant (kPCr) in hypothyroid patients compared to controls that resulted in decrease in oxidative capacity of muscle by 50% in hypothyroids. These findings are consistent with a defect of high energy phosphate mitochondrial metabolism in thyroid dysfunction.  相似文献

The energy spectrum and dipole matrix elements of a multiple quantum well (MQW) system has been calculated numerically by solving the time-independent Schrödinger equation using finite difference method, in the presence of magnetic field. The effect of barrier width is also investigated. The energy difference between the levels of various minibands and the energy difference between various levels of the same miniband is calculated for different number of wells in the MQW. Finally, the dynamics of the system in short laser pulses has been calculated numerically by solving the time-dependent Schrödinger equation. The effect of magnetic field on the dynamics is clearly shown and explained.  相似文献

Background and purpose

Proton magnetic resonance spectroscopy (PMRS) has high sensitivity and specificity for the detection of pyogenic brain abscess and the categorization of bacteria. But the metabolite patterns failed to evaluate the etiology of disease when the culture results are sterile. The aim of the present study is to compare the multimodality techniques viz., conventional culture, MR spectroscopy and 16S rRNA PCR and sequencing for rapid diagnosis of etiology in brain abscess and evaluate the PMRS in culture sterile samples and also demonstrate the sensitivity and specificity of these techniques.

Methods

Thirty five patients underwent MRI on a 3 T MRI and in-vivo PMRS for the diagnosis and evaluation of various resonances of metabolites such as lipid (LIP), lactate (LAC), acetate (AC), amino acid (AC), succinate (SUC). Pus was collected for identification of etiologic agents by culture and molecular method.

Results

In 35 samples, metabolite patterns were as follows: LIP/LAC/AA, n = 17, LIP/LAC/AA/SUC with or without AC, n = 17 and LIP/LAC/AA/AC, n = 1. Culture showed bacterial growth in 22 samples (18 aerobic/facultative anaerobic, 9 anaerobic) whereas molecular method was detected 26 aerobic/facultative anaerobic, 13 anaerobic, 4 microaerophilic bacteria. Among the 13 sterile samples, molecular method detected 16 microorganisms along with 3 mixed infections and PMRS recognized metabolite patterns as LIP/LAC/AA, n = 5 and LIP/LAC/AA/SUC with or without AC, n = 8. The sensitivity of in-vivo PMRS in sterile samples was 100% and 75%, and specificity was 75% and 100% for aerobic and anaerobic organisms respectively.

Conclusion

Based on metabolite resonances, PMRS can detect slow growing and fastidious organisms and classify them into aerobic and anaerobic bacteria which are difficult to culture by conventional method. It can categorize microorganisms even in culture sterile samples with rational sensitivity and specificity which may allow early choice of targeted therapy.  相似文献

The electron-phonon interaction in the periodic Anderson model (PAM) is considered. The PAM incorporates the effect of onsite Coulomb interaction (U) between f-electrons. The influence of Coulomb correlation U on the phonon response of the system is studied by evaluating the phonon spectral function for various parameters of the model. The numerical evaluation of the spectral function is carried out in the long wavelength limit at finite temperatures keeping only linear terms in U. The observed behaviour is found to agree well with the general features obtained experimentally for some heavy fermion (HF) systems.  相似文献

In this study, we investigate the expansion of the FRLW universe in the open, closed, and flat geometries. The universe is dominated by a scalar field (spatially homogeneous) as a source of dark energy. We consider the three different classes of scalar fields – quintessence, tachyonic, and phantom field – for our analysis. A mathematical analysis is carried out by considering these three scalar fields with exponential and power-law potentials. Both potentials give exponential expansion in the open, closed, and flat FRLW universes. It is found that quintessence, tachyonic, and phantom scalar fields are indistinguishable under the slow roll approximation.  相似文献

A statistical analysis has been carried out to determine the configurational preference of a pair of 4-cyanophenyl 4-n-pentylbenzoate (CPPB) molecules with respect to translatory and orientational motions. The CNDO/2 method has been employed to evaluate the net atomic charge and atomic dipole components at each atomic centre of the molecule. Configurational energy has been computed using the Rayleigh—Schodinger perturbation method. The total interaction energy values obtained through these computations were used to calculate the probability of each configuration at the phase transition temperature using the Maxwell—Boltzmann formula. An attempt has been made to identify the most probable configuration at the phase transition temperature. Further, the flexibility of various configurations has been studied in terms of variation of probability due to small departures from the most probable configuration. On the basis of stacking, in-plane and terminal interaction energy calculations, all possible geometrical arrangements of the molecular pair have been considered. The results are discussed in the light of experimental as well as theoretical observations. The nature of the mesophase has been correlated with the parameter introduced in this paper.  相似文献

The Fe3Al alloy with D03 structure exhibits large recoverable strains due to reversible slips. Tension and compression experiments were conducted on single crystals of Fe3Al, and the onset of slip in forward and reverse directions were obtained utilizing high-resolution digital image correlation technique. The back stress provides the driving force for reversal of deformation upon unloading, resulting in a superelastic phenomenon as in shape memory alloys. Using density functional theory simulations, we obtain the energy barriers (GSFE – generalized stacking fault energy) for {1?1?0}〈1?1?1〉 and {1?1?2}〈1?1?1〉 slips in D03 Fe3Al and the elastic moduli tensor, and undertake anisotropic continuum calculations to obtain the back stress and the frictional stress responsible for reversible slip. We compare the theoretically obtained slip stress magnitudes (friction and back stress) with the experimental measurements disclosing excellent agreement.  相似文献

Chikungunya is an infectious disease caused by mosquito-transmitted chikungunya virus (CHIKV). It was reported that NS1 and E2 siRNAs administration demonstrated CHIKV inhibition in in vitro as well as in vivo systems. Cationic lipids are promising for designing safe non-viral vectors and are beneficial in treating chikungunya. In this study, nanodelivery systems (hybrid polymeric/solid lipid nanoparticles) using cationic lipids (stearylamine, C9 lipid, and dioctadecylamine) and polymers (branched PEI-g-PEG -PEG) were prepared, characterized, and complexed with siRNA. The four developed delivery systems (F1, F2, F3, and F4) were assessed for stability and potential toxicities against CHIKV. In comparison to the other nanodelivery systems, F4 containing stearylamine (Octadecylamine; ODA), with an induced optimum cationic charge of 45.7 mV in the range of 152.1 nm, allowed maximum siRNA complexation, better stability, and higher transfection, with strong inhibition against the E2 and NS1 genes of CHIKV. The study concludes that cationic lipid-like ODA with ease of synthesis and characterization showed maximum complexation by structural condensation of siRNA owing to high transfection alone. Synergistic inhibition of CHIKV along with siRNA was demonstrated in both in vitro and in vivo models. Therefore, ODA-based cationic lipid nanoparticles can be explored as safe, potent, and efficient nonviral vectors overcoming siRNA in vivo complexities against chikungunya.  相似文献

The structural, electronic and optical properties of HgAl2Se4 are investigated using the full potential linear augmented plane wave method based on density functional theory. The calculated structural parameters using LDA are in excellent agreement with the available experimental result. The obtained energy band gap (2.24 eV) using EV-GGA approximation is in excellent agreement with experimental data (2.20 eV). Variation in the energy band gap as a function of the unit cell lattice parameter has been studied. The optical properties show a considerable anisotropy, which makes this compound very useful for various linear–nonlinear optical devices.  相似文献

Zeolites have taken a leading position in petrochemical, fine, and bulk chemical industries due to their porous architecture, pore sizes, tunable acidity, and thermal stability. Various strategies of zeolites preparation, including template‐free, solvent‐free, and toxic mineral‐free strategies are summarized. Moreover, the zeolite synthesis using naturally occurring minerals and sustainable natural templates is also discussed, which involves the synthesis of nanocrystalline zeolites of different framework structures using plant‐based natural templates and biomass‐derived renewable chemicals. Overall this personal account provides the fundamentals of various sustainable synthetic strategies reported in the literature for the synthesis of zeolites with suitable examples that will be useful for the students and will motivate experienced researchers to develop various novel sustainable methods for the synthesis of zeolites and other inorganic materials of industrial relevance.  相似文献
