In view of the pharmacological importance of dietary fibre, psyllium, to cure the constipation and diverticulitis, in the present study, an attempt has been made to modify psyllium polysaccharide with PVP to develop the hydrogel meant for slow and controlled drug delivery systems. The polymer was characterized by SEMs, FTIR, XRD, TGA and swelling studies. Swelling of hydrogels and drug (ciprofloxacin) release profile from the drug loaded hydrogels were determined for the evaluation of the swelling/release mechanism. Biomedical properties; biocompatibility and mucoadhesion of the hydrogels, were also studied. Swelling of the hydrogels and release of drugs from drug loaded hydrogels occurred through non-Fickian diffusion mechanism. Here it is pertinent to mention that both psyllium husk polysaccharide and antibiotic drug ciprofloxacin are used for gastrointestinal tract (GIT) problem, especially in case of diverticulitis. A highly water‐soluble phenothiazine (PTZ)–boron dipyrromethene (BODIPY)‐based electron donor–acceptor dyad ( WS‐Probe ), which contains BODIPY as the signaling antennae and PTZ as the OCl? reactive group, was designed and used as a fluorescent chemosensor for the detection of OCl?. Upon addition of incremental amounts of NaOCl, the quenched fluorescence of WS‐Probe was enhanced drastically, which indicated the inhibition of reductive photoinduced electron transfer (PET) from PTZ to 1BODIPY*; the detection limit was calculated to be 26.7 nm . Selectivity studies with various reactive oxygen species, cations, and anions revealed that WS‐Probe was able to detect OCl? selectively. Steady‐state fluorescence studies performed at varied pH suggested that WS‐Probe can detect NaOCl and exhibits maximum fluorescence in the pH range of 7 to 8, similar to physiological conditions. ESI‐MS analysis and 1H NMR spectroscopy titrations showed the formation of sulfoxide as the major oxidized product upon addition of hypochlorite. More interestingly, when WS‐Probe was treated with real water samples, the fluorescence response was clearly visible with tap water and disinfectant, which indicated the presence of OCl? in these samples. The in vitro cell viability assay performed with human embryonic kidney 293 (HEK 293) cells suggested that WS‐probe is non‐toxic up to 10 μm and implicates the use of the probe for biological applications. 相似文献

QSAR has been used to elucidate the origin of the hydrophobicity and binding affinity of a small library of fluoroaromatic inhibitors of F131V carbonic anhydrase II. Our analysis predicted the presence of a twisted amide conformation for several bound inhibitors, which we confirmed crystallographically. We also determined that the hydrophobicity of the inhibitors as a whole results from the fragment hydrophobicities of their fluorobenzyl rings, corrected for field effects and the presence of an intramolecular F.H contact in solution. The loss of this interaction on binding to the enzyme makes the affinity sensitive to the same terms, but with the opposite dependence on the F.H contact. In the case of the four inhibitors bound as twisted amides, this F.H contact must be retained to some extent in the bound state in order for their affinities to be consistent with our QSAR analysis of the entire set of 17 molecules. 相似文献
