In all organisms, genetic information is stored in DNA and RNA. Both of these macromolecules are damaged by many exogenous and endogenous events, with UV irradiation being one of the major sources of damage. The major photolesions formed are the cyclobutane pyrimidine dimers (CPD), pyrimidine-pyrimidone-(6-4)-photoproducts, Dewar valence isomers and, for dehydrated spore DNA, 5-(α-thyminyl)-5,6-dihydrothymine (SP). In order to be able to investigate how nature's repair and tolerance mechanisms protect the integrity of genetic information, oligonucleotides containing sequence and site-specific UV lesions are essential. This tutorial review provides an overview of synthetic procedures by which these oligonucleotides can be generated, either through phosphoramidite chemistry or direct irradiation of DNA. Moreover, a brief summary on their usage in analysing repair and tolerance processes as well as their biological effects is provided. 相似文献

A high-precision Penning trap mass measurement of the exotic 8He nuclide (T(1/2)=119 ms) has been carried out resulting in a reduction of the uncertainty of the halo binding energy by over an order of magnitude. The new mass, determined with a relative uncertainty of 9.2 x 10(-8) (deltam=690 eV) is 13 keV less bound than the previously accepted value. The mass measurement is of great relevance for the recent charge-radius measurement of 8He [P. Mueller, Phys. Rev. Lett. 99, 252501 (2007).10.1103/PhysRevLett.99.252501]. Sensitivity of drag to tangential surface motion is calculated in flow past a circular cylinder in both two- and three-dimensional conditions at Reynolds number \(\textit{Re} \le 1000\). The magnitude of the sensitivity maximises in the region slightly upstream of the separation points where the contour lines of spanwise vorticity are normal to the cylinder surface. A control to reduce drag can be obtained by (negatively) scaling the sensitivity. The high correlation of sensitivities of controlled and uncontrolled flow indicates that the scaled sensitivity is a good approximation of the nonlinear optimal control. It is validated through direct numerical simulations that the linear range of the steady control is much higher than the unsteady control, which synchronises the vortex shedding and induces lock-in effects. The steady control injects angular momentum into the separating boundary layer, stabilises the flow and increases the base pressure significantly. At \(\textit{Re}=100\), when the maximum tangential motion reaches 50% of the free-stream velocity, the vortex shedding, boundary-layer separation and recirculation bubbles are eliminated and 32% of the drag is reduced. When the maximum tangential motion reaches 2.5 times of the free-stream velocity, thrust is generated and the power savings ratio, defined as the ratio of the reduced drag power to the control input power, reaches 19.6. The mechanism of drag reduction is attributed to the change of the radial gradient of spanwise vorticity (\(\partial _{r} \hat{\zeta }\)) and the subsequent accelerated pressure recovery from the uncontrolled separation points to the rear stagnation point. 相似文献

The fast iterative solution of optimal control problems, and in particular PDE-constrained optimization problems, has become an active area of research in applied mathematics and numerical analysis. In this paper, we consider the solution of a class of time-dependent PDE-constrained optimization problems, specifically the distributed control of the heat equation. We develop a strategy to approximate the (1, 1)-block and Schur complement of the saddle point system that results from solving this problem, and therefore derive a block diagonal preconditioner to be used within the MINRES algorithm.
