A novel, sensitive and specific method for the quantification of alpha-tocopherol in two infant foods (milk and cereals) using liquid chromatography on-line with positive atmospheric pressure chemical ionisation mass spectrometry detection (LC/APCI-MS) has been developed. The samples were first saponified in order to eliminate fats and to transform tocopherol esters into free tocopherol, followed up by a liquid-liquid extraction of the analyte in petroleum benzine/diisopropyl ether (75:25, v/v) prior to injection onto the LC system. For the quantification, deuterium-labelled tocopherol was used as internal standard and the samples were monitored in selected ion monitoring (SIM) mode. Calibration curves between 1-40 microg/mL of alpha-tocopherol showed a good linear correlation (r(2) = 0.99994), and the detection limit was determined to be 2.5 ng/mL. The within-day and between-day precision were determined for several dietetic infant formulae and certified reference samples, and found to be below 3.5%. The accuracy determined on a Nestlé reference sample (milk powder) was calculated to be 115.2 +/- 1.2%, which confirms the robustness of the proposed method. This study shows that single quadrupole LC/MS can be applied for the quantification of vitamins in food and the method offers better sensitivity and selectivity than traditional method such as LC-UV. This would simplify the preparation of the food samples and consequently enhance the vitamin analysis throughput in the food area.  相似文献

Chiral p-tert-butylcalix[4]arenes functionalized at the lower rim with amino acid residues have been prepared. The (1)H and (13)C NMR spectra indicate that the macrocycles preferably adopt a cone conformation. Calix[4]arenes bearing amino acid moieties were prepared as a class of receptors selective for anions that are bound through hydrogen bonding with the NH group. The association constants are dependent on the nature of the substituents at the lower rim. Derivative 9 shows the strongest complexation and the largest selectivity for N-tosyl-(L)-alaninate. Finally, a preliminary X-ray crystal study of the difunctionalized receptor 6f shows the "flattened cone" conformation in the solid state.  相似文献

Polyfluorene (PFO) embedded in a nematic liquid crystal (LC) matrix is investigated. For low PFO weight contents, a homogeneous dispersion is obtained which displays a strong fluorescence anisotropy along the LC director, indicating a significant alignment of the polymeric chains along this direction. Besides, for relatively high PFO weight contents, phase separation takes place. Under these conditions, the sample is composed of micrometer‐sized domains, where the two species are in solution, enclosed by segregated polymeric boundaries. By polarized‐photoluminescence imaging and spectroscopy, it is found that most of the light emission originates from these boundaries and gets strongly pinned along their orientation. Since boundaries are mainly oriented orthogonal to the LC chains, this morphological alignment results in a system in which the orientation of the polarization emission can be predicted and possibly controlled. Conversely, in the homogeneous sample one can obtain a homogeneous emission polarization by controlling the alignment of the LC. The preparation of 2-acyl-6,7-dimethoxy-1,2,3,4-tetrahydro-1-isoquinolinecarboxamides is described. The mixed anhydride of 2-acyl-6,7-dimethoxy-1,2,3,4-tetrahydro-1-isoquinolinecarboxylic acid with isobutyl chloroformate was reacted with the corresponding hydrazines at — 10, — 15°. Besides those compounds, isoquinolones occur through a new side reaction. The structure of those products was established by 13C and 1H nmr and mass spectroscopy. A possible mechanism of the reaction is discussed.  相似文献

Single crystals of Bi(C2O4)OH were obtained by the slow diffusion of Bi3+ cations through silica gel impregnated with oxalic acid. The structure was solved in the Pnma space group with a=6.0853(2) Å, b=11.4479(3) Å, c=5.9722(2) Å, leading to R=0.0188 and wR=0.0190 from 513 unique reflections. The bismuth coordination polyhedron is a BiO6E pentagonal bipyramid with the lone pair E sitting at an axial vertex. The opposite axial vertex is occupied by a hydroxyl oxygen atom, which is also an equatorial corner of a neighboring BiO6E bipyramid. The sharing of the hydroxyl oxygen atoms build zig-zag chains running down the [100] direction. These chains are aligned in a sheet parallel to the (010) plane and are further connected through oxalate ions to form a three-dimensional arrangement. On heating, Bi(C2O4)OH decomposes to the meta-stable quadratic β-Bi2O3 phase.  相似文献

Three new cyclopropanic oleanane triterpenoids and ten known compounds were obtained from Donella ubanguiensis using chromatographic methods. The structures were established on the basis of mass spectrometric and NMR data and by comparison with values reported in the literature. The structures of the new compounds were confirmed by X-ray crystallography. A part of the isolated compounds was evaluated for cytotoxic and antimicrobial activities.  相似文献

An in-depth study of the cobalt-catalyzed [2+2+2] cycloaddition between yne-ynamides and nitriles to afford aminopyridines has been carried out. About 30 nitriles exhibiting a broad range of steric demand and electronic properties have been evaluated, some of which open new perspectives in metal-catalyzed arene formation. In particular, the use of [CpCo(CO)(dmfu)] (dmfu=dimethyl fumarate) as a precatalyst made possible the incorporation of electron-deficient nitriles into the pyridine core. Modification of the substitution pattern at the yne-ynamide allows the regioselectivity to be switched toward 3- or 4-aminopyridines. Application of this synthetic methodology to the construction of the aminopyridone framework using a yne-ynamide and an isocyanate was also briefly examined. DFT computations suggest that 3-aminopyridines are formed by formal [4+2] cycloaddition between the nitrile and the intermediate cobaltacyclopentadiene, whereas 4-aminopyridines arise from an insertion pathway.  相似文献

An immunochromatographic assay using nitrocellulose membrane was combined with electrochemical detection using an electrode chip in order to quantitatively detect testosterone as a model analyte. The electrode chip consisted of a gold working electrode, a counter electrode and a pseudo-reference electrode, all fabricated on the bottom of a 3.2 mm × 3.2 mm well. Competitive immunoreactions on the membrane were initiated by flowing a solution containing testosterone and horseradish peroxidase (HRP)-labeled testosterone (a competitor) over the membrane. Prepared membrane was placed in a solution containing ferrocenemethanol (FcOH) and H2O2 in the well of the electrode chip, and the enzyme reaction was detected by amperometry. Labeled HRP captured on the membrane catalyzed the oxidation of FcOH to the oxidized form FcOH+, which was reduced electrochemically by the electrode chip. The electrochemical response of the reduction current decreased with increasing concentration of testosterone over the range 1–625 ng/ml.  相似文献
