A rapid, sensitive, and reproducible pre-column derivatisation procedure has been established for the simultaneous determination of 20 amino acids by high-performance liquid chromatography using fluorescence detection. The amino acids were derivatized using o-phthalaldehyde and 9-fluorenylmethyl-chloroformate reagents. The optimal conditions for simultaneous separation and detection of both primary and secondary amino acids were investigated. The developed method has several advantages, namely automated pre-column derivatization, short analysis time with optimal separation, a simple and economical mobile phase, high level of precision for peak area and retention time, and higher sensitivity with more reliability of peak identification. The biological media development is the key parameter for macromolecule drug discovery. Biological media amino acids in three consecutive discovery batches were determined and the results showed a good agreement with hypothetical value. The method appears suitable for application to measure biological media amino acids at various stages of macromolecule drug discovery.  相似文献

The reaction of a Cu(II)-nitrosyl complex (1) with hydrogen peroxide at -20 °C in acetonitrile results in the formation of the corresponding Cu(I)-peroxynitrite intermediate. The reduction of the Cu(II) center was monitored by UV-visible spectroscopic studies. Formation of the peroxynitrite intermediate has been confirmed by its characteristic phenol ring nitration reaction as well as isolation of corresponding Cu(I)-nitrate (2). On air oxidation, 2 resulted in the corresponding Cu(II)-nitrate (3). Thus, these results demonstrate a possible decomposition pathway for H(2)O(2) and NO through the formation of a peroxynitrite intermediate in biological systems.  相似文献

Cu(II) complexes of three tridentate ligands, L(1), L(2) and L(3), [L(1), N-((1H-imidazole-2-yl)methyl)-2-(pyridine-2-yl)ethanamine; L(2), N-((1-methyl-1H-imidazole-2-yl)methyl)-2-(pyridine-2-yl)ethanamine; L(3), 2-(pyridine-2-yl)-N-((pyridine-2-yl)methyl)ethanamine] respectively, were synthesized and characterized. The single crystal X-ray structure of complex 1 reveals the pseudo octahedral coordination geometry around the copper center. Absorption and fluorescence experimental evidence show good DNA binding propensity (in the order of 10(5) M(-1)) of the complexes. Thermal denaturation and circular dichroism (CD) analyses reveal minor structural changes of calf thymus (CT) DNA in presence of complexes and groove and/or surface binding of the complexes to CT-DNA. Kinetic DNA cleavage assay shows pseudo-first-order kinetic reaction between the complex and supercoiled (SC) DNA. In addition, mechanistic SC DNA cleavage results show higher DNA cleavage activity in presence of reducing agent, due to the presence of hydroxyl radicals. In vitro cytotoxicity assay of the complexes demonstrate that the complexes have low toxicity for different cancer cell lines and IC(50) values were between 37 and 156 μM.  相似文献

Abstract

MnFe2O4/(Pb0.8Sr0.2)TiO3 (MFO/PST20) heterostructured composite films with three different structures have been grown on Pt/TiO2/SiO2/Si substrates by metal–organic decomposition processing via spin coating technique. The structural analysis revealed that the crystal axes of the MnFe2O4 are aligned with those of the PST20 ferroelectric matrix with obvious interfaces and no diffusions exist in all the three composite films. These composite films exhibit simultaneously multiferroic and magnetoelectric responses at room temperature. The growth structure of MFO and PST20 layers has an effect on multiferroic and magnetoelectric coupling behaviours of the composite films. The bi- and four-layered MFO/PST20 composite films exhibit superior ferroelectric properties compared to the tri-layered film. The increasing MFO and PST20 layers in the composite films enhance ferromagnetic properties and are closely related to the strain release in MnFe2O4 phase. The MFO/PST20 bi-layered composite film shows a high magnetoelectric voltage co-efficient αE ~ 194 mVcm?1Oe?1 at a dc magnetic field Hdc ~ 2.5 kOe. A significant decrease in αE value has been observed for tri- and four- layered composite films. A close correlation between phase selective residual stress and magnetoelectric properties has been emerged. The results are reasonably encouraging for employing MnFe2O4 for growing multiferroic–magnetoelectric composite films.  相似文献

We have investigated the properties of liquid crystals (LCs) doped with ZnO (8% Cu doped) nanoparticles. The electro-optic properties of LCs have changed with varying concentration of ZnO nanoparticles. The dielectric anisotropy obtained from the values of dielectric permittivity at 5 kHz in the nematic and smectic phases was found to increase with increasing concentration of nanoparticles in LCs. It has been established that the effect of nanoparticles on the dielectric anisotropy depends on the physical properties of LCs; the nanoparticle disturbs the orientation ordering of LC molecules. The nanoparticle also influences the switching behavior, splay elastic constant, rotational viscosity and threshold voltage of pure LCs. A small quantity of nanoparticles causes slight reduction of the splay elastic constant and rotational viscosity of LC cells.  相似文献

This paper presents a procedure to solve a chance constraint programming problem with sum-of-fractional objectives. The problem and the solution procedure are described with the help of a practical problem – assembled printed circuit boards (PCBs). Errors occurring during assembling PCBs are in general classified into three kinds, viz. machine errors, manual errors and other errors. These errors may lead to the rejection of the major portion of the production and hence result the manufacturer a huge loss. The problem is decomposed to have two objective functions; one is a sum-of-fractional objectives and the other is a non-linear objective with bounded constraints. The interest is to maximize the sum-of-fractional objectives and to minimize the non-linear objective, subject to stochastic and non-stochastic bounded environment. The first problem deals with the maximization of the profit (maximizing sum-of-fractional objectives) and the second deals with the minimization of the loss (errors), so as to obtain the maximum profit after removing the number of defective PCBs.  相似文献

This communication describes GC-MS analysis of bis(trimethylsilyl) (bis-TMS) derivatives of bis(2-hydroxyethylthio)alkanes (BHETAs) and bis(2-hydroxyethylsulfonyl) alkanes (BHESAs) which are important markers of sulfur mustard class of chemical warfare agents. The study was undertaken with a view to develop spectral database of these compounds for verification analysis of Chemical Weapons Convention (CWC). Based on the obtained mass spectra of bis-TMS derivatives of BHETAs and BHESAs, the fragmentation routes are proposed, which explain most of the characteristic ions.  相似文献
