Synthesis of 1-azabicyclo[3.3.0]octane derivatives and their effects as piracetam-like nootropics

A useful pharmaceutical intermediate, 5-nitromethyl-1-azabicyclo[3.3.0]octane (1), was prepared in one step from 1,7-dichloro-4-heptanone (4) under mild conditions. Catalytic hydrogenation of 1 over Raney Ni in the presence of sodium hydroxide afforded 5-aminomethyl-1-azabicyclo[3.3.0]octane (2) in high yield. Piracetam analogues 20-23, which were pyrrolidine derivatives involving a 1-azabicyclo[3.3.0]octane ring, were synthesized. Pharmacological tests showed that N-[(1-azabicyclo[3.3.0]octan-5-yl)methyl]-2-oxo-1-pyrrolidineacetamid e (20) improves cerebral function.     

Application of ion-exchange cartridge clean-up in food analysis I. Simultaneous determination of thiabendazole and imazalil in citrus fruit and banana using high-performance liquid chromatography with ultraviolet detection

A simple, rapid and reproducible analytical method for thiabendazole (TBZ) and imazalil (IMA) in citrus fruit and banana has been developed. The method involves the use of an ion-exchange cartridge for sample clean-up followed by ion-pair high-performance liquid chromatography with ultraviolet detection. The recoveries of TBZ and IMA from citrus fruits spiked at levels of 10 μg/g and 5 μg/g were in the range of 94–98% and 93–98% with coefficients of variation of 0.5–2.2% and 1.6–2.7%, respectively. The recoveries of TBZ and IMA from banana spiked at levels of 3 μg/g and 2 μg/g were 94% and 94% with coefficients of variation 1.1% and 4.9%, respectively. The detection limits for TBZ and IMA were 0.1 μg/g in citrus fruit and 0.05 μg/g in banana.     

Comparative Study of Structure of Silica Gels from Different Sources

The medium-range-network structure of silicas from different sources was comparatively examined by X-ray diffraction, IR and NMR analyses. Silica gels made through the conventional sol-gel route were found to have a different structure from silica glass, and considered to be composed of 4-fold siloxane ring units, irrespective of preparation conditions. Amorphous silicas in rice-hulls and bamboo leaves showed structure data close to silica glass, being suggested to consist mainly of 6-fold siloxane ring units. The modified sol-gel method, in which siloxane oligomers containing cyclic siloxane tetramers may have been ring-opened and polymerized under the catalytic reaction of NaOH or NH₄F in the water-free solution, provided the silica gel of which structure was very similar to silica glass.     

Separation of WAP-8294A components, a novel anti-methicillin-resistant staphylococcus aureus antibiotic, using high-speed counter-current chromatography.

The WAP-8294A complex was isolated from the fermentation broth of Lysobacter sp. WAP-8294, whose major component, WAP-8294A2, showed a strong activity against Gram-positive bacteria including methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant Enterococci in vitro, and also exhibited a potent activity against MRSA in vivo. The previous separation procedure using the conventional chromatographic methods was laborious and time-consuming, and the recovery of the desired compound was often unsatisfactory. In the present study, high-speed counter-current chromatography (HSCCC) was applied to the separation of the main components of the WAP-8294A complex. Due to the high polarity of the target compounds, we selected a hydrophilic two-phase solvent system composed of n-butanol-ethyl acetate-aqueous 0.005 M trifluoroacetic acid (1.25:3.75:5, v/v/v) which provided a suitable range of partition coefficient values for these compounds. Although the settling time of this solvent system was much longer than the optimum range, suggesting a low retention of the stationary phase under the standard experimental conditions, the separation was successfully performed at the low flow-rate of 0.5 ml/min. A sample size of 25 mg yielded pure fractions of three components (1-6 mg). The identification of each component was carried out by HPLC and fast atom bombardment mass spectrometry. The method will contribute to the clinical development of WAP-8294A2 as an anti-MRSA agent.     

Ground-state decay rate for the Zener breakdown in band and Mott insulators

Nonlinear transport of electrons in strong electric fields, as typified by dielectric breakdown, is reformulated in terms of the ground-state decay rate originally studied by Schwinger in nonlinear QED. We discuss the effect of electron interaction on Zener tunneling by comparing the dielectric breakdown of the band insulator and the Mott insulator, where the latter is studied by the time-dependent density-matrix renormalization group. The relation with the Berry's phase theory of polarization is also established.     

Evolution operators generated by non‐densely defined operators

In this paper it is shown that an evolution operator is generated by a family of closed linear operators whose common domain is not necessarily dense in the underlying Banach space, under the stability condition proposed by the second author from the viewpoint of finite difference approximations.
