3-Allyl- and 3-prop-1-ynylsulfanyl-2-arylazo-3-cycloalkylamino-acrylonitriles undergo cyclization under mild conditions to afford the novel heterocyclic systems 1,4,6,7,8,8a-hexahydropyrrolo[2,1-c][1,2,4]-triazine-4-thione, 1,4,6,7,9,9a-hexahydro-[1,4]oxazino[3,4-c][1,2,4]triazine and 1,6,7,8,9,9a-hexahydro-4H-pyrido[2,1-c][1,2,4]triazine via a number of consecutive pericyclic reactions.  相似文献

Fhit, the product of tumor suppressor fragile histidine triad (FHIT) gene, exhibits antitumor activity of still largely unknown cellular background. However, it is believed that Fhit-Ap(3)A or Fhit-AMP complex might act as a second class messenger in cellular signal transduction pathway involved in cell proliferation and apoptosis. We demonstrate here for the first time that the photosensitizer, protoporphyrin IX (which is a natural precursor of heme) binds to Fhit protein and its mutants in the active site in vitro. Furthermore, PpIX inhibits the enzymatic activity of Fhit. Simultaneously, PpIX shows lower binding capacity to mutant Fhit-H96N of highly reduced hydrolase activity. In cell-based assay PpIX induced HeLa cell death in Fhit and Fhit-H96N-dependent manner which was measured by means of MTT assay. Moreover, HeLa cells stably expressing Fhit or mutant Fhit-H96N were more susceptible to protoporphyrin IX-mediated photodynamic therapy (2J/cm(2)) than parental cells.  相似文献

Asymmetric cross-coupling of aryl iodides (ArI) with secondary arylphosphines (PHMe(Ar'), Ar' = (2,4,6)-R3C6H2; R = i-Pr (Is), Me (Mes), Ph (Phes)) in the presence of the base NaOSiMe3 and a chiral Pd catalyst precursor, such as Pd((R,R)-Me-Duphos)(trans-stilbene), gave the tertiary phosphines PMe(Ar')(Ar) in enantioenriched form. Sterically demanding secondary phosphine substituents (Ar') and aryl iodides with electron-donating para substituents resulted in the highest enantiomeric excess, up to 88%. Phosphination of ortho-substituted aryl iodides required a Pd(Et-FerroTANE) catalyst but gave low enantioselectivity. Observations during catalysis and stoichiometric studies of the individual steps suggested a mechanism for the cross-coupling of PhI and PHMe(Is) (1) initiated by oxidative addition to Pd(0) yielding Pd((R,R)-Me-Duphos)(Ph)(I) (3). Reversible displacement of iodide by PHMe(Is) gave the cation [Pd((R,R)-Me-Duphos)(Ph)(PHMe(Is))][I] (4), which was isolated as the triflate salt and crystallographically characterized. Deprotonation of 4-OTf with NaOSiMe3 gave the phosphido complex Pd((R,R)-Me-Duphos)(Ph)(PMeIs) (5); an equilibrium between its diastereomers was observed by low-temperature NMR spectroscopy. Reductive elimination of 5 yielded different products depending on the conditions. In the absence of a trap, the unstable three-coordinate phosphine complex Pd((R,R)-Me-Duphos)(PMeIs(Ph)) (6) was formed. Decomposition of 5 in the presence of PhI gave PMeIs(Ph) (2) and regenerated 3, while trapping with phosphine 1 during catalysis gave Pd((R,R)-Me-Duphos)(PHMe(Is))2 (7), which reacted with PhI to give 3. Deprotonation of 1:1 or 1.4:1 mixtures of cations 4-OTf gave the same 6:1 ratio of enantiomers of PMeIs(Ph) (2), suggesting that the rate of P inversion in 5 was greater than or equal to the rate of reductive elimination. Kinetic studies of the first-order reductive elimination of 5 were consistent with a Curtin-Hammett-Winstein-Holness (CHWH) scheme, in which pyramidal inversion at the phosphido ligand was much faster than P-C bond formation. The absolute configuration of the phosphine (SP)-PMeIs(p-MeOC6H4) was determined crystallographically; NMR studies and comparison to the stable complex 5-Pt were consistent with an RP-phosphido ligand in the major diastereomer of the intermediate Pd((R,R)-Me-Duphos)(Ph)(PMeIs) (5). Therefore, the favored enantiomer of phosphine 2 appeared to be formed from the major diastereomer of phosphido intermediate 5, although the minor intermediate diastereomer underwent P-C bond formation about three times more rapidly. The effects of the diphosphine ligand, the phosphido substituents, and the aryl group on the ratio of diastereomers of the phosphido intermediates Pd(diphos*)(Ar)(PMeAr'), their rates of reductive elimination, and the formation of three-coordinate complexes were probed by low-temperature 31P NMR spectroscopy; the results were also consistent with the CHWH scheme.  相似文献

We report the efficient aqueous dispersion of pristine HiPco single-walled carbon nanotubes (SWNTs) with ionic liquid (IL)-based surfactants 1-dodecyl-3-methylimidazolium bromide (1) and 1-(12-mercaptododecyl)-3-methylimidazolium bromide (2), the thiolation of nanotube sidewalls with 2, and the controlled self-assembly of positively charged SWNT-1,2 composites on gold. Optical absorption spectra and resonance Raman (RR) data of obtained aqueous SWNT-1,2 dispersions are consistent with debundled and noncovalently functionalized nanotubes whose electronic properties have not been disturbed. Additionally, the dispersion of pristine nanotube material with surfactants 1 and 2 leads to a high degree of purification from carbonaceous particles. The chiralities of the 14 smallest semiconducting HiPco SWNTs in resonance with Raman excitation at 1064 nm (1.165 eV) were determined in SWNT-2 aqueous dispersion using UV-vis-NIR and RR spectra. X-ray photoelectron spectroscopy (XPS) and surface-enhanced resonance Raman scattering (SERRS) spectroscopy of SWNT-2 submonolayers on gold verified the encapsulation of individualized SWNTs with IL surfactants, the cleavage of S-S disulfide bonds formed in aqueous SWNT-2 suspensions, and the direct chemisorption of the SWNT-2 composite on bare gold via the Au-S bond. Aqueous dispersions of SWNTs with IL-based surfactants add biofunctionality to carbon nanotubes by imparting the positive surface charge necessary for interactions with cell membranes. Our technique, which purifies pristine nanotube material and produces water-soluble, positively charged nanotubes with pendent surface-active thiol groups, may also be translated to other carbon nanotubes and carbon nanostructures. Self-assembled, positively charged submonolayers of SWNTs can be further used for applications in cell biology and sensor technology.  相似文献

Interaction of red blood cells (RBCs) with unmodified and partially (50%) silylated fumed silica A-300 (nanosilica)was studied by microscopic, XRD and thermally stimulated depolarisation current (TSDC) methods. Nanosilica at a low concentration C A-300 < 0.01 wt.% in buffered aqueous suspension is characterised by a weak haemolytic effect on RBCs. However, at C A-300 = 1 wt% all RBCs transform into shadow corpuscles because of 100% haemolysis. Partial (one-half) hydrophobization of nanosilica leads to reduction of the haemolytic effect in comparison with unmodified silica at the same concentrations. A certain portion of the TSDC spectra of the buffered suspensions with RBC/A-300 is independent of the amounts of silica. However, significant portions of the low-and high-temperature TSDC bands have a lower intensity at C A-300 = 1 wt% than that for RBCs alone or RBC/A-300 at C A-300 = 0.01 wt.% because of structural changes in RBCs. Results of microscopic and XRD investigations and calculations using the TSDC-and NMR-cryoporometry suggest that the intracellular structures in RBCs (both organic and aqueous components) depend on nanosilica concentration in the suspension.   相似文献

Even after emergence of most advanced instrumental techniques for the final separation, detection, identification and determination of analytes, sample handling continues to play a basic role in environmental analysis of complex matrices. In fact, sample preparation steps are often the bottleneck for combined time and efficiency in many overall analytical procedures. Thus, it is not surprising that, in the last two decades, a lot of effort has been devoted to the development of faster, safer, and more environment friendly techniques for sample extraction and extract clean up, prior to actual instrumental analysis. This article focuses on the state of the art in sample preparation of environmental solid biological samples dedicated to persistent organic pollutants (POPs) analysis. Extraction techniques such as Soxhlet extraction, sonication-assisted extraction, supercritical fluid extraction (SFE), microwave-assisted extraction (MAE), pressurised liquid extraction (PLE) and matrix solid-phase dispersion (MSPD) are reviewed and their most recent applications to the determination of POPs in biota samples are provided. Additionally, classical as well as promising novel extraction/clean-up techniques such as solid phase microextraction (SPME) are also summarized. Finally, emerging trends in sample preparation able to integrate analytes extraction and their adequate clean-up are presented.  相似文献

To investigate the role and importance of nondirectional electrostatic interactions in mineralization, we explored the use of Langmuir monolayers in which the charge density can be tuned using supramolecular interactions. It is demonstrated that, in mixed Langmuir monolayers of bis-ureido surfactants containing oligo(ethylene oxide) and ammonium head groups associated with matching or nonmatching spacers between the two urea groups, the organization is controlled by molecular recognition. These different organizations of the molecules lead to different nucleation behavior in the mineralization of calcium carbonate. The formation of modified calcite and vaterite crystals was induced selectively by different phases of mixed monolayers, and they were characterized by SEM, TEM, and SAED. To understand the influence of the mixed Langmuir monolayers on the crystallization process, we studied the mixtures by means of (pi-A) isotherms and Brewster angle microscopy observations. Infrared reflection-absorption spectroscopy experiments were also performed on Langmuir-Schaefer films. From these results, we conclude that the local organization of the two systems discussed here gives rise to differences in both charge density and flexibility that together determine not only polymorph selection and the nucleation face but also the morphology of the resulting crystals.  相似文献

A rapid and simple procedure is reported for the determination of six ethylene glycol ethers in cleaning products and detergents using gas chromatography with mass spectrometry. The analytes were extracted from 2.0 g samples in acetonitrile (3 mL) and the extract was submitted to a clean‐up step by QuEChERS method, using a mixture containing 0.3 g magnesium sulfate, 0.15 g primary/secondary amine, and 0.05 g C18. The clean acetonitrile extract (1 μL) was injected into the chromatographic system. No matrix effect was observed, so the quantification of the samples was carried out against external standards. Detection limits were in the range 3.0–27 ng/g for the six ethylene glycol ethers. The recoveries obtained, using the optimized procedure, were in the 89.4–118% range, with relative standard deviations lower than 14%. Twenty‐three different household cleaning products, including glass cleaner, degreaser, floor, softeners, and clothes and dishwashing detergents, were analyzed. Large interindividual variations were observed between samples and compounds.  相似文献

An analytical method for the quantitative determination of the principal phenolic compounds (benzoic acids, hydroxycinnamic acids, 3-phenylpropionic acids, flavanols, procyanidins, dihydrochalcones, quercetin glycosides) in ciders, which successfully employs a RP-HPLC and photodiode-array detection system without prior treatment of the sample, is described. Parameters usually examined in the method validation were evaluated. Good linearity was obtained with correlation coefficients exceeding 0.999 and the detection limits ranged from 0.07 mg/L (p-hydroxybenzoic acid) to 2 mg/L (hydrocaffeic acid). Recoveries ranging between 90 and 104% and the reproducibility of the method was always < 8% (RSD). The method was applied to a set of commercial samples and the results obtained may be helpful to establish a phenolic profile in Asturian cider.  相似文献
