Twenty fungi, which all formed a clear zone around the colony on a poly(ethylene succinate) (PESu)-containing medium, were isolated from various environmental samples. Mesophilic strain NKCM1003, with the highest PESu hydrolytic activity among all the isolates, degraded a PESu film at the rate of 21 ± 2 μg/cm2/h when it was aerobically incubated at 30 °C on a medium containing PESu as the sole carbon source. SEM observations showed that the strain gradually degraded the film starting from the amorphous regions of the surface. Phylogenetic analysis revealed that the strain was closely related to the species Aspergillus clavatus. Zymogram analysis suggested that the secreted enzyme with PESu hydrolytic activity is a P(3HB) depolymerase. The strain also utilized the enzymatic products of PESu, permitting it to grow well. These results indicate that the strain NKCM1003 plays an important role in the PESu-degrading process in the field.  相似文献

Antimony(III) was preconcentrated on activated carbon (AC) as the antimony(III)–pyrogallol complex. Prior to the preconcentration, antimony(V) was reduced to antimony(III) with potassium iodide and ascorbic acid. The antimony adsorbed on the AC was determined by graphite furnace atomic absorption spectrometry as an AC suspension. The method was applied to differential determination of trace amounts of antimony in natural water.  相似文献

The risk of the improved estimator for || or {ei329-1} based on X and S under entropy loss (=d/|| –log(d/||)–1 or d||–log(d||)–1) is evaluated in terms of incomplete beta function of matrix argument and its derivative. A capillary electrophoresis-mass spectrometry method for the simultaneous chiral determination of enantiomers of methamphetamine (MA), amphetamine (AP), dimethylamphetamine (DMA) and p-hydroxymethamphetamine (pOHMA), in urine has been developed. The internal standards used were 2-phenylethylamine and 1-amino4-phenylbutane. The electrolyte was 1 M formic acid (pH 2.2). The chiral selector, which was added to the electrolyte, was a mixture of 3 mM beta-cyclodextrin and 10 mM heptakis(2,6-di-O-methyl)-beta-cyclodextrin. The detection limits were 0.03 microg ml(-1) for the enantiomers of MA and AP and 0.05 microg ml(-1) for the enantiomers of pOHMA using selected ion monitoring. In the analysis of healthy adult urine samples spiked with MA, AP and pOHMA, the precision of within-run assays (n = 4) for the migration time after correction with two internal standards were under 0.04%, and the detection yields utilizing solid phase extraction were 95-105%. This method was applicable to the analysis of urine samples of MA addicts and DMA addicts.  相似文献

Solid-state structures and enzymatic degradability have been investigated for cocrystallized blends between poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [PHBV] and poly(3-hydroxybutyrate-co-3-hydroxypropionate) [PHBP]. From wide-angle X-ray diffraction patterns, small-angle X-ray scattering data, and the comparison of the enzymatic degradability of these blends, the solid-state structures of PHBV/PHBP blend samples, in which the PHBV component has higher isothermal crystal growth rate (G) value than the PHBP one, might be similar to those of the component PHBVs; while those of the PHBP/PHBV blend samples, in which PHBP component has higher G value, were similar to the component PHBPs. Normalized one-dimensional correlation functions gamma(x) of PHBV/PHBP binary blends crystallized at 90 degrees C.  相似文献

Isolation and characterization of poly(butylene succinate)-degrading fungi   总被引:3,自引:0,他引:3  
We isolated 12 poly(butylene succinate) (PBSu)-degrading fungi from various soil environments. Among the isolates, the NKCM1706 strain exhibited the fastest degradation rate for the PBSu film (10.5 μg cm−2 h−1). Phylogenetic analysis revealed that this strain is closely related to Aspergillus fumigatus (internal transcribed spacer (ITS) identity, 100%). Further, this strain exhibited PBSu-hydrolytic activity in the presence of poly(?-caprolactone) (PCL), PBSu, and poly(butylene succinate-co-adipate) (PBSA). On adding this strain into the soil sample, the PBSu degradation rate accelerated approximately sixfold, suggesting that this strain plays a crucial role in PBSu degradation in actual soil environments. In addition to PBSu, the NKCM1706 strain could degrade PBSA, poly(ethylene succinate) (PESu), poly(3-hydroxybutyrate) (P(3HB)), and PCL.  相似文献
