The aqueous, lyotropic liquid-crystalline phase behavior of the alpha-helical polypeptide, poly(N(epsilon)-2-[2-(2-methoxyethoxy)ethoxy]acetyl-lysine) (1), has been studied using optical microscopy and X-ray scattering. Solutions of optically pure 1 were found to form cholesteric liquid crystals at volume fractions that decreased with increasing average chain length. At very high volume fractions, the formation of a hexagonal mesophase was observed. The pitch of the cholesteric phase could be varied by a mixture of enantiomeric samples L-1 and D-1, where the pitch increased as the mixture approached equimolar. The cholesteric phases could be untwisted, using either magnetic field or shear flow, into nematic phases, which relaxed into cholesterics upon removal of field or shear. We have found that the phase diagram of 1 in aqueous solution parallels that of poly(gamma-benzyl glutamate) in organic solvents, thus providing a useful system for liquid-crystal applications requiring water as solvent.  相似文献

Quantification of orotic acid (uracil-6-carboxylic acid) in urine is an important tool to diagnose some inherited diseases, such as urea cycle disorder (OTCD) and hereditary orotic aciduria. New rapid analytical methods are necessary to provide high-throughput orotic acid analyses. A new analytical method has been developed for the rapid analysis of orotic acid in urine by liquid chromatography coupled with ion spray tandem mass spectrometry (LC/MS/MS). After a sample dilution 1:20, the analysis was performed in the selected reaction monitoring mode in which orotic acid was detected through the transition m/z 155 to 111. The retention time was 3.9 min in a 4.5-min analysis. Daily calibration between 0.5-5.0 micromol/L of orotic acid, corresponding to 10-100 micromol/L in urine before the 1:20 dilution, offered consistent linearity and reproducibility. Interassay coefficient of variance (c.v.) was 4.97% at a mean concentration of 10.99 micromol/L. The sensitivity and specificity of tandem mass spectrometry permitted a high volume of analyses of orotic acid. The sample preparation is simple, inexpensive and not time demanding.  相似文献

[reaction: see text] A new mild method for protecting alcohols as t-butyl ethers is reported. The reaction proceeds with Mg(ClO4)2 and Boc2O and shows general applicability. The deprotection of t-butyl ethers has also been revisited. Preliminary results indicate the CeCl3 x 7H2O/NaI system is a very suitable catalyst for their removal.  相似文献

The functionalized cyclodextrin 6-deoxy-6-[1-(2-amino)ethylamino]--cyclodextrin was synthesized, and an NMR, EPR, pH-metric, and calorimetric investigation was carried out in aqueous solution in order to ascertain its behaviour towards protonation and copper(II) complex formation. The thermodynamic parameters of the ternary complex formation with alanine, phenylalanine and tryptophan enantiomeric pairs were also determined (25° C andI=0.1 mol dm–3). No thermodynamic enantioselectivity was observed in these systems, while a chiral, though poor, discrimination was observed in LEC: c.d. spectra also show enantiomeric stereoselectivity. The results of the present investigation, compared with previously reported results, suggest the occurence of acis-complex trans-complex equilibrium in such systems.  相似文献

A new method was developed for the determination of 2-furfural (2-F) and 5-methylfurfural (5-MF), two products of Maillard reaction in vinegar, with head-space solid-phase microextraction (HS-SPME) coupled to gas chromatography-mass spectrometry (GC-MS). A divinylbenzene (DVB)/carboxen (CAR)/polydimethylsiloxane (PDMS) fibre was used and SPME conditions were optimised, studying ionic strength effect, temperature effect and adsorption time. Both analytes were determined by calibration established on 2-furfural-d4 (2-F-d4). The method showed good linearity in the range studied (from 16 to 0.12 mg/l), with a regression coefficient r2 of 0.9999. Inter-batch precision and accuracy were found between 14.9 and 6.0% and between -11.7 and 0.2%, respectively. Detection limit was 15 microg/l. The method is simple and accurate and it has been applied to a series of balsamic and non-balsamic vinegars.  相似文献

The gas-phase acidity of ionized benzyl alcohol and of some of its derivatives with selected reference bases has been studied by Fourier Transform Ion Cyclotron Resonance (FT-ICR) mass spectrometry. The aim was to relate the gas-phase reactivity to the behavior in aqueous solution of the radical cations of benzyl alcohols bearing methoxy substituent(s) on the phenyl ring which are known to undergo deprotonation at both the CH2 and OH groups. The dual reactivity behavior is confirmed in the gas phase, in which the prototypical ion, C6H5CH2OH*+, is deprotonated at both the CH2 and OH groups, whereas the ring hydrogens are not involved. An increasing extent of O-deprotonation is shown as the strength of the base increases. Appropriate methyl substitution, as in the radical cations of C6H5C(Me)2OH and C6H5CH2OMe, allows only O- or C-acidity. The two processes are characterized by comparable thermodynamic features with a Gas-phase Basicity (GB) value of 852 kJ mol(-1) for the cumyloxyl radical and 850 kJ mol(-1) for the alpha-methoxybenzyl radical. The possible origin of the observed mechanistic dichotomy is discussed.  相似文献

Partition coefficients of benzene, toluene, ethylbenzene and xylenes (BTEX), between crosslinked polydimethylsiloxane and water, were determined at room temperature by capillary extraction (a form of in-tube solid-phase microextraction, SPME) coupled to open tubular gas chromatography (in-tube SPME-high-resolution GC). A series of 7-9 repetitive extractions, performed on a 1-ml volume of diluted aqueous BTEX sample by the double-syringe squeeze method, gave exponential regression curves which fit very well with those predicted by partition theory. From the equations of the curves of relative FID response vs. extraction number, experimental Kd were easily calculated and the results compared with literature values. The whole measurement requires about 1 h from the start of the experiment to the final calculation of all BTEX partition coefficients. In-tube SPME resulted in a fast, clean, efficient, and cheaper alternative than the classic 1-cm, externally coated, SPME fiber-holder technique.  相似文献

Electrochemical oxidation of α- and β-diastereomers of a range of steroid hormone receptor marker chromium tricarbonyl complexes, (steroid)Cr(CO)3, have been examined at platinum electrodes in dichloromethane. Data confirm the general nature of previously published conclusions on the oxidation of (arene)Cr(CO)3 complexes (arene = benzene or steroid). That is, with 0.1 M Bu4NPF6 as the electrolyte, and in the absence of nucleophiles, a reversible oneelectron process, (steroid)Cr(CO)3 ? [(steroid)Cr(CO)3]+ + e?, is observed, followed by an irreversible one-electron process at considerably more positive potentials. The reversible half-wave potentials (approximately E°-values) calculated for the [(steroid)Cr(CO)3]+/(steroid)Cr(CO)3 redox couple are shown to be dependent on whether the α- or β-diastereomer is oxidized. Similarly the rate of nucleophilic attack on the 17-electron cation [(steroid)Cr(CO)3]+ by nucleophiles such as ClO, PPh3 and bis(diphenylphosphine)methane confirms a previous observation that the stereochemistry of this class of compound is important with respect to redox, kinetic and hormone receptor properties. The nature of the electrochemical data obtained on the (arene)Cr(CO)3 complexes in the presence of nucleophiles suggests that reactions associated with the nucleophilic attack on the 17-electron cations are complex and that a range of reaction pathways occur simultaneously. Electrochemical studies on the oxidation of (benzene)Cr(CO)2PPh3 and (oestradiol)Cr(CO)2PPh3 confirm some aspects of the proposed mechanisms, although it is clear that a great deal still has to be learned concerning mechanistic aspects of nucleophilic attack on these 17-electron complexes.  相似文献
