A new luciferase reporter gene assay for the detection of androgenic and antiandrogenic effects based on a human prostate specific antigen promoter and PC3/AR human prostate cancer cells.

We developed a new mammalian cell-based luciferase reporter gene assay for androgenic and antiandrogenic activities of chemicals and environmental samples. Environmental samples usually have a complex matrix that may contain the constituents acting as androgen receptor (AR) agonists, AR antagonists or aryl hydrocarbon receptor (AhR) agonists. AhR agonists are known to elicit the antiandrogenic effect through cross-talk between AR and AhR signal transduction pathways. In this study, PC3/AR human prostate carcinoma cells were transiently transfected with a prostate-specific antigen (PSA) promoter-driven luciferase expression plasmid. The cells were treated with a test compound or an environmental sample for 24 h at 37 degrees C and then measured for luciferase activity. The luciferase activity was induced by dihydrotestosterone (DHT) in a concentration-dependent manner in a concentration range from 10 fM to 1 nM. R1881, a synthetic androgen receptor agonist, induced luciferase activity and its inductive effects was additive to that of DHT. The luciferase activity was not induced by cortisol, a glucocorticoid, progesterone, a progestin, and 17beta-estradiol, an estrogen in a concentration range of up to 1 microM. DHT-induced luciferase activity was reduced by bicalutamide and cyproterone acetate, AR antagonists, and also by benzo[a]pyrene, an aryl hydrocarbon receptor agonist, through AhR-mediated pathways. All of these findings indicate that the present assay system correctly responds to AR agonists, AR antagonists and AhR agonist and, therefore, it is a powerful tool for the sensitive and selective screening of chemicals and environmental samples for their androgenic and antiandrogenic activities. We developed the first assay system, in which the expression of luciferase was driven by the promoter of a prostate-specific antigen gene, a typical human androgen-regulated gene.     

Investigation of transitions and relaxation processes in polystyrene by using positron annihilation

The glass transition and relaxation processes in polystyrene resins with the number average molecular weight ranging from 7.0·10² to 9.8·10⁴ were studied with the positron annihilation technique. The pick-off annihilation lifetime of ortho-positronium (₃) and its intensity (I ₃) were measured in the temperature range from 20 to 430 K. The glass transition temperature (T _g) was determined as an onset temperature coefficient of ₃.T _g shows the molecular weight dependence in these samples. BelowT _g, local motions were detected by measurements ofI ₃. The local motions could be observed above 100 K in this experiment.I ₃ show the minimum at around 250 K and it does not show molecular weight dependence.     

Site-selective RNA cleavage by DNA bearing a base pair-mimic nucleoside

We have synthesized the deoxyadenosine derivative tethering a phenyl group (X), which mimics the Watson-Crick A/T base pair. The RNA/DNA hybrid duplexes containing X in the middle of the DNA sequence showed a similar thermal stability regardless of the ribonucleotide species (A, G, C, or U) opposite to X, probably because of the phenyl group stacking inside of the duplex accompanied by the opposite ribonucleotide base flipped in an extrahelical position. The RNA strand hybridized with the DNA strand bearing X was cleaved on the 3'-side of the ribonucleotide opposite to X in the presence of MgCl2, and the RNA sequence to be cleaved was not restricted. The site-specific RNA hydrolysis suggests that the DNA strand bearing X has the advantage of the site-selective base flipping in the target sequence and the development of a "universal deoxyribozyme" to exclusively cleave a target RNA sequence.     

Use of 1-alkyl-3-methylimidazolium hexafluorophosphate room temperature ionic liquids as chelate extraction solvent with 4,4,4-trifluoro-1-(2-thienyl)-1,3-butanedione

Possible use of room temperature ionic liquids (RTILs) as chelate extraction solvent was evaluated by using 1-butyl-3-methylimidazolium hexafluorophosphate ([bmim][PF₆]), 1-hexyl-3-methylimidazolium hexafluorophosphate ([hmim][PF₆]) and 1-octyl-3-methylimidazolium hexafluorophosphate ([omim][PF₆]). These RTILs showed high extraction performance for divalent metal cations with 4,4,4-trifluoro-1-(2-thienyl)-1,3-butanedione (Htta). The extracted metals were back-extracted into 1 mol dm⁻³ nitric acid quantitatively. Furthermore, the extracted species were estimated as neutral hydrated complexes M(tta)₂(H₂O)_n (n= 1 or 2) for M = Ni, Cu and Pb and anionic complexes M(tta)₃⁻ for M = Mn, Co, Zn and Cd.     

Kunzeanones A, B, and C: novel alkylated phloroglucinol metabolites from Kunzea ambigua

Three new metabolites, kunzeanones A (1), B (2), and C (3), along with three known compounds, cryptostrobin (4), (+)-spathulenol (5), and (−)-globulol (6), were isolated from the non-polar fraction of the dried leaves of Kunzea ambigua (Myrtaceae), which shows ichthyotoxicity toward a small fish, medaka. The structures of these new compounds were elucidated as condensates of alkylated phloroglucinol with methylflavanone and germacrane-type sesquiterpene, respectively, on the basis of spectral analyses including 1-D and 2-D NMR spectra. The stereochemistries of kunzeanones A and B were determined by X-ray crystallographic analysis. A sesquiterpene, (+)-spathulenol (5), among the isolates was characterized as the ichthyotoxic principle of the extract.     

Equivalent cross-relaxation rate imaging positively correlates with pathological grade and cell density of adipocytic tumors

Objective

Equivalent cross-relaxation rate imaging (ECRI) is an MRI technique used to evaluate qualitative changes in protein-water interactions. We aimed to prospectively evaluate the utility of ECRI for classification of adipocytic tumors.

Materials and Methods

Institutional Review Board approval was obtained and all patients provided informed consent. Study participants included 40 patients with adipocytic tumors who were diagnosed with lipomas (n = 22), atypical lipomatous tissue/well-differentiated liposarcoma (ALT/WDL; n = 9), myxoid/round cell liposarcoma (MyL; n = 6), and dedifferentiated liposarcoma (DDL; n = 3), and 20 control patients for whom subcutaneous fat in the buttock or thigh was analyzed.

Results

Mean ECR values of lipomas, ALT/WDL, and subcutaneous fat were low, and those of MyL and DDL were high. Mean ECR values of MyL and DDL were significantly higher than those of ALT/WDL. The cut-off value was 5.1%. There was a positive correlation between ECR value, pathological grade, and cell density in adipocytic tumors.

Conclusion

The ECR value positively correlates with pathological grade and cell density of adipocytic tumors. Our findings suggest that ECRI is a useful method for preoperative evaluation of adipocytic tumors.     

Synthesis and characterization of Ti–Ni shape memory alloy thin films by pulsed laser deposition

Shape-memory alloys are crucial in various industrial fields. However, a high-quality film synthesis method has not been established yet. Here we examine optimum conditions for synthesis of thin films by pulsed laser deposition of Ti–Ni alloy target in vacuum. We investigated surface morphologies and chemical compositions of the films which were obtained under various conditions. We found that the suitable Ti/Ni ratio was obtained by adjusting the distance between the target and the substrate in vacuum. In parallel, we analyzed plasma plume by optical emission spectroscopy and time-of-flight mass spectrometry. We discuss the basic behavior of ablated particles in vacuum.     

Coaxial holographic data storage without recording the dc components

A technique of recovering the data pages from Fourier holograms recorded without the dc components is demonstrated theoretically and experimentally by use of a coaxial holographic storage system. A reconstructed image is obtained by adding a phase-modulated dc component of the signal beam on reading. The bit error rate of the reconstructed image is comparable with that for the hologram recorded with the dc component as well. Since high intensities of the dc components are not recorded in this technique, the dynamic range of the recording media can be saved, which potentially contributes to increasing the number of multiplexed holograms.
