We have studied the interaction between metal ions and the metal ion-binding motif in hammerhead ribozymes, as well as the functions of the metal ion at the motif, with heteronuclear NMR spectroscopy. In this study, we employed model RNA systems which mimic the metal ion-binding motif and the altered motif. In Co(NH3)6(III) titrations, we observed large 1H and 31P chemical shift perturbations for the motif and found that outer-sphere complexation of Co(NH3)6(III) is possible for this motif. From the reinvestigation of our previous 15N chemical shift data for Cd(II) binding, in comparison with those of organometallic compounds, we conclude that Cd(II) can form an inner-sphere complex with the nucleobase in the motif. Therefore, the A9/G10.1 site was found to accept both inner-sphere and outer-sphere complexations. The Mg(II) titration for a slightly different motif from the A9/G10.1 site (G10.1-C11.1 to A10.1-U11.1) revealed that its affinity to Mg(II) was drastically reduced, although the ribozyme with this altered motif is known to retain enzymatic activities. This observation suggests that the metal ion at these motifs is not a catalytic center of hammerhead ribozymes.  相似文献

The molecular structure of a three-coordinate palladium(II)-styrene complex, [Pd(η5-C5H5)(PEt3)(styrene)]BF4 has been determined by means of X-ray diffraction. The crystal belongs to the monoclinic system, space group P21/c, with four formula units in a cell of dimensions: a 10.229(3), b 11.262(3), c 18.760(5) Å and β 103.77(2)°. The structure was solved by the heavy atom method, and refined by the least-squares procedure to R = 0.050 for 3635 observed reflections. The palladium atom is surrounded by the cyclopentadienyl group, the triethylphosphine ligand and the olefinic bond of styrene in the cationic complex. In the palladiumstyrene bonding, the olefinic bond is inclined by 77.3° to the coordination plane defined by the Pd and P atoms and the center of the cyclopentadienyl ring (PdC(1) 2.176(6), PdC(2) 2.234(5) and C(1)C(2) 1.369(8) Å).  相似文献

1-Ehtoxycarbonyl-1, 2-dihydroquinoline-2-phosphonates (3) were treated with n-butyllithium followed by alkyl halides to afford the corresponding 4-alkylated phosphonates (7) with complete regioselectivity in 67–97% yields. The phosphonates (7) were converted to 4-alkylquinolines (8) in ca. 50% yields by treatment with sodium iodide in HMPA or by alkaline hydrolysis in aqueous ethanol.  相似文献

A procedure is described by which plutonium and americium can be determined in environmental samples. The sample is leached with nitric acid and hydrogen peroxide, and the two elements are co-precipitated with ferric hydroxide and calcium oxalate. The calcium oxalate is incinerated at 450 degrees and the ash is dissolved in nitric acid. Plutonium is extracted with tri-n-octylamine solution in xylene from 4M nitric acid and stripped with ammonium iodide/hydrochloric acid. Americium is extracted with thenoyltrifluoroacetone solution in xylene at pH 4 together with rare-earth elements and stripped with 1M nitric acid. Americium and the rare-earth elements thus separated are sorbed on Dowex 1 x 4 resin from 1M nitric acid in 93% methanol, the rare-earth elements are eluted with 0.1M hydrochloric acid/0.5M ammonium thiocyanate/80% methanol and the americium is finally eluted with 1.5M hydrochloric acid in 86% methanol. Plutonium and americium in each fraction are electro-deposited and determined by alpha-spectrometry. Overall average recoveries are 81% for plutonium and 59% for americium.  相似文献

Analysis of 2-arachidonoylglycerol (2-AG) and N-arachidonoylethanolamide (anandamide) via alkali or alkaline earth metal-adduct high-energy collision-induced dissociation (CID) in fast-atom bombardment (FAB) ionization-mass spectrometry (MS) is described. The CID-MS/MS of the [2-AG+Li](+) or [2-AG+Na](+) ion undergoes charge-remote fragmentation (CRF), which is useful for the determination of the double-bond positions in the hydrocarbon chain, while the CID-MS/MS of the [2-AG-H+Cat](+) (Cat = Mg(2+), Ca(2+), Ba(2+)) ion provides an abundant fragment ion of the cationized arachidonic acid species, which is derived from cleaving the ester bond via a McLafferty-type rearrangement in addition to structurally informative CRF ions in small amounts. On the other hand, the CID-MS/MS spectra of anandamide cationized with both alkali metal (Li(+) or Na(+)) and alkaline earth metal (Mg(2+), Ca(2+), or Ba(2+)) show CRF patterns: the spectra obtained in lithium or sodium adduct are more clearly visible than those in magnesium, calcium, or barium adduct. The McLafferty rearrangement is not observed with metal-adduct anandamide. The characteristics in each mass spectrum are useful for the detection of these endogenous ligands. m-Nitrobenzyl alcohol (m-NBA) is the most suitable matrix. A lithium-adduct [2-AG+Li](+) or [anandamide+Li](+) ion is observed to be the most abundant in each mass spectrum, since the affinity of lithium for m-NBA is lower than that for other matrices examined.  相似文献

The reaction γ+d → π+? +p + n has been measured in a kinematically complete way at incident photon energies from 570 to 850 MeV in steps of 40 MeV. From detailed comparison of measured data with results of event simulations, it is concluded that three different mechanisms, the quasi-free, double-delta and phase space productions, contribute to the reaction. Each of the cross sections corresponding to these mechanisms is determined separately.  相似文献

Sorption rate curves of CO2, N2, and He gases below 1 atm were measured for polyimide films prepared from benzophenone tetracarboxylic dianhydride (BTDA) with 3,5-diaminotoluene trifluoride (DATF), 2,4-diaminotoluene (DAT), m-phenylenediamine (MPD), and diaminobenzoic acid (DABA). The molecular structures of these four polyimides differ only in the substituent groups of the diamine structure. These polyimides exhibit dualmode type sorption isotherms for carbon dioxide that are concave to the pressure axis, typical of glassy polymer/gas system. The apparent diffusion coefficients below 1 atm pressure of carbon dioxide for this series of compounds decrease in the order: BTDA-DATF > BTDA-DAT > BTDA-MPD > BTDA-DABA. A linear relation between the logarithm of the apparent diffusion coefficient and the reciprocal of free volume, calculated by the method of Bondi using density data, is found for these polyimides. However, this tendency is not observed for the other two gases. The activation energies of the apparent diffusion coefficients at 20 cmHg pressure of carbon dioxide increase with increasing cohesive energy density of the polyimides. The energy per mole of free volume elements in a liquidlike structure in each cohesive energy density may be equated to the activation energy and used to calculate the free volume. The values from the activation energy are almost the same as those from Bondi's method.  相似文献

We investigate the molecular orientation dependence of the (o)rtho-(p)ara H2 conversion on Fe(OH)3 cluster. Taking Fermi's contact interaction as perturbation, we find that the o-p H2 conversion yield for the molecular orientation angle θ= 10 with respect to the surface normal is larger than that for θ= 70.  相似文献
