Convenient synthesis of photoaffinity probes and evaluation of their labeling abilities

Convenient synthesis of a variety of photoaffinity probes was accomplished by utilizing our Ns strategy and novel resin. The synthetic probes were evaluated via the labeling ability with the preseniline 1 C-terminal fragments, which was identified as a therapeutic target for Alzheimer's disease.     

Synthesis,resolution, and absolute stereochemistry of (-)-blestriarene C

A naturally occurring 1,1'-biphenanthrene, blestriarene C (1), was prepared in 13 steps and 30% overall yield. The key steps are the ester-mediated nucleophilic aromatic substitution on 2,6-di-tert-butyl-4-methoxyphenyl 5-isopropoxy-2-methoxybenzoate (4) by 2-methoxy-4-methoxymethoxy-6-methylphenylmagnesium bromide (5) and a novel intramolecular cyclization of the resulting 4-isopropoxy-2'-methoxy-4'-methoxymethoxy-6'-methylbiphenyl-2-carboxylic ester 14 to 7-isopropoxy-4-methoxy-2-(methoxymethoxy)phenanthren-9-ol (15). The racemic blestriarene C was optically resolved by chiral HPLC on a preparative scale to give several 10-mg yields of both the enantiomers in up to 95% ee. The absolute stereochemistry was determined to be S(a)-(-) by the axial chirality recognition method, which was based on the stereospecific formation of a 12-membered cyclic diester containing two biaryl-o,o'-diyl unites joined by ester -CO(2)- linkages. The validity of the method was confirmed by an X-ray crystallographic analysis and ab initio conformational analyses of such 12-membered cyclic diesters. It was found that blestriarene C and its 7,7'-diisopropyl ether 2 underwent rapid photoracemization even under ambient light exposure.     

Unique inclusion behaviour of 5,11,17,23-tetra-tert-butyl-25,26,27,28-tetraaminothiacalix[4]-arene towards small organic molecules

Tetraaminothiacalixarene 3, bearing four amino groups instead of the hydroxy groups of p-tert-butylthiacalix[4]arene 2, exhibits inclusion properties different from those of compound 2 towards small organic molecules upon crystallisation from neat solvents or guest solutions. X-ray crystallographic analyses reveal that nitromethane and acetonitrile are included into the cone-shaped cavity of compound 3, as is often seen in inclusion crystals of compound 2, whereas dichloromethane occupies a space between two distal benzene rings of compound 3, adopting a 1,3-alternate conformation. Acetic acid, which forms a dimer, fills a pore surrounded by four host molecules with a pinched cone conformation. Furthermore, guest-free crystals of compound 3 with a 1,3-alternate conformation absorb acetonitrile to give inclusion crystals with the same crystal structure as that obtained by the crystallisation. Thus, compound 3 flexibly changes its conformation according to the structures of guest compounds.     

Widely repetition-tunable 200 fs pulse source using a Mach-Zehnder-modulator-based flat comb generator and dispersion-flattened dispersion-decreasing fiber

By combining a Mach-Zehnder-modulator-based flat comb generator (MZ-FCG) with a dispersion-flattened dispersion-decreasing fiber, femtosecond pulses have been generated from a cw light. Near-Fourier-transform-limit picosecond pulses generated from the MZ-FCG were compressed into femtosecond order by pulse compression. Our system enables flexible tuning of the repetition rate and pulse width, because those depend on the driving signal of the MZ-FCG. Pulse trains of 200 fs width were continuously and stably generated without mode hopping, with a repetition rate range from 5 to 17 GHz. Our system consists of a modulator and compression fiber; thus, the configuration is simpler and more stable.
