Whereas the cleavage of alkenes by ozone typically generates peroxide intermediates that must be decomposed in an accompanying step, ozonolysis in the presence of pyridine directly generates ketones or aldehydes through a process that neither consumes pyridine nor generates any detectable peroxides. The reaction is hypothesized to involve nucleophile-promoted fragmentation of carbonyl oxides via formation of zwitterionic peroxyacetals.  相似文献

A two-phase polycondensation reaction was used to prepare high molecular weight linear poly-(amino-s-triazine)s from aminodichloro-s-triazines and diamines. These polymers contain a high concentration of melamine-type units in the polymer backbone. Molded specimens exhibited good mechanical properties and resistance to organic solvents. Dimethyl substitution at the exocyclic amino group led to a significant reduction in indentation resistance, strength, and modulus.  相似文献

Gaseous bis(hexamethydisilylamido)mercury(II), Hg{N(SiMe3)2)2}2, has been studied by electron diffraction at a nozzle temperature of ca 390 K.

The diffraction data are consistent with a model consisting only of monomers. By assuming the NHgN chain to be linear and the HgHSi2 fragments to be planar, an equilibrium conformer with a staggered Si2NHgNSi2 skeleton of Dad-symmetry may be brought into a nice agreement with the observed diffraction data. The relatively large value of the vibrational amplitude of the inter-ligand SiSi distance, 0.26(12) A, indicates that the ligands undergo large amplitude vibrations about the NHgN axis. Steric considerations as well as the magnitude of the rotational barrier as estimated from the diffraction data (ca. 2 kcal mol−1) show that this motion is hindered. A model with an eclipsed, co-planar Si2NHgNSi2 backbone of Dadsymmetry could not satisfactorily be brought into agreement with the observed diffraction data.

The values of some relevant key-parameters are: ra(Hg---N) = 2.01(2) A, ra(Si---N) = 1.732(9) A, ra(Si---C) = 1.883(6) A;HgNSi = 116.0(1.0)°, SiNSi = 128.0(2.0)°, NSiC= 111.8(1.2)° and SiCH = 111.0(2.0)°. The trimethylsilyl groups are twisted 25(3)° away from their references positions typified by one Si---C bond of each such group eclipsing the adjacent Hg---N bond, in such a way that the overall symmetry of the model is lowered from Dad to S4.  相似文献

The thermal decomposition of SEX in a nitrogen atmosphere was studied by coupled thermogravimetry-Fourier transform infrared spectroscopy (TG-FTIR), and by pyrolysis-gas chromatography-mass spectrometry (py-GC-MS). The TG curve exhibited two discrete mass losses of 45.8% and 17.8% respectively, at 200 and 257–364°C. The evolved gases identified as a result of the first mass loss were carbonyl sulfide (COS), ethanol (C2H5OH), ethanethiol (C2H5SH), carbon disulfide (CS2), diethyl sulfide ((C2H5)2S), diethyl carbonate ((C2H5O)2CO), diethyl disulfide ((C2H5)2S2), and carbonothioic acid, O, S, diethyl ester ((C2H5S)(C2H5O)CO). The gases identified as a result of the second mass loss were carbonyl sulfide, ethanethiol, and carbon disulfide. Hydrogen sulfide was detected in both mass losses by py-GC-MS, but not detected by FTIR. The solid residue was sodium hydrogen sulfide (NaSH).SEX was adsorbed onto activated carbon, and heated in nitrogen. Two discrete mass losses were still observed, but in the temperature ranges 100–186°C (7.8%) and 186–279°C (11.8%). Carbonyl sulfide and carbon disulfide were now the dominant gases evolved in each of the mass losses, and the other gaseous products were relatively minor. It was demonstrated that water adsorbed on the carbon hydrolysed the xanthate to cause the first mass loss, and any unhydrolysed material decomposed to give the second mass loss.Mr. N. G. The excitation and emission properties of several psoralen derivatives are compared using conventional single-photon excitation and simultaneous two-photon excitation (TPE). Two-photon excitation is effected using the output of a mode-locked titanium: sapphire laser, the near infrared output of which is used to promote non-resonant TPE directly. Specifically, the excitation spectra and excited-state properties of 8-methoxypsoralen and 4′-aminomethyl-4,5,8-trimethylpsoralen are shown to be equivalent using both modes of excitation. Further, in vitro feasibility of two-photon photodynamic therapy (PDT) is demonstrated using Salmonella typhimurium. Two-photon excitation may be beneficial in the practice of PDT because it would allow replacement of visible or UV excitation light with highly penetrating, nondamag-ing near infrared light and could provide a means for improving localization of therapy. Comparison of possible laser excitation sources for PDT reveals the titanium: sapphire laser to be exceptionally well suited for nonlinear excitation of PDT agents in biological systems due to its extremely short pulse width and high repetition rate that together provide efficient PDT activation and greatly reduced potential for biological damage  相似文献

Increased Photosensitivity in HL60 Cells Expressing Wild-Type p53   总被引:4,自引:0,他引:4  
Loss of p53 function has been correlated with decreased sensitivity to chemotherapy and radiation therapy in a variety of human tumors. Comparable analysis of p53 status with sensitivity to oxidative stress induced by pho-todynamic therapy has not been reported. In the current study we examined photosensitivity in human promye-locytic leukemia HL60 cells exhibiting either wild-type p53, mutated p53 or deleted p53 expression. Experiments were performed using a purpurin, tin ethyl etiopurpurin (SnET2)-, or a porphyrin, Photofrin (PH)-based photo-sensitizer. Total SnET2 accumulation was comparable in all three cell lines. Uptake of PH was highest in cells expressing wild-type p53 but incubation conditions could be adjusted to achieve equivalent cellular PH levels during experiments that analyzed photosensitivity. Survival measurements demonstrated that HL60 cells expressing wild-type p53 were more sensitive to PH- and SnET2-mediated photosensitization, as well as to UVC irradiation, when compared to HL60 cells exhibiting deleted or mutated p53 phenotypes. A rapid apoptotic response was observed following purpurin- and porphyrin-induced photosensitization in all cell lines. Results of this study indicate that photosensitivity is increased in HL60 cells expressing wild-type p53 and that photosensitizer-medi-ated oxidative stress can induce apoptosis through a p53-independent mechanism in HL60 cells .  相似文献

Reactivity assays previously suggested that two quinol-containing MRI contrast agent sensors for H2O2, [Mn(H2qp1)(MeCN)]2+ and [Mn(H4qp2)Br2], could also catalytically degrade superoxide. Subsequently, [Zn(H2qp1)(OTf)]+ was found to use the redox activity of the H2qp1 ligand to catalyze the conversion of O2˙ to O2 and H2O2, raising the possibility that the organic ligand, rather than the metal, could serve as the redox partner for O2˙ in the manganese chemistry. Here, we use stopped-flow kinetics and cryospray-ionization mass spectrometry (CSI-MS) analysis of the direct reactions between the manganese-containing contrast agents and O2˙ to confirm the activity and elucidate the catalytic mechanism. The obtained data are consistent with the operation of multiple parallel catalytic cycles, with both the quinol groups and manganese cycling through different oxidation states during the reactions with superoxide. The choice of ligand impacts the overall charges of the intermediates and allows us to visualize complementary sets of intermediates within the catalytic cycles using CSI-MS. With the diquinolic H4qp2, we detect Mn(iii)-superoxo intermediates with both reduced and oxidized forms of the ligand, a Mn(iii)-hydroperoxo compound, and what is formally a Mn(iv)-oxo species with the monoquinolate/mono-para-quinone form of H4qp2. With the monoquinolic H2qp1, we observe a Mn(ii)-superoxo ↔ Mn(iii)-peroxo intermediate with the oxidized para-quinone form of the ligand. The observation of these species suggests inner-sphere mechanisms for O2˙ oxidation and reduction that include both the ligand and manganese as redox partners. The higher positive charges of the complexes with the reduced and oxidized forms of H2qp1 compared to those with related forms of H4qp2 result in higher catalytic activity (kcat ∼ 108 M−1 s−1 at pH 7.4) that rivals those of the most active superoxide dismutase (SOD) mimics. The manganese complex with H2qp1 is markedly more stable in water than other highly active non-porphyrin-based and even some Mn(ii) porphyrin-based SOD mimics.

Manganese complexes with polydentate quinol-containing ligands are found to catalyze the degradation of superoxide through inner-sphere mechanisms. The redox activity of the ligand stabilizes higher-valent manganese species.  相似文献

A generic high-throughput liquid chromatography (HTLC) tandem mass spectrometry (MS/MS) assay for the determination of compound I in human urine and dialysate (hemodialysis) was developed and validated. By using the HTLC on-line extraction technique, sample pretreatment was not necessary. The sample was directly injected onto a narrow bore large particle size extraction column (50 x 1.0 mm, 60 microm) where the sample matrix was rapidly washed away using a high flow rate (5 mL/min) aqueous mobile phase while analytes were retained. The analytes were subsequently eluted from the extraction column onto an analytical column using an organic-enriched mobile phase prior to mass spectrometric detection. The analytes were then eluted from the analytical column to the mass spectrometer for the determination. The linear dynamic range was 2.0-6000 ng/mL for the urine assay and 0.1-300 ng/mL for the dialysate assay. Intraday accuracy and precision were evaluated by analyzing five replicates of calibration standards at all concentrations used to construct the standard curve. For the urine assay, the precision (RSD%, n=5) ranged from 1.9 to 8.0% and the accuracy ranged from 87.8 to 105.2% of nominal value. For the dialysate assay, the precision (RSD%, n=5) ranged from 1.1 to 10.0% and the accuracy from 94.5 to 105.2% of nominal value. In-source fragmentation of the acyl glucuronide metabolite (compound III) did not interfere with the determination of parent compound I. The developed HTLC/MS/MS methodology was specific for compound I in the presence of compound III. Column life-time is increased and sample analysis time is decreased over traditional reversed-phase methods when direct injection assays for urine and dialysate are coupled with the technology of HTLC.  相似文献

An antibody that binds rare earth complexes selectively could be used as a docking station for a set of probe molecules, of particular interest for medical imaging and therapy. The rare earths are rich in probe properties, such as the paramagnetism of Gd, the luminescence of Tb and Eu, and the nuclear properties of Lu and Y. We find that antibody 2D12.5, initially developed to bind analogues of Y-DOTA (1,4,7,10-tetraazacyclododecane-N,N',N' ',N' '-tetraacetic acid) for radiotherapy, binds not only Y-DOTA analogues but also analogous DOTA complexes of all of the lanthanides. Surprisingly, chelates of some metals such as Gd3+ bind more tightly than the original Y3+ complex. When the shape of the complex is perturbed by either increasing or decreasing the radius of the lanthanide ion, the thermodynamic stability of the protein-ligand complex changes in a regular fashion. The behavior of DeltaDeltaG as a function of ionic radius fits a parabola, as might be expected for a system that behaves in a thermodynamically elastic way. The broad specificity and high affinity of this antibody for all rare earth-DOTA complexes make it particularly interesting for applications that take advantage of the unique characteristics of lanthanides. For example, UV excitation of the Tb-DOTA-2D12.5 complex leads to energy transfer from aromatic side chains of the antibody to bound Tb-DOTA, enhancing green terbium luminescence >104 relative to unbound Tb-DOTA.  相似文献

Background  

Nudix hydrolases form a protein family whose function is to hydrolyse intracellular nucleotides and so regulate their levels and eliminate potentially toxic derivatives. The genome of the radioresistant bacterium Deinococcus radiodurans encodes 25 nudix hydrolases, an unexpectedly large number. These may contribute to radioresistance by removing mutagenic oxidised and otherwise damaged nucleotides. Characterisation of these hydrolases is necessary to understand the reason for their presence. Here, we report the cloning and characterisation of the DR0975 gene product, a nudix hydrolase that appears to be unique to this organism.  相似文献
