Fluorescent-based single-strand conformation polymorphism/heteroduplex capillary electrophoretic mutation analysis of the P53 gene.

Fluorescent-based single-strand conformation polymorphism (F-SSCP) analysis with capillary electrophoresis (CE) is the most common method for the detection of mutation because of its high sensitivity and resolution. In this study, we prepared an inexpensive linear polyacrylamide (LPA), and successfully applied it to CE-SSCP analysis and tandem CE-SSCP/heteroduplex analysis (HA) of the P53 gene on an ABI capillary genetic analyzer. A comparison of the sieving capabilities of a homemade LPA and commercial polydimethylacrylamide (PDMA) demonstrates that the homemade LPA has a higher resolution, a shorter analysis time, and is more suitable for tandem SSCP/HA than commercial PDMA. To show the usefulness, mutations of P53 gene exon 7 - 8 in 37 tumor samples were investigated by using homemade LPA. The results indicate that 10 mutations were found in 9 of 37 cases; the majority of P53 mutations were missense mutations, and 70% were located in exon 7, which plays an important role in neoplastic progression in human tumorigenesis.     

Transition‐metal‐catalyzed reactions of 5‐methylene‐2‐oxazolidinone and 5‐methylene‐1,3‐thiazolidine‐2‐thione with isocyanates

5‐Methylene‐2‐oxazolidinone (1) and 5‐methylene‐1,3‐thiazolidine‐2‐thione (4) react with various isocyanates to give the corresponding urethanes 3 and 5 in high yields in the presence of palladium(0) or palladium(II) catalyst under mild reaction conditions. Study on reproductive endocrinology of human placenta (III)--Hormonal regulation of progesterone production by trophoblast tissue of first trimester.

The effect of hormones on progesterone secretion by 6-8 week human trophoblast tissue cultured in serum-free medium has been investigated. GnRH at low concentration (10(-10)-10(-8) mol/L) stimulated progesterone secretion, while high dose (10(-6)-10(-5) mol/L) produced inhibitory effect. The progesterone secretion could be significantly decreased by addition of anti-hCG antiserum or monoclonal anti-hCG IgG in a dose- and time-dependent manner. Various concentrations of TRH, PGE2, PGF2 alpha, testosterone and estradiol were found to be ineffective. These data indicate clearly that progesterone production by human trophoblast tissue at early gestation stage is under the modulation of GnRH and hCG.     

Linear Ramsey numbers of sparse graphs

The Ramsey number R(G₁,G₂) of two graphs G₁ and G₂ is the least integer p so that either a graph G of order p contains a copy of G₁ or its complement G^c contains a copy of G₂. In 1973, Burr and Erd?s offered a total of $25 for settling the conjecture that there is a constant c = c(d) so that R(G,G)≤ c|V(G)| for all d‐degenerate graphs G, i.e., the Ramsey numbers grow linearly for d‐degenerate graphs. We show in this paper that the Ramsey numbers grow linearly for degenerate graphs versus some sparser graphs, arrangeable graphs, and crowns for example. This implies that the Ramsey numbers grow linearly for degenerate graphs versus graphs with bounded maximum degree, planar graphs, or graphs without containing any topological minor of a fixed clique, etc. © 2005 Wiley Periodicals, Inc. Synthesis and application as polymer electrolyte of hyperbranched polyether made by cationic ring‐opening polymerization of 3‐{2‐[2‐(2‐hydroxyethoxy)ethoxy]ethoxymethyl}‐3′‐methyl‐oxetane

A novel cyclic ether monomer 3‐{2‐[2‐(2‐hydroxyethoxy)ethoxy]ethoxy‐methyl}‐3′‐methyloxetane (HEMO) was prepared from the reaction of 3‐hydroxymethyl‐3′‐methyloxetane tosylate with triethylene glycol. The corresponding hyperbranched polyether (PHEMO) was synthesized using BF₃·Et₂O as initiator through cationic ring‐opening polymerization. The evidence from ¹H and ¹³C NMR analyses revealed that the hyperbranched structure is constructed by the competition between two chain propagation mechanisms, i.e. active chain end and activated monomer mechanism. The terminal structure of PHEMO with a cyclic fragment was definitely detected by MALDI‐TOF measurement. A DSC test implied that the resulting polyether has excellent segment motion performance potentially beneficial for the ion transport of polymer electrolytes. Moreover, a TGA assay showed that this hyperbranched polymer possesses high thermostability as compared to its liquid counterpart. The ion conductivity was measured to reach 5.6 × 10^?5 S/cm at room temperature and 6.3 × 10^?4 S/cm at 80 °C after doped with LiTFSI at a ratio of Li:O = 0.05, presenting the promise to meet the practical requirement of lithium ion batteries for polymer electrolytes. © 2006 Wiley Periodicals, Inc. Chemical constituents of Taraxacum formosanum

Three new compounds, taraxacine-A (1), taraxacine-B (2) and taraxafolin (3) together with twenty-five known compounds, which include two beta-carboline alkaloids, two indole alkaloids, two chlorophylls, two flavonoids, one coumarin, two triterpenoids, one monoterpenoid, one ionone, four steroids and eight benzenoids, were isolated and characterized from the fresh aerial parts of Taraxacum formosanum. Structures of new compounds were determined by spectral analysis.     

Application of co-eluting structural analog internal standards for expanded linear dynamic range in liquid chromatography/electrospray mass spectrometry

Non-linear standard calibration curves occur frequently in liquid chromatography/electrospray ionization mass spectrometry (LC/ESI-MS), necessitating the use of non-linear regression curve fitting or a reduction in assay dynamic range. Non-linearity can be minimized by the use of stable isotope internal standards, if the cause of curve bending is ionization saturation. Unfortunately, stable isotope internal standards can be difficult or expensive to synthesize. Structural analog internal standards, if sufficiently close to analytes both in chemical structures and HPLC retention times, may mimic the effect of stable isotope internal standards. Tentative experimental evidence supporting this concept is presented in this paper.
