An integrative passive sampler (Chemcatcher®) consisting of a 47 mm C18 Empore™ disk as the receiving phase overlaid with a thin cellulose acetate diffusion membrane was developed and calibrated for the measurement of time-weighted average water concentrations of organotin compounds [monobutyltin (MBT), dibutyltin (DBT), tributlytin (TBT) and triphenyltin (TPhT)] in water. The effect of water temperature and turbulence on the uptake rate of these analytes was evaluated in the laboratory using a flow-through tank. Uptake was linear over a 14-day period being in the range: MBT (3-23 mL day−1), DBT (40-200 mL day−1), TBT (30-200 mL day−1) and TPhT (30-190 mL day−1) for all the different conditions tested. These sampling rates were high enough to permit the use of the Chemcatcher® to monitor levels of organotin compounds typically found in polluted aquatic environments. Using gas chromatography (GC) with either ICP-MS or flame photometric detection, limits of detection for the device (14-day deployment) for the different organotin compounds in water were in the range of 0.2-7.5 ng L−1, and once accumulated in the receiving phase the compounds were stable over prolonged periods. Due to anisotropic exchange kinetics, performance reference compounds could not be used with this passive sampling system to compensate for changes in sampling rate due to variations in water temperature, turbulence and biofouling of the surface of the diffusion membrane during field deployments. The performance of the Chemcatcher® was evaluated alongside spot water sampling in Alicante Habour, Spain which is known to contain elevated levels of organotin compounds. The samplers provided time-weighted average concentrations of the bioavailable fractions of the tin compounds where environmental concentrations fluctuated markedly in time.  相似文献

High-throughput screening (HTS) is an effective method for lead and probe discovery that is widely used in industry and academia to identify novel chemical matter and to initiate the drug discovery process. However, HTS can be time consuming and costly and the use of subsets as an efficient alternative to screening entire compound collections has been investigated. Subsets may be selected on the basis of chemical diversity, molecular properties, biological activity diversity or biological target focus. Previously, we described a novel form of subset screening: plate-based diversity subset (PBDS) screening, in which the screening subset is constructed by plate selection (rather than individual compound cherry-picking), using algorithms that select for compound quality and chemical diversity on a plate basis. In this paper, we describe a second-generation approach to the construction of an updated subset: PBDS2, using both plate and individual compound selection, that has an improved coverage of the chemical space of the screening file, whilst only selecting the same number of plates for screening. We describe the validation of PBDS2 and its successful use in hit and lead discovery. PBDS2 screening became the default mode of singleton (one compound per well) HTS for lead discovery in Pfizer.  相似文献

When the nucleus independent chemical shifts, NICS(1)zz, for a set of aromatic and antiaromatic hydrocarbons are summed, they show an excellent linear relationship with the magnetic susceptibility exaltation, Lambda, for neutral, cationic, and monoanionic species. Aromatic and antiaromatic dianions show a similar relationship but with a different slope. However, when both Lambda and the summation of NICS(1)zz are divided by the area of the ring squared, the vast majority of the aromatic and antiaromatic species fall on the same line, indicating that both NICS and Lambda are affected by the size of the ring. The species that deviate slightly from the line are a few of the anionic compounds, which may be a result of the difficulties of calculating magnetic properties of anions. This is the first report of the relationship of NICS to ring area. In addition, the excellent correlation between Lambda and the summation of NICS(1)zz demonstrates that summation of NICS(1)zz values for individual ring systems of polycyclic ring systems to give a measure of the aromaticity of the entire system is justified. By extension, the excellent correlation also serves to demonstrate that the NICS(1)zz values for individual ring systems are reliable measures of local aromaticity/antiaromaticity. Finally, the excellent correlation between experimental shifts and the 13C NMR shifts calculated with density functional theory, B3LYP/6-311+g(d,p), serves as an indirect validation of the accuracy of the NICS(1)zz calculated by the same method and basis set.  相似文献

A library of 200 2-alkyl-3-alkyloxy-2H-indazole-6-carboxamides was synthesized using parallel solution-phase methods. The indazole cyclization reaction was optimized for library production with the best yields resulting from controlled alcohol/water solvent ratios. The key step, a heterocyclization reaction, proceeds by N,N-bond formation and delivers the 2H-indazole scaffold. Automated preparative HPLC was utilized to provide pure compounds on a 10+ mg scale.  相似文献

We present calculations of the laser-induced force between metallic nanospheres, similar and dissimilar in character, and that between a metallic nanosphere and a planar surface. When the separation between these objects is in the 0.5-2 nm range, we find very strong resonances in the laser-induced force associated with excitation of plasmon resonances. Measurement of such forces will provide direct access to the plasmon enhancements of laser fields so critical to optical spectroscopy in the nanoenvironment.  相似文献

Ni-44 at.% Al and Ni-50 at.% Al single crystals were tested in compression in the hard d001 ¢orientation. The dislocation processes and deformation behaviour were studied as a function of temperature, strain and strain rate. A slip transition in NiAl occurs from a?111? slip to non-a?111? slip at intermediate temperatures. In Ni-50 at.% Al single crystals, only a?010? dislocations are observed above the slip transition temperature. In contrast, a a?101?{101} glide has been observed to control deformation beyond the slip transition temperature in Ni-44 at.% Al. a?101? dislocations are observed primarily along both ?111? directions in the glide plane. High-resolution transmission electron microscopy observations show that the core of the a?101? dislocations along these directions is decomposed into two a?010? dislocations, separated by a distance of approximately 2 nm. The temperature window of stability for these a?101? dislocations depends upon the strain rate. At a strain rate of 1.4 210?4 s?1, a?101? dislocations are observed between 800 and 1000 K. Complete decomposition of a?101? dislocations into a?010? dislocations occurs beyond 1000 K, leading to a?010? climb as the deformation mode at higher temperatures. At lower strain rates, decomposition of a?101? dislocations has been observed to occur along the edge orientation at temperatures below 1000 K. Embedded-atom method calculations and experimental results indicate that a?101? dislocations have a large Peierls stress at low temperatures. Based on the present microstructural observations and a survey of the literature with respect to vacancy content and diffusion in NiAl, a model is proposed for a?101?{101} glide in Ni-44 at.% Al, and for the observed yield strength versus temperature behaviour of Ni-Al alloys at intermediate and high temperatures.  相似文献
