This paper describes the development of two independent analytical methods for the extraction and quantification of methylmercury
from marine biota. The procedures involve microwave extraction, followed by derivatization and either headspace solid-phase
microextraction (SPME) with a polydimethylsiloxane (PDMS)-coated silica fiber or back-extraction into iso-octane. The identification
and quantification of the extracted compounds is carried out by capillary gas chromatography/mass spectrometric (GC/MS) and
inductively coupled plasma mass spectrometric (GC/ICP-MS) detection. Both methods were validated for the determination of
methylmercury (MeHg) concentrations in a variety of biological standard reference materials (SRMs) including fresh-frozen
tissue homogenates of SRM 1946 Lake Superior fish tissue and SRM 1974a organics in mussel tissue (Mytilus edulis) and then applied to the certification effort of SRM 1947 Lake Michigan fish tissue and SRM 1974b organics in mussel tissue
(Mytilus edulis). While past certifications of methylmercury in tissue SRMs have been based on two independent methods from the National
Institute of Standards and Technology (NIST) and participating laboratories, the methods described within provide improved

Well-characterized certified reference materials are needed by laboratories in the food testing, dietary supplement, and nutrition
communities to facilitate compliance with labeling laws and improve the accuracy of information provided on product labels,
so that consumers can make good choices. As a result of the enactment of the Nutrition Labeling and Education Act of 1990
and the Infant Formula Act of 1980, the National Institute of Standards and Technology (NIST) worked to develop a series of
food-matrix standard reference materials (SRMs) characterized for nutrient concentrations. These include SRM 1544 Fatty Acids
and Cholesterol in a Frozen Diet Composite, SRM 1546 Meat Homogenate, SRM 1548a Typical Diet, SRM 1566b Oyster Tissue, SRM
1846 Infant Formula, SRM 1946 Lake Superior Fish Tissue, SRM 1947 Lake Michigan Fish Tissue, SRM 2383 Baby Food Composite,
SRM 2384 Baking Chocolate, SRM 2385 Slurried Spinach, and SRM 2387 Peanut Butter. With the enactment of the Dietary Supplement
Health and Education Act of 1994, NIST has been working to develop suites of dietary supplement SRMs characterized for active
and marker compounds and for toxic elements and pesticides, where appropriate. An updated SRM 1588b Organics in Cod Liver
Oil, a suite of ephedra-containing materials (SRMs 3240–3245), a carrot extract in oil (SRM 3276), and a suite of ginkgo-containing
materials (SRMs 3246–3248) are available. Several other materials are currently in preparation. Dietary supplements are sometimes
provided in forms that are food-like; for these, values may also be assigned for nutrients, for example SRM 3244 Ephedra-Containing
Protein Powder. Both the food-matrix and dietary supplement reference materials are intended primarily for validation of analytical
methods. They may also be used as “primary control materials” in assignment of values to in-house (secondary) control materials
to confirm accuracy and to establish measurement traceability to NIST. 相似文献

Standard reference materials (SRMs) are valuable tools in developing and validating analytical methods to improve quality
assurance standards. The National Institute of Standards and Technology (NIST) has a long history of providing environmental
SRMs with certified concentrations of organic and inorganic contaminants. Here we report on new certified and reference concentrations
for 27 polybrominated diphenyl ether (PBDE) congeners in seven different SRMs: cod-liver oil, whale blubber, fish tissue (two
materials), mussel tissue and sediment (two materials). PBDEs were measured in these SRMs, with the lowest concentrations
measured in mussel tissue (SRM 1974b) and the highest in sediment collected from the New York/New Jersey Waterway (SRM 1944).
Comparing the relative PBDE congener concentrations within the samples, we found the biota SRMs contained primarily tetrabrominated
and pentabrominated diphenyl ethers, whereas the sediment SRMs contained primarily decabromodiphenyl ether (BDE 209). The
cod-liver oil (SRM 1588b) and whale blubber (SRM 1945) materials were also found to contain measurable concentrations of two
methoxylated PBDEs (MeO-BDEs). Certified and reference concentrations are reported for 12 PBDE congeners measured in the biota
SRMs and reference values are available for two MeO-BDEs. Results from a sediment interlaboratory comparison PBDE exercise

A large‐scale synthetic route to a variety of phosphaformamidines and phosphaformamidinates, a type of derivative that was not accessible by the methods previously known for preparing phosphaamidines and phosphaamidinates, is reported. Thermally stable ethyl N‐arylformimidates 1 (ArN?CH(OEt), Ar=2,4,6‐(Me)3Ph or 2,6‐(iPr)2Ph) readily reacted with lithium dialkyl‐ and diarylphosphanides to afford the corresponding N‐aryl phosphaformamidines in 80 and 60 % yield, respectively, whereas with lithium (aryl)(silyl)phosphanide, the N‐aryl‐N‐silylphosphaformamidine (60 % yield) was obtained. Addition of primary lithium arylphosphanides to 1 followed by addition of a stoichiometric amount of nBuLi gave rise to the respective phosphaformamidinates (70–88 % yield). Methanolysis of the products afforded the N‐aryl‐N‐hydrogenophosphaformamidines (90–95 % yield). The solid‐state structure of one of the phosphaformamidinates is also presented. 相似文献

An experimental and theoretical study of the electronic structure of copper phthalocyanine (CuPc) molecule is presented. We performed x-ray photoemission spectroscopy (XPS) and photoabsorption [x-ray absorption near-edge structure (XANES)] gas phase experiments and we compared the results with self-consistent field, density functional theory (DFT), and static-exchange theoretical calculations. In addition, ultraviolet photoelectron spectra (UPS) allowed disentangling several outer molecular orbitals. A detailed study of the two highest occupied orbitals (having a(1u) and b(1g) symmetries) is presented: the high energy resolution available for UPS measurements allowed resolving an extra feature assigned to vibrational stretching in the pyrrole rings. This observation, together with the computed DFT electron density distributions of the outer valence orbitals, suggests that the a(1u) orbital (the highest occupied molecular orbital) is mainly localized on the carbon atoms of pyrrole rings and it is doubly occupied, while the b(1g) orbital, singly occupied, is mainly localized on the Cu atom. Ab initio calculations of XPS and XANES spectra at carbon K edge of CuPc are also presented. The comparison between experiment and theory revealed that, in spite of being formally not equivalent, carbon atoms of the benzene rings experience a similar electronic environment. Carbon K-edge absorption spectra were interpreted in terms of different contributions coming from chemically shifted C 1s orbitals of the nonequivalent carbon atoms on the inner ring of the molecule formed by the sequence of CN bonds and on the benzene rings, respectively, and also in terms of different electronic distributions of the excited lowest unoccupied molecular orbital (LUMO) and LUMO+1. In particular, the degenerate LUMO appears to be mostly localized on the inner pyrrole ring. 相似文献

Oxytetracycline (OXY) is a broad-range antimicrobial routinely used in pig production, at doses in the range of few g/kg of medicated feed, during the weaning period. It could persist at ppm level in pig liquid manure that routinely is used for organic fertilisation. In the present work we describe a methodology to study OXY environmental fate in arable land where crops are cultivated for animal feeding purposes. A liquid-liquid extraction followed by metal chelate affinity chromatography was applied to environmental samples of manures and soils drawn within a case-control study. Extracts were then analysed by high performance liquid chromatography with UV/DAD detection, using a reverse phase column, and expressing the results as 4-epioxytetracycline epimer. Results indicate OXY is well retained at mg kg(-1) levels in soil exposed to contaminated pig manure fertilisation. Such compartment could constitute an abiotic reservoir for the systemic and/or for the external contamination of corn. 相似文献

The simulation of charged and/or strongly polar solutes represents a challenge for standard molecular-dynamics techniques. The use of periodic boundary conditions (PBCs) leads to artifacts due to the interaction between two replicas in the presence of the long-range Coulomb forces. A way to avoid these problems is the use of nonperiodic boundary conditions. A possible realization is to consider a finite system, a sphere, embedded in a reaction field described by the method of the images. In the present work the modified image approximation has been implemented in a molecular-dynamics code and optimized for the use of two standard solvents, water and acetonitrile. The methodology has then been applied to investigate the conformational changes in water-solvated alanine dipeptide. The free-energy surface calculated with this method is comparable to that obtained with PBC. 相似文献

[reaction: see text] Electrochemical oxidation of meta-substituted diphenylmethylidenefluorenes (3a-g) results in the formation of fluorenylidene dications that are shown to be antiaromatic through calculation of the nucleus independent chemical shift (NICS) for the 5- and 6-membered rings of the fluorenyl system. There is a strong linear correlation between the redox potential for the dication and both the calculated NICS and sigma(m). Redox potentials for formation of dications of analogously substituted tetraphenylethylenes shows that, with the exception of the p-methyl derivative, the redox potentials for these dications are less positive than for formation of the dications of 3a-g and for dications of p-substituted diphenylmethylidenefluorenes, 2a-g. The greater instability of dications of 2a-g and 3a-g compared to the reference system implies their antiaromaticity, which is supported by the positive NICS values. The redox potentials for formation of the dications of meta-substituted diphenylmethylidenes (3a-g) are more positive than for the formation of dications of para-substituted diphenylmethylidenes (2a-g), indicating their greater thermodynamic instability. The NICS values for dications of 3a-g are more antiaromatic than for dications of 2a-g, which is consistent with their greater instability of the dications of 3a-g. Although the substituted diphenylmethyl systems are not able to interact with the fluorenyl system through resonance because of their geometry, they are able to moderate the antiaromaticity of the fluorenyl cationic system. Two models have been suggested for this interaction, sigma to p donation and the ability of the charge on the substituted ring system to affect delocalization. Examination of bond lengths shows very limited variation, which argues against sigma to p donation in these systems. A strong correlation between NICS and sigma constants suggests that factors that affect the magnitude of the charge on the benzylic (alpha) carbon of the diphenylmethyl cation affect the antiaromaticity of the fluorenyl cation. Calculated atomic charges on carbons 1-8 and 10-13 show an increase in positive charge, and therefore greater delocalization of charge in the fluorenyl system, with increasing electronegativity of the substituent. The change in the amount of positive charge correlated strongly with NICS, supporting the model in which the amount of delocalization of charge is related to the antiaromaticity of the species. Thus, both aromatic and antiaromatic species are characterized by extensive delocalization of electron density. 相似文献
