Catalytic asymmetric allylic oxidation of cyclic olefins ocurrs for the first time in very high (94-99% ee) enantioselectivity using copper(I) complexes of malonyl derived bisoxazolines and tert-butyl p-nitroperbenzoate giving allyl benzoates in moderate yield. The copper complex, 15 mol %, was used in acetonitrile at -20 degrees C over an extended period, 5-12 d, with excess olefin together with one equivalent of perester. The S-esters were generated in accord with the model proposed previously for the (S,S)-bisoxazoline ligand. An eta2 intermediate was ruled out using low-temperature 13C NMR with the complex in the presence of olefin.  相似文献

We report on a search forv μ-induced events where the single emerging muon carries lepton number opposite that of the incident neutrino. The rate and kinematic quantities of the candidate events are compared with known backgrounds from \(\bar v_\mu \) -induced charged current interactions and ν-induced interactions that produce dileptons. We derive an upper limit on the rate of wrong-sign single muon production relative to the rate ofv μ charged current interactions to be 1.6×10?4 fory<0.5 and 3.1×10?4 fory>0.5 (90% CL). These upper limits enable us to constrain exotic sources of wrong-sign muons such as the charm component of the nucleon sea, flavor changing neutral currents and lepton number violating processes. Finally, the rate and kinematic properties of these events are compared with those of the neutrino-induced opposite-sign dimuon events.  相似文献

The structure-based design of multivalent ligands offers an attractive strategy toward high affinity protein inhibitors. The spatial arrangement of the receptor-binding sites of cholera toxin, the causative agent of the severe diarrheal disease cholera and a member of the AB(5) bacterial toxin family, provides the opportunity of designing branched multivalent ligands with 5-fold symmetry. Our modular synthesis enabled the construction of a family of complex ligands with five flexible arms each ending with a bivalent ligand. The largest of these ligands has a molecular weight of 10.6 kDa. These ligands are capable of simultaneously binding to two toxin B pentamer molecules with high affinity, thus blocking the receptor-binding process of cholera toxin. A more than million-fold improvement over the monovalent ligand in inhibitory power was achieved with the best branched decavalent ligand. This is better than the improvement observed earlier for the corresponding nonbranched pentavalent ligand. Dynamic light scattering studies demonstrate the formation of concentration-dependent unique 1:1 and 1:2 ligand/toxin complexes in solution with no sign of nonspecific aggregation. This is in complete agreement with a crystal structure of the branched multivalent ligand/toxin B pentamer complex solved at 1.45 A resolution that shows the specific 1:2 ligand/toxin complex formation in the solid state. These results reiterate the power of the structure-based design of multivalent protein ligands as a general strategy for achieving high affinity and potent inhibition.  相似文献

Amido analogs of geldanamycin, an ansamycin anticancer agent, were designed, synthesized, and assayed with SKBR3 cells, in which stability of HER2 receptor tyrosine kinase is dependent on the chaperone Hsp90. An amide was employed as a trisubstituted alkene isostere at the C8,9 position, which provided for a simplified, convergent synthesis through two major fragments, an aniline-amine left-hand portion and a dicarboxylic acid right-hand piece.  相似文献

The yttrium complex of 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetra(1′-13C-acetic acid) [13C]DOTA was synthesized. Fast dissolution dynamic nuclear polarization (DNP) nuclear magnetic resonance (NMR) studies revealed that the 89Y, 13C, and 15N nuclei present in the complex could be co-polarized at the same optimum microwave irradiation frequency. The liquid-state spin–lattice relaxation time T 1 of these nuclei were found to be reasonably long to preserve some or most of the DNP-enhanced polarization after dissolution. The hyperpolarized 13C and 89Y NMR signals were optimized in different glassing mixtures. The overall results are discussed in light of the thermal mixing model of DNP.  相似文献

This paper presents a comparative assessment of three types of electrical discharge reactors: 1) pulsed corona, 2) dielectric-barrier discharge, and 3) dielectric-pellet bed reactor. The emphasis is on the efficiency for electron-impact dissociation of N2(e+N2 →e+N+N) and the subsequent chemical reduction of NO by nitrogen atoms (N+NO→N2+O). By measuring the concentration of NO as a function of input energy density in dilute mixtures of NO in N2, it is possible to determine the specific energy cost for the dissociation of N2. Our experimental results show that the specific energy consumption (eV per NO molecule reduced) of different types of electrical discharge reactors are all similar. These results imply that, during radical production in electrical discharge reactors, the electric field experienced by the plasma is space-charge shielded to approximately the same value. The specific energy consumption for the dissociation of N2 using electrical discharge processing is measured to be around 240 eV per nitrogen atom produced. In the NO-N2 mixture, this corresponds to a specific energy consumption of around 240 eV per NO molecule reduced  相似文献

Hyperpolarized [1-(13)C]pyruvate has become an important diagnostic tracer of normal and aberrant cellular metabolism for in vitro and in vivo NMR spectroscopy (MRS) and imaging (MRI). In pursuit of achieving high NMR signal enhancements in dynamic nuclear polarization (DNP) experiments, we have performed an extensive investigation of the influence of Gd(3+) doping, a parameter previously reported to improve hyperpolarized NMR signals, on the DNP of this compound. [1-(13)C]Pyruvate samples were doped with varying amounts of Gd(3+) and fixed optimal concentrations of free radical polarizing agents commonly used in fast dissolution DNP: trityl OX063 (15 mM), 4-oxo-TEMPO (40 mM), and BDPA (40 mM). In general, we have observed three regions of interest, namely, (i) a monotonic increase in DNP-enhanced nuclear polarization P(dnp) upon increasing the Gd(3+) concentration until a certain threshold concentration c(1) (1-2 mM) is reached, (ii) a region of roughly constant maximum P(dnp) from c(1) until a concentration threshold c(2) (4-5 mM), and (iii) a monotonic decrease in P(dnp) at Gd(3+) concentration c > c(2). Of the three free radical polarizing agents used, trityl OX063 gave the best response to Gd(3+) doping, with a 300% increase in the solid-state nuclear polarization, whereas addition of the optimum Gd(3+) concentration on BDPA and 4-oxo-TEMPO-doped samples only yielded a relatively modest 5-20% increase in the base DNP-enhanced polarization. The increase in P(dnp) due to Gd(3+) doping is ascribed to the decrease in the electronic spin-lattice relaxation T(1e) of the free radical electrons, which plays a role in achieving lower spin temperature T(s) of the nuclear Zeeman system. These results are discussed qualitatively in terms of the spin temperature model of DNP.  相似文献

Cinchona phase-transfer catalysts (PTC) were developed for glycolate aldol reactions to give differentially protected 1,2-diol products. Silyl enol ether 9 reacted to generate benzhydryl-protected products. O-Allyl trifluorobenzyl cinchonium hydrofluoride CN-4 (20 mol %) catalyzed the addition of 9 to benzaldehyde to give 8 as a single syn-product in 76% yield and 80% ee. Recrystallization enriched the product to 95% ee, and a Baeyer-Villiger reaction transformed the product into useful ester intermediates. [reaction: see text]  相似文献

The antihypertension agent iodoazidoaryl prazosin (IAAP) has been made using a convergent route involving addition of an acylated piperazine 7 to 2-chloroquinazoline 5. IAAP has been shown to function as a multidrug resistance (MDR) reversal agent and bind to P-glycoprotein, a transmembrane transport protein. A study is also reported involving palladium-catalyzed substitution with amine heterocycles. With N,N-bis(2,6-diisopropyl)dihydroimidazolium chloride (10) as the ligand (2 mol %) for palladium(II) acetate (2 mol %) in THF at room temperature, morpholine added to 5 in 81% yield.  相似文献
