This article describes an electrochemical strategy to achieve low background‐current levels in horse‐radish peroxidase (HRP)‐based electrochemical immunosensors. The strategy consists of (i) the use of an HRP substrate/product redox couple whose formal potential is high and (ii) the use of an electrode that shows moderate electrocatalytic activity for the redox couple. The strategy is proved by a model biosensor using a catechol/o‐benzoquinone redox couple and an indium tin oxide (ITO) electrode. The combined effect of high formal potential and moderate electrocatalytic activity allows o‐benzoquinone electroreduction with minimal catechol electrooxidation and H2O2 electroreduction. The detection limit for mouse‐IgG is 100 pg/mL.  相似文献

We report a versatile platform for highly sensitive alkaline phosphatase (ALP)‐based electrochemical biosensors that uses an avidin‐modified indium tin oxide (ITO) electrode as a sensing electrode and 1‐naphthyl phosphate (NPP) as an ALP substrate. Almost no electrocatalytic activity of NPP and good electrocatalytic activity of 1‐naphthol (ALP product) on the ITO electrodes allow a high signal‐to‐background ratio. The effective surface covering of avidin on the ITO electrodes allows very low levels of nonspecific binding of proteins to the sensing electrodes. The platform technology is used to detect mouse IgG with a detection limit of 1.0 pg/mL.  相似文献

The reaction of 1 equiv of primary silanes, SiH(3)R (R = Ph, Mes), with [RhIr(CO)(3)(dppm)(2)] yields mono(silylene)-bridged complexes of the type [RhIr(H)(2)(CO)(2)(μ-SiHR)(dppm)(2)] (R = Ph or Mes), while for R = Ph the addition of 2 equiv yields the bis(silylene)-bridged complexes, [RhIr(CO)(2)(μ-SiHPh)(2)(dppm)(2)]. The kinetic isomer of this bis(silylene)-bridged product has the phenyl substituent axial on one silylene unit and equatorial on the other, and in the presence of excess silane this rearranges to the thermodynamically preferred "axial-axial" isomer, in which the phenyl substituents on each bridging silylene unit are axial and parallel to one another. The reaction of 1 equiv of diphenylsilane with [RhIr(CO)(3)(dppm)(2)] produces the mono(silylene)-bridged product, [RhIr(H)(2)(CO)(2)(μ-SiPh(2))(dppm)(2)], and the subsequent addition of silane in the presence of CO yields the silyl/silylene product [RhIr(H)(SiPh(2)H)(CO)(3)(κ(1)-dppm)(μ-SiPh(2))(dppm)]. The reaction of [RhIr(CO)(3)(dppm)(2)] with 2 equiv of SiH(2)Me(2) yields the analogous product [RhIr(H)(SiMe(2)H)(CO)(3)(κ(1)-dppm)(μ-SiMe(2))(dppm)]. Low-temperature NMR spectroscopic observation of some key intermediates, such as [RhIr(H)(SiH(2)Ph)(CO)(2)(μ-CO)(dppm)(2)], formed during the formation of the mono(silylene)-bridged species provides evidence for a mechanism involving initial Si-H bond activation at Rh, followed by the subsequent Si-H bond activation at Ir. The Si-H bond activation of a second equivalent of silane seems to be initiated by dissociation of the Rh-bound end of one diphosphine. The reaction of diphenylsilane with the cationic complex [RhIr(CH(3))(CO)(2)(dppm)(2)][CF(3)SO(3)] gives rise to a different reactivity pattern in which Si-H bond activation is initiated at Ir. In this case, the cationic silyl-bridged species, [RhIr(CH(3))(CO)(2)(κ(1):η(2)-SiHPh(2))(dppm)(2)][CF(3)SO(3)], contains an agostic Si-H interaction with Rh. In solution, at ambient temperature, this complex converts to two species, [RhIr(H)(COCH(3))(CO)(μ-H)(μ-SiPh(2))(dppm)(2)][CF(3)SO(3)] and [RhIr(CO)(2)(μ-H)(μ-SiPh(2))(dppm)(2)] [CF(3)SO(3)], formed by the competing methyl migration to CO and reductive elimination of methane, respectively. In the diphenylsilylene dihydride product, a weak interaction between the bridging silicon and the terminal Ir-bound hydride is proposed on the basis of NMR evidence.  相似文献

An analytical method of CE-MS and CE with an online preconcentration technique induced by a dynamic pH junction, addition of organic solvent and large volume injection was developed for sensitive determination of peptides in biological samples. Leucine enkephalin, methionine enkephalin, dynorphin A, β-endorphin and angiotensin II were used as model peptides. The optimal online preconcentration conditions were obtained at a sample matrix consisting of 100?mM borate buffer (pH 10.0) with 50% v/v acetonitrile and a BGE containing 1?M formic acid at pH 2.0, along with a 25-cm injection length. Under the optimized conditions, a 4.0×10(3)-1.1×10(4)-fold increase in peak intensity was achieved without degrading the peak shape. This online preconcentration method was applied to analyze the intracellular angiotensin II within the peptides extracted from HL1 cells and approximately increase of 1×10(4)-fold sensitivity was achieved compared to normal condition. Thus, the developed method could be applied to the analysis of various peptides for peptidomics study in biological samples.  相似文献

A simple, fast, repeatable and less laborious sample preparation protocol was developed and applied for the analysis of biocontrol fungus Trichoderma harzianum strain FA1132 by using gas chromatography-mass spectrometry. The match factors for sample spectra with respect to the mass spectra library of fungal volatile compounds were determined and used to study the complex hydrocarbons and other volatile compounds, which were separated by using different capillary columns with nonpolar, medium polar and high polar stationary phases. To date, more than 278 volatile compounds (with spectral match factor at least 90%) such as normal saturated hydrocarbons (C7-C30), cyclohexane, cyclopentane, fatty acids, alcohols, esters, sulfur-containing compounds, simple pyrane and benzene derivatives have been identified. Most of these compounds have not previously been reported. The method described in this paper is a more convenient research tool for the detection of volatile compounds from the cultures of T. harzianum.  相似文献

The bacterial reduction of Cr(VI) from industrial wastewater was evaluated using a 2.0-m3 bioreactor. Liquid pineapple waste was used as a nutrient for the biofilm community formed inside the bioreactor. The use of rubber wood sawdust as packing material was able to immobilize more than 106?CFU?mL?1 of Acinetobacter haemolyticus cells after 3?days of contact time. Complete reduction of 15?C240?mg?L?1 of Cr(VI) was achieved even after 3?months of bioreactor operation. Cr(VI) was not detected in the final effluent fraction indicating complete removal of Cr from solution from the flocculation/coagulation step and the unlikely re-oxidation of Cr(III) into Cr(VI). Impatiens balsamina L. and Gomphrena globosa L. showed better growth in the presence of soil?Csludge mixture compared to Coleus scutellarioides (L.) Benth. Significant amounts of Cr accumulated at different sections of the plants indicate its potential application in Cr phytoremediation effort. The bacterial-based system was also determined not to be detrimental to human health based on the low levels of Cr detected in the hair and nail samples of the plant operators. Thus, it can be said that bacterial-based Cr(VI) treatment system is a feasible alternative to the conventional system especially for lower Cr(VI) concentrations, where sludge generated can be used as growth supplement for ornamental plant as well as not detrimental to the health of the workers.  相似文献

A new end-off type acyclic ligand with four hydroxyethyl arms, 2,6-bis[bis(2-hydroxyethyl)aminomethyl]-4-methylphenol [H(bhmp)], formed dinuclear cobalt(II) complexes [Co(2)(bhmp)(OAc)(2)]BPh(4) (1) and [Co(2)(bhmp)(OBz)(2)]BPh(4) (2). The complex 1.2.5CH(3)CN (C(50)H(62.5)BCo(2)N(4.5)O(9)) crystallizes in the monoclinic space group C2/c with dimensions a = 25.424(5) A, b = 13.376(2) A, c = 29.913(6) A, beta = 105.930(3) degrees, and V = 9781(3) A(3) and with Z = 8. X-ray diffraction analysis revealed a mu-phenoxo-bis(mu-acetato)dicobalt(II) core structure containing two octahedral cobalt(II) ions. Electronic spectra were investigated for 1 and 2 in the range 400-1800 nm, and the data were typical for the octahedral high-spin cobalt(II) complexes. Magnetic susceptibility was measured for 1 and 2 over the temperature range 4.5-300 K, and the data were analyzed well using our theoretical method. The best fitting parameters were kappa = 0.77, lambda = -116 cm(-1), Delta = 572 cm(-1), and J = -0.44 cm(-1) for complex 1 and kappa = 0.96, lambda = -93 cm(-1), Delta = 616 cm(-1), and J = -0.33 cm(-1) for complex 2.  相似文献

We have concurrently studied the surface pressure (pi) versus area (A) isotherms and microscopic surface morphological features of Langmuir monolayers of diethylene glycol mono-n-octadecyl ether (C18E2) by film balance and Brewster angle microscopy (BAM) over a wide range of temperature. At temperatures < or =10 degrees C, the monolayers exist in the form of condensed phase even just after the evaporation of the spreading solvent, suggesting that the melting point of the condensed phase is above this temperature. At > or =15 degrees C, the monolayers can exist as gas (G), liquid expanded (LE), and liquid condensed (LC) phases and undergo a pressure-induced first-order phase transition between LE and LC phases showing a sharp cusp point followed by a plateau region in the pi-A isotherms. A variety of 2-D structures, depending on the subphase temperature, are observed by BAM just after the appearance of the cusp point. It is interesting to note here that the domains attain increasingly large and compact shape as the subphase temperature increases and finally give faceted structures with sharp edges and corners at > or =30 degrees C. The BAM observations were coupled with polarization modulation infrared reflection absorption spectroscopy (PM-IRRAS) to gain better understanding regarding the conformational order and subcell packing of the molecules. The constancy of the methylene stretching modes over the studied temperature range suggests that the hydrocarbon chains do not undergo any conformational changes upon compression of the monolayer. However, the full width at half-maximum (fwhm) values of the asymmetric methylene stretching mode (nu(as)(CH(2))) are found to respond differently with changes in temperature. It is concluded that even though the trans/gauche ratio of the hydrocarbon chains remains virtually constant, the LE-LC phase transition upon compression of the monolayer is accompanied by a loss of the rotational freedom of the molecules.  相似文献
