Linezolid is an antimicrobial agent for the treatment of multiresistant Gram‐positive infections. A practical high‐performance liquid chromatography method was developed for the determination of linezolid in human plasma and saliva. Linezolid and an internal standard (o‐ethoxybenzamide) were extracted from plasma and saliva with ethyl acetate and analyzed on a Capcell Pak C18 MG column with UV detection at 254 nm. The calibration curve was linear through the range 0.5–50 µg/mL using a 200 μL sample volume. The intra‐ and interday precisions were all <6.44% for plasma and 5.60% for saliva. The accuracies ranged from 98.8 to 110% for both matrices. The mean recoveries of linezolid were 80.8% for plasma and 79.0% for saliva. This method was used to determine the plasma and saliva concentrations of linezolid in healthy volunteers who were orally administered a 600 mg dose of linezolid. Our liquid–liquid extraction procedure is easy and requires a small volume of plasma or saliva (200 μL). This small volume can be advantageous in clinical pharmacokinetic studies, especially if children participate. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

Lolitrems are tremorgenic indole diterpenes that exhibit a unique 5/6 bicyclic system of the indole moiety. Although genetic analysis has indicated that the prenyltransferase LtmE and the cytochrome P450 LtmJ are involved in the construction of this unique structure, the detailed mechanism remains to be elucidated. Herein, we report the reconstitution of the biosynthetic pathway for lolitrems employing a recently established genome-editing technique for the expression host Aspergillus oryzae. Heterologous expression and bioconversion of the various intermediates revealed that LtmJ catalyzes multistep oxidation to furnish the lolitrem core. We also isolated the key reaction intermediate with an epoxyalcohol moiety. This observation allowed us to establish the mechanism of radical-induced cyclization, which was firmly supported by density functional theory calculations and a model experiment with a synthetic analogue.  相似文献

We designed and synthesized a new type of small helical molecule exhibiting intense circularly polarized luminescence (CPL) ( 12H ) by modifying a 20π-electron hemiporphyrazine with a large transition magnetic dipole moment. The hemiporphyrazine ring was opened and one additional pyridine unit was introduced, resulting in an overlap of two pyridine rings. X-ray structure analysis confirmed that 12H and its zinc complex ( 1Zn ) adopt a helical geometry. A racemic mixture of 1Zn was resolved into two enantiomers ((P)- and (M)- 1Zn ), which exhibited CPL with a high luminescence dissymmetry factor (glum) value of ±2.1×10−2. The origin of the large glum value was rationalized by means of DFT calculations. Helical structures could be formed in a diastereoselective manner by covalently attaching chiral units to the skeleton ( 1’2H and 1’Zn ). 1Zn was found to possess chiral recognition ability for amines.  相似文献

Herein we report a versatile Mizoroki–Heck-type photoinduced C(sp3)−N bond cleavage reaction. Under visible-light irradiation (455 nm, blue LEDs) at room temperature, alkyl Katritzky salts react smoothly with alkenes in a 1:1 molar ratio in the presence of 1.0 mol % of commercially available photoredox catalyst without the need for any base, affording the corresponding alkyl-substituted alkenes in good yields with broad functional-group compatibility. Notably, the E/Z-selectivity of the alkene products can be controlled by an appropriate choice of photoredox catalyst.  相似文献

Abstract

For improving process efficiency of polymer-sustained-alignment liquid crystal displays (PSA-LCDs), new photo-initiation monomers (PI-monomers), 2-methoxy-2-methacryloyl-oxy-ethoxy-2-phenylacetophenon (2-MMOEP) and 2,2-dimethacryloyl-oxy-ethoxy-2-phenylacetophenon (2,2-DMOEP), were developed and investigated image sticking characteristics. LC cells having PSA layers formed from a monomer 4,4′-dimethacryloyl-oxy-biphenyl (4,4′-DMABiph) and each PI-monomer were fabricated, and voltage holding ratio (VHR) and residual direct current voltage (VrDC) were clarified as the image sticking characteristics. The VHR and the VrDC were improved with the LC cells having the PSA layers formed from 4,4′-DMABiph and 2,2-DMOEP owing to large rate constant of polymerization. In contrast, the VHR and the VrDC were deteriorated with the LC cells having the PSA layers formed from 4,4′-DMABiph and 2-MMOEP. The deterioration of the VHR and the VrDC was derived from the increase in the concentration of radical and/or ion generated from 2-MMOEP. The results indicate that the number of polymerizable group in the PI-monomer is important, and plural number of polymerizable group in the PI-monomer leads to high VHR and low VrDC.  相似文献

The magnetic field effects (MFEs) on photoinduced hydrogen abstraction reactions between benzophenone and thiophenol in an ionic liquid, N,N,N-trimethyl-N-propylammonium bis(trifluoromethanesulfonyl) imide (TMPA TFSI), were studied by a nanosecond laser flash photolysis technique under ultrahigh fields of up to 28 T.
