Synthesis of 1-arylacenaphtho[1,2-d]pyrazoles and 5,7-Dehydro-5H,7H-benzo[b]acenaphtho[1,2-e]-1,3a,6a-triazapentalenes

2-Benzoyl- 5 and 2-acetylacenaphthenone 6 , prepared from the corresponding 1-acyl-2-(1-pyrrolidinyl)-acenaphthylenes 2 and 3 , reacted with arylhydrazines 8 under acidic conditions to give the corresponding 1-arylacenaphtho[1,2-d]pyrazoles 9 and 10 . Novel heteropentalene mesomeric betaines, 5,7-dehydro-5H,7H-benzo[b]acenaphtho[1,2-e]-1,3a,6a-triazapentalenes 13 and 14 were prepared by reductive cyclization of 1-(o-nitrophenyl)acenaphtho[1,2-d]pyrazoles 9d and 10d , respectively.     

Y-931, a novel atypical antipsychotic drug,is less sensitive to oxidative phenomena

The oxidation behavior of Y-931, a potent atypical antipsychotic drug, was compared with that of clozapine and olanzapine. In two enzymatic systems (horseradish peroxidase (HRP)/glutathione (GSH) and HRP/H(2)O(2)/GSH) which generate thiyl radicals, clozapine markedly strengthened the electron paramagnetic resonance (EPR) signal for the radical. Olanzapine, Y-931 and the major metabolites (compounds 1-3) had no or minimal effect on the intensity of this signal. In addition, the redox potential values for the three derivatives were in accord with the EPR spin trapping results. In toxicological experiments in human leukocytes, a concentration-dependent toxicity was observed when neutrophils were incubated with clozapine (1-10 micromol/l) and H(2)O(2) (1 mmol/l). However, Y-931 and olanzapine did not show remarkable toxicity under the conditions.     

Validation of the CALUX bioassay for the screening of PCDD/Fs and dioxin-like PCBs in retail fish

The chemical-activated luciferase expression (CALUX) assay is a reporter gene assay that detects dioxin-like compounds based on their ability to activate the aryl hydrocarbon receptor (AhR) and thus expression of the reporter gene. In this paper, the CALUX assay was examined for its application in the screening of polychlorinated dibenzo-p-dioxins (PCDDs), dibenzofurans (PCDFs) and dioxin-like polychlorinated biphenyls (dioxin-like PCBs) in retail fish. The sample extracts were cleaned up on a sulfuric acid-silica gel column followed by an activated carbon column, and the AhR activity of the separated PCDD/F and dioxin-like PCB fractions was determined using the assay. The quantitative limit for 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) was 0.98 pg ml(-1) (0.19 pg assay(-1) in the standard curve, corresponding to 0.16 pg g(-1) of CALUX-based toxic equivalency (2,3,7,8-TCDD equivalents) in the tested sample. Recovery tests in which dioxins were added to fish samples resulted in acceptable recoveries (77-117%). The CALUX assay performed well in the analysis of dioxins in fish samples and a comparative study revealed a strong correlation between the CALUX assay and high-resolution gas chromatography-high-resolution mass spectrometry analysis for the determination of PCDD/Fs (r = 0.89) and dioxin-like PCBs (r = 0.91) in retail fish (n = 22). These data revealed that the CALUX assay would be a useful screening method for PCDD/Fs and dioxin-like PCBs in retail fish.     

Effect of Static Magnetic Field on Photocatalytic Degradation of Methylene Blue over ZnO and TiO2 Powders

Magnetic field effects (MFEs) on photocatalytic degradation of methylene blue (MB) solution over ZnO and TiO₂ powders are investigated under static magnetic field up to 0.7 T with light irradiation by ultraviolet (UV) light emitting diode. The UV–visible-near-infrared spectrometer is used to monitor the MB concentrations. The positive MFE is observed for ZnO, while the negative MFE for TiO₂, and both MFEs are increased with the increase in the magnetic field applied. By increasing the settling time (the time interval between the preparation of MB solution and the powder dispersion into the MB solution), the photodegradation abilities under MFEs are decreased for both the catalysts. The cause of MFE is discussed in terms of dissolved oxygen in the MB solution and magnetic adsorption of the constituent molecules.     

RAFT synthesis of an amphiphilic block copolymer bearing chlorin rings in the hydrophobic segment and its application in photodynamic therapy

RAFT polymerization of styrene (St) in the presence of 5,10,15,20‐tetrakis(pentafluorophenyl)porphyrin (TFPP) was conducted using 4‐cyano‐4‐(thiobenzoyl)thiopentanoic acid as a chain‐transfer agent and azobisisobutyronitrile as an initiator at 60 °C. The resulting polymer exhibited a chlorin‐like UV‐vis spectrum, which indicated that the polymer possessed a reduced TFPP structure. Furthermore, an SEC trace recorded using UV‐vis detector (λ = 410 nm), which selectively detected the TFPP‐incorporated polymer, shifted toward higher molecular mass as the polymerization progressed. This evidence indicated that TFPP acted as a vinylene‐type monomer, such as maleimide, to form a copolymer, namely, poly(St‐co‐TFPP). The mole fraction of TFPP units was estimated to be 0.74 × 10^?3, which was close to that in the feed (1 × 10^?3). Chain extension of poly(St‐co‐TFPP) with polyethylene glycol monomethyl ether acrylate (PEGA) was performed to afford the amphiphilic block copolymer poly(St‐co‐TFPP)‐b‐poly(PEGA). The degrees of polymerization of St and PEGA were determined to be 64 and 75, respectively. Poly(St‐co‐TFPP)‐b‐poly(PEGA) formed micelles following dialysis. The median diameter of the micelles in solution was determined to be 16 nm by DLS. The photocytotoxicity of the micelle solution was evaluated in a human glioblastoma cell line (U251) and an N‐methyl‐N'‐nitro‐N‐nitrosoguanidine‐induced mutant of a rat murine RGM‐1 gastric carcinoma mucosal cell line (RGK‐1). © 2017 Wiley Periodicals, Inc. Synthetic Studies on Spirolides A and B: Formation of the Upper Carbon Framework Based on a Lewis Acid Template-Catalyzed Diels–Alder Reaction

The upper fragment of spirolides A and B, which are marine phycotoxins that exhibit strong antagonistic activities on nicotinic acetylcholine receptors, was constructed. The functionalized cyclohexene in spirolides was stereoselectively synthesized from the bicyclic lactone, which could be readily accessed by the Lewis acid template-catalyzed asymmetric Diels–Alder reaction of the pentadienol and methyl acrylate.     

Total Synthesis of Lajollamycin B

The first total synthesis of lajollamycin B, a structurally novel nitro-tetraene spiro-β-lactone/γ-lactone antibiotic, is described. The convergent synthesis involves the construction of the C8′–C11′ nitrodienylstannane and its coupling with the segment prepared from the C1′–C7′ ω-iodoheptadienoic acid and the right-hand heterocyclic fragment, which has been utilized for our previous syntheses of oxazolomycin A. The revision of the geometry of the terminal Δ^{10′, 11′}-double bond from E to Z is also described for the structure of natural lajollamycin B.
