The constitution and absolute configuration of the rhoeadine alkaloids (+)-alpinigenine and (+)-cis-alpinigenine. The fundamental structure of the hemi-acetal phenylbenzazepine alkaloid (+)-alpinigenine ( 1 ), isolated from Papaver bracteatum LINDL ., was derived essentially from 1H-NMR.- and mass-spectra of 1 and its derivatives 7, 10 and 14 (cf. Scheme 2). The positioning of the four methoxy groups in the two aromatic rings could be deduced from the 1H-NMR.-spectra of the N-oxides 14 and 15 in which, as a result of favourable sterical and conformational behaviour, an interaction exists between the N-oxide oxygen atom and one of the two ortho protons in ring C. The B/D-trans-fused 1 undergoes isomerization in 1N HCl to cis-alpinigenine ( 16 ). A stereochemical correlation between bases in the trans-and cis-series was enabled via an Emde degradation of the corresponding methylacetal-methyliodides 21 resp. 19 leading to the enantiomeric isochroman derivatives 22 resp. 23 which are achiral at C (2) (Scheme 4). The configuration at C (14) in the hemi-acetals (eg. 1 and 16 ) and the methyl ethers (eg. 7 and 8 ) is discussed in detail (cf. Scheme 7). (+)-Alpinigenine ( 1 ) has the (1S, 2R, 14R) configuration and (+)-cis-alpinigenine ( 16 ), in chloroform or acetone solution, the (1R, 2R, 14R) configuration.  相似文献

A two-step protocol for the first chemoselective cleavage of 2-hydroxy acid amides has been developed. Mesylation of the model substrate 2-(hydroxypropionylamino)-4-methylpentanoic acid methyl ester (11) followed by treatment with N-ethylthiourea (13) allows cleavage of 2-hydroxy acid amides under smooth conditions. Successful application of this methodology to the open-chain transesterification product 15 (methylester) of the cyclic heptadepsipeptide HUN-7293, a potent inhibitor of inducible cell adhesion molecule expression, delivered the corresponding hexapeptide 18 with unprotected N-terminus in 70-75% yield. This result demonstrates that the protocol developed even works in the presence of an ester and several methylated and unmethylated amide bonds. Finally, a sequence of ligation of methyl D-dehydroglutaminate (20) to the C-terminus of the saponification product 21, followed by the degradation protocol and ring closure, allowed chemical "point mutation" at the DGCN site affording the aza analogue of HUN-7293 (24) in 15% overall yield. To the best of our knowledge this is the first report on chemoselective cleavage of 2-hydroxy acid amides.  相似文献

Several independent synthetic routes are described leading to the formation of a novel unsaturated tetracyclic phosphorus carbon cage compound tBu4C4P6 (1), which undergoes a light-induced valence isomerization to produce the first hexaphosphapentaprismane cage tBu4C4P6 (2). A second unsaturated isomer tBu4C4P6 (9) of 1 and the bis-[W(CO)5] complex 13 of 1 are stable towards similar isomerization reactions. Another starting material for the synthesis of the hexaphosphapentaprismane cage tBu4C4P6 (2) is the trimeric mercury complex [(tBu4C4P6)Hg]3 (11), which undergoes elimination of mercury to afford the title compound 2. Single-crystal X-ray structural determinations have been carried out on compounds 1, 2, 9, 11, and 13.  相似文献

Fluoro-organic Syntheses VI: The Solvolysis of Chlorofluoro-cyclopropanes 2-Fluoro-allylic carbocations, generated from 1-chloro-1-fluoro-cyclopropanes or 2-fluoro-allyl p-toluene-sulfonates in water or acetic acid, undergo either proton loss or addition of hydroxyl or acetoxyl. In the latter case, an alkyl-substituted 2-fluoro-allyl ion leads predominantly to the more branched product (e.g. 3-fluoro-2-methyl-3-buten-2-ol) which may be converted into the less branched one (e.g. 2-fluoro-3-methyl-2-buten-2-ol) through reversible reactions.  相似文献

Abstract— The excited state behavior of the red light-absorbing form of phytochrome (Pr) was studied on the femtosecond time scale. After excitation of Pr with 75 fs laser pulses at 616 nm the kinetics of the transient absorption changes was recorded at selected wavelengths probing mainly the bleaching of the Pr ground-state absorption and the stimulated emission. The kinetic data obtained indicate the population of an excited state with a 3 ps lifetime immediately after excitation. This state precedes the formation of another excited state with a 32 ps lifetime. The decay of the latter state is followed by the appearance of a first product state that is assumed to represent lunii-R. In addition, 2,3-dihydrobiliverdin, which is considered to be an adequate model of the Pr chro-mophore, was included in the femtosecond studies. The absorption difference spectra recorded at various delay times show an immediate bleaching of the ground-state absorption. Simultaneously with bleaching a broad transient absorption appears between 410 and 525 nm. The data analysis yields similar kinetic components as they were observed in the decay of Pr. It is suggested from this finding that within the first tens of picoseconds after excitation the excited-state properties of Pr are mainly determined by the properties of the chromophore itself.  相似文献

Zusammenfassung Alkyl-und Dialkylamino-phenoxy-methylsilane ließen sich sowohl durch Umsetzung von Phenoxy-methyl-chlorsilanen mit primären und sekundären Aminen unter Cl/NRR-Austausch als auch durch Reaktion von Phenoxymethylsilanen mit substituierten Alkaliamiden unter C6H5O/NRR-Austausch gemäß den Gl. (1) bis (6) darstellen. Ihre Eigenschaften sind aus Tab. 1 zu ersehen.
We succeeded in preparing alkyl- and dialkylamino-phenoxymethylsilanes by reaction of phenoxy-methyl-chlorosilanes with primary and secondary amines (Cl/NRR-exchange) as well as by reaction of phenoxymethylsilanes with substituted alkaliamides (C6H5O/NRR-exchange)according equations (1) to (6). Their properties are to be seen in table 1.
