Five pathways leading to the deamination of cytosine (to uracil) after formation of its deprotonated radical cation are investigated in the gas phase, at the UB3LYP/6‐311G(d,p) level of theory, and in bulk aqueous solvent. The most favorable pathway involves hydrogen‐atom transfer from a water molecule to the N3 nitrogen of the deprotonated radical cation, followed by addition of the resulting hydroxyl radical to the C4 carbon of the cytosine derivative. Following protonation of the amino group (N4), the C4? N4 bond is broken with elimination of the NH3?+ radical and formation of a protonated uracil. The rate‐determining step of this mechanism is hydrogen‐atom transfer from a water molecule to the cytosine derivative. The associated free energy barrier is 70.2 kJ mol?1.  相似文献

An efficient and original stereocontrolled transannular rearrangement starting from activated 2,5-diketopiperazines has been developed, an opportunity for the medicinal chemistry field, which requests access to novel biological scaffolds. This powerful ring contraction, which can be related to a stereoselective aza-version of the Chan rearrangement, allows for example the one-step synthesis of various tetramic acids, access to 2-disubstituted statins, or the synthesis of relevant lactam-constrained dipeptide mimetics using a TRAL-RCM sequence.  相似文献

5',8-Purine cyclonucleosides constitute an important class of oxidatively generated tandem lesions whose formation involves initial hydroxyl radical-mediated hydrogen atom abstraction from the 5-hydroxymethyl group of 2-deoxyribose followed by intramolecular cyclization. The present study deals with the synthesis of the 5'S diastereomer of 5',8-cyclo-2'-deoxyadenosine containing di- and tri-oligodeoxynucleotides as an attempt to delineate the conformational changes induced in the DNA fragments by the presence of a rigid modified nucleoside. For this purpose, extensive 1D and 2D NMR measurements that were completed by DFT theoretical calculations were performed. As a striking result, it was found that the covalent bond between C(5') and C(8) in the investigated purine cyclonucleoside induces an unusual West ((0)T(1)) conformation of the furanose ring. Thus it can be postulated that the rigid structure of the tandem lesion would strongly perturb the global geometry of oligonucleotides at the site of the modification and therefore affect the enzymatic activity of DNA polymerases and repair enzymes.  相似文献

The role of local geometric and stereo-electronic effects in tuning the preference for different cross-linked adducts between thymine and purinic bases has been analyzed by a computational approach rooted in density functional theory. Our study points out that G--T and T--G tandem lesions are produced according to the same mechanism as A--T and T--A intrastrand adducts, and in both cases purine--T adducts are preferred rather than the opposite sequences. Moreover, use of conceptual DFT tools allows the rationalization of the preferential occurrence of G--T and T--G tandem lesions in place of their A--T and T--A counterparts.  相似文献

In this paper, we present the results of simultaneous screening of eight gibberellins (GAs) in coconut (Cocos nucifera L.) water by MEKC directly coupled to ESI-MS detection. During the development of MEKC-MS, partial filling (PF) was used to prevent the micelles from reaching the mass spectrometer as this is detrimental to the MS signal, and a cationic surfactant, cetyltrimethylammonium hydroxide, was added to the electrolyte to reverse the EOF. On the basis of the resolution of the neighboring peaks, different parameters (i.e., the pH and concentration of buffer, surfactant concentrations, length of the injected micellar plug, organic modifier, and applied separation voltage) were optimized to achieve a satisfactory PF-MEKC separation of eight GA standards. Under optimum conditions, a baseline separation of GA standards, including GA1, GA3, GA5, GA6, GA7, GA9, GA12, and GA13, was accomplished within 25 min. Satisfactory results were obtained in terms of precision (RSD of migration time below 0.9%), sensitivity (LODs in the range of 0.8-1.9 microM) and linearity (R2 between 0.981 and 0.997). MS/MS with multiple reaction monitoring (MRM) detection was carried out to obtain sufficient selectivity. PF-MEKC-MS/MS allowed the direct identification and confirmation of the GAs presented in coconut water (CW) sample after SPE, while, the quantitative analysis of GAs was performed by PF-MEKC-MS approach. GA1 and GA3 were successfully detected and quantified in CW. It is anticipated that the current PF-MEKC-MS method can be applicable to analyze GAs in a wide range of biological samples.  相似文献
