A rapid, sensitive and specific method to quantify ticlopidine in human plasma using clopidogrel as the internal standard (IS) is described. The analyte and the IS were extracted from acidified plasma by liquid-liquid extraction using diethyl ether-hexane (80 : 20, v/v). The extracts were analyzed by high-performance liquid chromatography coupled to electrospray tandem mass spectrometry (HPLC/MS/MS). Chromatography was performed isocratically on a Jones Genesis C(8) 4 microm analytical column (150 x 4.1 mm i.d.). The method had a chromatographic run time of 3.0 min and a linear calibration curve over the range 1.0-1000 ng ml(-1) (r(2) > 0.999427). The limit of quantification was 1.0 ng ml(-1). This HPLC/MS/MS procedure was used to assess the bioequivalence of two ticlopidine 250 mg tablet formulations (ticlopidine test formulation from Apotex do Brasil, Brazil, and Ticlid from Sanofi-Synthelabo, standard reference formulation). A single 250 mg dose of each formulation was administered to healthy volunteers. The study was conducted using an open, randomized, two-period crossover design with a 2 week washout interval. Since the 90% confidence interval for C(max) and area under the curve ratios were all inside the 80-125% interval proposed by the US Food and Drug Administration, it was concluded that ticlopidine formulation from Apotex do Brasil is bioequivalent to Ticlid formulation with respect to both the rate and the extent of absorption.  相似文献

A square-wave voltammetric (SWV) method using a hanging mercury drop electrode (HMDE) has been developed for determination of the herbicide molinate in a biodegradation process. The method is based on controlled adsorptive accumulation of molinate for 10 s at a potential of –0.8 V versus AgCl/Ag. An anodic peak, due to oxidation of the adsorbed pesticide, was observed in the cyclic voltammogram at ca. –0.320 V versus AgCl/Ag; a very small cathodic peak was also detected. The SWV calibration plot was established to be linear in the range 5.0×10–6 to 9.0×10–6 mol L–1; this corresponded to a detection limit of 3.5×10–8 mol L–1. This electroanalytical method was used to monitor the decrease of molinate concentration in river waters along a biodegradation process using a bacterial mixed culture. The results achieved with this voltammetric method were compared with those obtained by use of a chromatographic method (HPLC–UV) and no significant statistical differences were observed.  相似文献

A simple method for benzene, toluene, and xylenes (BTX) quantitative analyses in human urine was developed, using headspace solid-phase microextraction (HS-SPME) and gas chromatography coupled to mass spectrometry detection in the single ion monitoring mode. The developed method is solventless, non-invasive, requires small volume of sample (1 ml), shows high selectivity, sensitivity, repeatability, and linearity (correlation coefficients >0.998), providing a useful alternative to assess human exposure to BTX compounds due to occupational reasons or eventual exposure to organic solvents. Detection limit varies from 0.28 to 0.5 ppb (v/v).  相似文献

On the basis of its electrochemical behaviour a new flow-injection analysis (FIA) method with amperometric detection has been developed for quantification of the herbicide bentazone (BTZ) in estuarine waters. Standard solutions and samples (200 microL) were injected into a water carrier stream and both pH and ionic strength were automatically adjusted inside the manifold. Optimization of critical FIA conditions indicated that the best analytical results were obtained at an oxidation potential of 1.10 V, pH 4.5, and an overall flow-rate of 2.4 mL min(-1). Analysis of real samples was performed by means of calibration curves over the concentration range 2.5x10(-6) to 5.0x10(-5) mol L(-1), and results were compared with those obtained by use of an independent method (HPLC). The accuracy of the amperometric determinations was ascertained; errors relative to the comparison method were below 4% and sampling rates were approximately 100 samples h(-1). The repeatability of the proposed method was calculated by assessing the relative standard deviation (%) of ten consecutive determinations of one sample; the value obtained was 2.1%.  相似文献

The IAEA-331 spinach material NIST SRM 1570a Spinach Leaves, submitted to an intercomparison run by the IAEA, has been analysed by k0-based INAA with counting using both low and high energy photon detectors. The results have been compared with (i) the certified values of the NIST SRM 1570a; the agreement is good, taking into account the uncertainties; and (ii) the certified and consensus values of the former NIST SRM 1570 Spinach Leaves, their composition in minor and trace-elements are quite similar; however a lower content has been observed for Co, Fe and Sc in the IAEA-331 and for Zn in the SRM 1570. For quality control, the NIST 1573 Tomato Leaves and the NIST 1575 Pine Needles have been analysed using the same conditions as for the IAEA-331. The results agree quite well with the certified and consensus values given in the literature.  相似文献

A methodology was developed for the separation and determination of microamounts of mercury from copper concentrate samples by wavelength dispersive X-ray fluorescence (WDXRF) after solid-phase extraction of mercury from iodide medium using polyurethane foam (PUF). The best sorption conditions for the Hg-KI-PUF system were settled using X-ray fluorescence technique after collection of ground PUF on a filter paper by vacuum filtration and direct measurement of the intensity signal of the sorbed mercury on PUF. The main parameters of sorption such as iodide concentration, pH, shaking time and sample dilution effect were studied. The system shows rapid kinetic sorption and maximum X-ray intensity signal was achieved after shaking for 2 min a 0.01 mol l−1 iodide solution containing microamounts of mercury in the pH range from 1.0 to 9.0. Effective sorption up to a volume of 0.9 l allows preconcentration of mercury. A linear fit up to 50 μg mercury was obtained by the plot of the initial mercury mass in the bulk solution (0.5 l) vs. its respective XRF intensity signal measurement on ground PUF after the sorption process. The calibration sensitivity, quantification and detection limits found were 9.09 CPS μg−1, 9.0 and 2.7 μg, respectively. The sorption of many elements was also evaluated under the best conditions. High concentrations of Cu(II) and Fe(III) interfere seriously. Mercury-selective separation could be achieved using citrate or EDTA as masking agent; no interference due to copper matrix samples was observed in citrate medium. This methodology was evaluated by recovery for mercury determination in copper concentrate ore samples supplied by a mining industry and copper sulfate salts; the results were between 98% and 106%.  相似文献
