This paper reports the surface activity of phytase at the air-water interface, its interaction with lipid monolayers, and the construction of a new phytic acid biosensor on the basis of the Langmuir-Blodgett (LB) technique. Phytase was inserted in the subphase solution of dipalmitoylphosphatidylglycerol (DPPG) Langmuir monolayers, and its incorporation to the air-water interface was monitored with surface pressure measurements. Phytase was able to incorporate into DPPG monolayers even at high surface pressures, ca. 30 mN/m, under controlled ionic strength, pH, and temperature. Mixed Langmuir monolayers of phytase and DPPG were characterized by surface pressure-area and surface potential-area isotherms, and the presence of the enzyme provided an expansion in the monolayers (when compared to the pure lipid at the interface). The enzyme incorporation also led to significant changes in the equilibrium surface compressibility (in-plane elasticity), especially in liquid-expanded and liquid-condensed regions. The dynamic surface elasticity for phytase-containing interfaces was investigated using harmonic oscillation and axisymmetric drop shape analysis. The insertion of the enzyme at DPPG monolayers caused an increase in the dynamic surface elasticity at 30 mN m(-)(1), indicating a strong interaction between the enzyme and lipid molecules at a high-surface packing. Langmuir-Blodgett (LB) films containing 35 layers of mixed phytase-DPPG were characterized by ultraviolet-visible and fluorescence spectroscopy and crystal quartz microbalance nanogravimetry. The ability in detecting phytic acid was studied with voltammetric measurements.  相似文献

The thermodynamic properties G h o,H h o, and C p,h oassociated with the transfer of non-ionic organic compounds from gas to dilute aqueous solution and the limiting partial molar properties C p o ,2 and V2 2 of these compounds in water are described through a simple scheme of group contributions. A distinction is made between groups made only of carbon and hydrogen, and functional groups i.e. groups containing at least one atom different from carbon and hydrogen. Each group is assigned a contribution, for each property, through a least squares procedure which utilizes only molecules containing at most one functional group. Finally, for compounds containing more than one functional group, correction parameters are evaluated as the differences between the experimental values and those calculated by means of the group contributions. The different behavior of hydrophilic compared with hydrophobic groups is discussed for the various properties. A rationale for the correction parameters, i.e. for the effects of the interactions among hydrophilic groups on the thermodynamic properties, is attempted.  相似文献

A fast atom bombardment (FAB) mass spectrometric study on the open-chain compound 1,3-bis(cyanomethylenetriphenylphosphorane)propane-1,3-dione and on the cyclic zwitterionic compounds 4-oxy-5-triphenylphosphonium-6-methyl-2-pyrone and 4-oxy-5-triphenylphosphonium-6-phenyl-2-pyrone, obtained by reaction of carbon suboxide, C3O2, with stabilized phosphorus ylides, Ph3PCHX (X?CN, COMe, COPh), is described. The FAB mass spectrometric behaviour of these compounds is compared with that shown by tri-phenylphosphoranilideneketene, Ph3P ? C ? C ? O, and by 4-hydroxy-6-methyl-2-pyrone, with the aid of metastable ion data and collision spectroscopy.  相似文献

The behavior of 5,6-dihydro-4H-pyrido[3,2,1-jk]carbazol-4-one (10) , 1,2,7,8-tetrahydro-3H-quino[1,8-ab][1]benzazepin-3-one (11) , 1,2-dihydro-9H-[1]benzazepino[1,9-ab] [4,1]benzoxazepin-4 (3H)one (13) , and 1,2-dihydro-8H-[1]benzazepino[1,9-cd] [1,5]benzoxazepin-4(3H)one (14) towards the Schmidt reaction has been determined in polyphosphoric acid and in benzeneor chloroform-sulfuric acid. A new apparatus for the extraction of organic compounds from sea water is described. With this apparatus it is possible to extract 54 standard compounds with high recovery percentages from 9 1 of sea water with 3 ml ofn-hexane. The analysis time (about 1 h) is appreciably lower than those of the extraction methods based on RP-18 and Carbopack-B adsorption (about 15 h). Furthermore, it is possible to analyze samples without filtration. An application of this method to the analysis of Tirreno sea water is reported.  相似文献

The CuA center is a dinuclear copper site that serves as an optimized hub for long‐range electron transfer in heme–copper terminal oxidases. Its electronic structure can be described in terms of a σu* ground‐state wavefunction with an alternative, less populated ground state of πu symmetry, which is thermally accessible. It is now shown that second‐sphere mutations in the CuA containing subunit of Thermus thermophilus ba3 oxidase perturb the electronic structure, which leads to a substantial increase in the population of the πu state, as shown by different spectroscopic methods. This perturbation does not affect the redox potential of the metal site, and despite an increase in the reorganization energy, it is not detrimental to the electron‐transfer kinetics. The mutations were achieved by replacing the loops that are involved in protein–protein interactions with cytochrome c, suggesting that transient protein binding could also elicit ground‐state switching in the oxidase, which enables alternative electron‐transfer pathways.  相似文献

Capillary zone electrophoresis (CZE) was used for characterisation of soil humic acid (HA) fractions obtained by coupling size-exclusion chromatography with polyacrylamide gel electrophoresis, on the basis of their molecular size and electrophoretic mobility. CZE was conducted using several low alkaline buffers as background electrolyte (BGE): 50 mM carbonate, pH 9.0; 50 mM phosphate, pH 8.5; 50 mM borate, pH 8.3; 50 mM Tris-borate+1 mM EDTA+7 M urea+0.1% sodium dodecyl sulphate (SDS), pH 8.3. Independently of BGE conditions, the effective electrophoretic mobility of HA fractions were in good agreement with their molecular size. The better resolution of HA were obtained in Tris-borate-EDTA buffer with urea and SDS. This results indicated that CZE, mostly with BGE-contained disaggregating agents, is useful for separating HAs in fractions with different molecular sizes.  相似文献

Abstract

The solution properties of the exopolysaccharide marginalan produced by Pseudomonas marginalis HT041B were investigated by means of low-angle laser light-scattering, capillary viscometry, and rheology. Potentiometric and viscosimetric data indicated the absence of a cooperative transition of the disorder-to-order type. The experimental findings obtained in dilute solution (Mark-Houwink coefficients, rigidity coefficient, characteristic ratio) suggested that the polymer behaves like a semiflexible chain which adopts a disordered conformation. The rheological behaviour of more concentrated marginalan solutions, as determined by means of both steady shear and oscillatory measurements, further confirmed the disordered conformational state of the polymer in solution.

Complete assignment of 1H and 13C NMR chemical shifts and J(1H/1H and 1H/19F) coupling constants for 22 1‐phenyl‐1H‐pyrazoles' derivates were performed using the concerted application of 1H 1D and 1H, 13C 2D gs‐HSQC and gs‐HMBC experiments. All 1‐phenyl‐1H‐pyrazoles' derivatives were synthesized as described by Finar and co‐workers. The formylated 1‐phenyl‐1H‐pyrazoles' derivatives were performed under Duff's conditions.
