Separation, identification, and interaction of heparin oligosaccharides with granulocyte-colony stimulating factor using capillary electrophoresis and mass spectrometry

A capillary electrophoresis (CE) method was developed for the separation of heparin oligosaccharides compatible to study the interactions between the oligosaccharides and granulocyte-colony stimulating factor (G-CSF). Unfractionated heparin was eliminitively degraded to heparin oligosaccharides by an endolytic heparinase. The degraded smaller oligosaccharides (M(r) < 1000) were baseline-separated by CE under a 50 mM phosphate buffer (pH 9.0) in 10 min. Standard heparin disaccharides and larger oligosaccharides (1000 < M(r) < 8000) were all separated under optimized separation conditions. Compared with standard heparin disaccharides, smaller oligosaccharides contained one nonsulfated, two monosulfated, and two disulfated disaccharides, but trisulfated disaccharides were not found. The smaller oligosaccharides were also identified and molecular mass was deduced by electrospray ionization-mass spectrometry (ESI-MS). Furthermore, interactions between G-CSF and the oligosaccharides were studied by using capillary zone electrophoresis (CZE) under the above separation conditions. It was found that larger oligosaccharides could interact with G-CSF while smaller oligosaccharides were not observed to bind to G-CSF under the experimental conditions. In conclusion, the purified heparinase could selectively degrade heparin into oligosaccharides and the interaction between G-CSF and heparin was correlated with the chain length of heparin.     

Growth of porous single-crystal Cr2O3 in a 3-D mesopore system

Theoretical studies of dissociative phosphoryl transfer in interconversion of phosphoenolpyruvate to phosphonopyruvate: solvent effects, thio effects, and implications for enzymatic reactions

The conversion of phosphoenolpyruvate (PEP) to phosphonopyruvate (P-pyr) is catalyzed by PEP mutase via a dissociative mechanism. In this work, we investigate the uncatalyzed reaction using ab initio methods, density functional theory, and the semiempirical MNDO/d method. Comparisons of geometries and relative energies of stationary points (minima and transition states) with density functional results indicate that the semiempirical method is reasonably accurate. Solvent effects are examined using implicit solvent models, including the recently extended smooth conductor-like screening model. Due to the large negative charge carried by the system, solvation is found to drastically alter the location and energy of stationary points along the dissociative reaction pathways. The influence of substituting a nonbridging phosphoryl oxygen by sulfur (thio effects) was also investigated. Implications of these results for the enzymatic reaction are discussed.     

液-液萃取电感耦合等离子体质谱法测定天然水中痕量稀土元素

建立了以单、二（2－乙基己基）磷酸酯混合螯合剂的庚烷溶液作为萃取剂，6.0mol/L HCl反萃取预分离富集，In和Rh做内,ICP-MS测定天然水中15种稀土元素的分析方法。方法的富集倍数为200倍，各元素的检出限范围在0.01-1.8ng/L之间，相对标准偏差RSD＜5％，萃取回收率＞93％。

Zs－93硒剂对移植性小鼠S＿（180）肉瘤的抑制作用及其免疫活性

用硒与尿素化合，新生的由杂环有机硒所组成的Ｚｓ－９３硒剂治疗移植性Ｓ１８０肉瘤，有显著的抑瘤作用（Ｐ＜０．０１），其作用表现出一定的剂量效应，以５μｇ／只小鼠为最适，抑瘤率达６０．７６％，治疗组小鼠的Ｔ细胞、ＳＯＤ活性均有增强。     

硒与消化系统癌关系的探讨

据不同地貌、地质条件在30个县市按剖面、同步、随机取土壤样品372份．测定硒等微量元素含量．作等级相关分析显示。土壤硒含量与消化系统癌呈负相关关系．作相对危险度、剂量-效应及多因素分析,认为此负相关关系并非偶然现象．     

镓改性ZSM-5沸石中镓活性中心的形成与表征

采用脉冲微反体系研究了经不同程度还原和不同镓含量的GaZSM-5沸石的芳构化性能与吡啶吸附红外光谱特征谱带吸收度的关联，证实了1458 cm~(-1)吸收带的用于表征与Ga物种有关的活性中心，并研究了沸石的硅铝比对Ga中心形成和Ga的价态对芳构化性能的影响。结果表明，对Ga含量相同的样品来说，硅铝比愈低，起始Ga中心生成量愈多，高价态与低价态的Ga中心对丙烷芳构化反应都有促进作用。     

叔醇脱水成烯反应的溶剂化能力与介电常数的关系

我们用Fe_2(SO_4)_3·xH_2O为催化剂,对2-甲基-2-已醇脱水生成烯烃反应的溶剂化能力及其动力学做了研究,测定了20种不同溶剂中的反应速率常数和以丙至辛6种直链醇为溶剂于不同温度下反应的速率常数,确定了溶剂化能力与溶剂介电常数之间的线性关系。这方面工作尚未见报道。     

Development of ELISA and immunochromatographic assay for ofloxacin

Two rapid,sensitive and reliable immunoassay methods,namely competitive indirect enzyme-linked immunosorbent assay(CI- ELISA)and colloidal gold-based immunochromatographic assay(CGIA),were developed to detect ofloxacin(OFL).The linear range of the CI-ELISAwas from 0.5 to 128 ng/mL with a limit of detection(LOD)of 0.35 ng/mL.Good recoveries were obtained in analyzing simulated swine urine samples.The CGIA could accurately estimate OFL at concentrations as low as 10 ng/mL in less than 10 min,and test results were read visually without any instrument.
