Fully automated polymer-assisted synthesis of 1,5-biaryl pyrazoles

The polymer-assisted solution-phase (PASP) synthesis of a 192-member 2-D array of 1,5-biaryl pyrazoles 4[1-12,1-16] is reported. The synthesis was performed in a fully automated manner using a multiprobe top-filtration robot and incorporates a "catch and release" step to afford library compounds directly in high yield and purity.     

Synthesis of methyl-1,6-dioxaspiro[4,5]decanes using organoselenium mediated cyclization reactions

Three naturally occurring methyl-1,6-dioxaspiro[4.5]decanes have been prepared in good yield using organoselenium mediated reactions during the crucial cyclization process.     

Spectroscopy of excited state positronium

Electric dipole transitions in the microwave range have been induced between the fine-structure levels of positronium in the excited staten=2. As an indication of the transitions, we used the increase in Lyman- radiation when the metastable 2³S₁-level is depopulated. The results for the transitions 2³S₁2³P_0,1,2 are ₀=18499.65±1.20±4.00 MHz, ₁=13012.42 ±0.65±1.54 MHz and ₂=8624.38±0.54±1.40 MHz. The first error is statistical and the second systematic. The precision of the present measurement has improved by a factor of 3, compared to previous data. Recent bound state QED-calculations have been extended to the orderR _t8 ⁴ln ^–1. The not yet completely calculated orderR _t8 ⁴ is estimated to contribute less than 1 MHz. Our experimental results are in good agreement with theory. By applying a weak magnetic field, we were able to observe the transition 2³S₁2¹P₁ which is strictly forbidden byC-invariance in zero field. Our result, corrected for Zeeman- and motional Starkeffect, is ₃=11180.0±5.0±4.0 MHz. An upper limit for theC-violating matrix element of MHz could be deduced. Our experiment used moderated slow positrons from the bremsstrahlung and pair production of a pulsed electron linear accelerator (TEPOS facility at the university of Giessen).     

Synthesis of trifluoromethyl ketones using polymer-supported reagents

A two step synthesis of trifluoromethyl ketones from aldehydes is reported. A combination of polymer-supported reagents and sequestering agents were employed to effect the transformation without the need for chromatographic purification.     

Total synthesis of the polyenoyltetramic acid polycephalin C

The total synthesis of the polyenoyltetramic acid polycephalin C is described. Key steps of the synthesis include a double Swern oxidation, double Takai reaction and a double Stille reaction. In addition, the absolute stereochemistry of the ring junction has been determined by synthesis of both isomers and comparison of their CD spectra with natural polycephalin C.     

Versatile solid-phase synthesis of secondary amines from alcohols. Development of an N-Boc-(o-nitrobenzene)sulfonamide linker

The development of a versatile amine releasing linker based on the modified o-nitrobenzene sulfonamide protective group is described. This new N-Boc-o-nitrobenzenesulfonamide (Boc-ONBS) linker enables the elaboration on resin of primary and secondary amines by sequential substitution of the sulfonamide moiety using the Mitsunobu reaction. A 16-member array of secondary and Boc protected primary amines was then prepared using this linker.     

Photoreactivation of cyclobutane dimers and (6-4) photoproducts in the epidermis of the marsupial, Monodelphis domestica

Radioimmunoassays were used to investigate the repair of cyclobutane pyrimidine dimers and pyrimidine (6-4)pyrimidone photoproducts ((6-4] photoproducts) in the epidermis of the South American opossum, Monodelphis domestica. In the absence of photoreactivating light, both types of photodamage were excised with similar kinetics, 50% of the damage remaining 8 h after UV irradiation in vivo. Exposure of UV-irradiated skin to photoreactivating light resulted in removal of most of the cyclobutane dimers and an enhanced rate of (6-4) photoproduct repair. Photoenhanced excision repair of non-dimer damage increases the range of biologically effective lesions removed by in vivo photoreactivation.     

Synthesis of the β-lactam antibiotic (+)- thienamycin via an intermediate π-allyltricarbonyliron lactone complex

The π-allyltricarbonyliron lactone complex (7), formed by reaction of E-1,2-epoxy-2-methyl-6,6-dimethoxyhex-3-ene(5) with co-ordinatively unsaturated iron carbonyl species, was reacted with benzylamine to give a lactam complex (8) by an S_N'-like mechanism. This complex upon oxidation with Ce(IV) afforded cis-3-isopropenyl-4-[(2',2'-dim (9) which was chemically modified into trans-3-(1'-hydroxyethyl)-4-[(2',2-dimethoxy)ethyl] azetidin-2-one (13), a key intermediate previously used in the synthesis of the antibiotic thienamycin. Similar reaction with (S)-(-)--methylbenzylamine afforded a separable mixture of diastereoisomeric iron lactam complexes (16 and 17). These complexes could be individually converted to the corresponding optically active β-lactam derivatives (27 and 28) and, hence, are precursors for the synthesis of either natural (+)-thienamycin or unnatural (-)-thienamycin.     

Modelling of polymer aggregates in solution

The self-consistent mean field model of Scheutjens and Fleer is used to model spherical aggregates of homopolymers and monomer—polymer particles in solution. For homopolymer aggregates we found that the chain ends are preferentially located at the exterior side of the polymer/solvent interface. The distribution of the end segments may be an important parameter in latex film formation. For monomer—polymer particles a “core-shell” structure is found with an extended core containing a monomer—polymer mixture and a thin shell a few nanometres thick strongly enriched with monomer. The monomer-enriched shell seems to function as a solvating envelope for the dangling chain ends. These results are compared with other simulations based on a single chain in a spherical-cavity model.
