A special type of glass discs—prepared of sodium tetraborate with varying concentrations of only one element—are used for the calibration of XRF spectrometers. The same glass discs are used for the determination of background influence coefficients Fi,j which makes it possible to calculate the background signal in the unknown samples concerning the analyte (i) when only the characteristic radiation from the interfering elements (j) are measured together with the i-signal on a blank disc of pure sodium tetraborate. For the first time the two enantiomeric forms of the glycosidase inhibitor 1-azafagomine have been synthesised starting from D- and L-xylose. D-Xylose was converted to the 2,3,5-tribenzylfuranose, which upon reductive amination with tert-butyl carbazate gave the protected 1-hydrazino-1-deoxypentitol in high yield. N-acetylation, mesylation of the 4-OH, removal of the Boc group, cyclisation and deprotection gave (+)-1-azafagomine ((+)-1). By a similar sequence of reactions, L-xylose was converted to (-)-1-azafagomine ((-)-1). Enzymatic and other routes to optically pure 1-azafagomine were also studied. Compound (-)-1 is a potent competitive glycosidase inhibitor, while (+)-1 has no biological activity. The inhibition of almond beta-glucosidase by (-)-1 was found to be slow owing to a slow binding step of inhibitor to enzyme, with no subsequent conformational rearrangement. The rate constants for binding and release were found to be 3.3 x 10(4)M(-1)s(-1) and 0.011 s(-1), respectively, yielding Ki = 0.33 microM. 相似文献

A method to define trivariate spline quasi-interpolation operators (QIOs) is developed by blending univariate and bivariate operators whose linear functionals allow oversampling. In this paper, we construct new operators based on univariate B-splines and bivariate box splines, exact on appropriate spaces of polynomials and having small infinity norms. An upper bound of the infinity norm for a general blending trivariate spline QIO is derived from the Bernstein-Bézier coefficients of the fundamental functions associated with the operators involved in the construction. The minimization of the resulting upper bound is then proposed and the existence of a solution is proved. Liquid-Liquid phase separation has emerged as fundamental process underlying the formation of biomolecular condensates. Insights into the composition and structure of biomolecular condensates is, however, complicated by their molecular complexity and dynamics. Here, we introduce an improved spatially-resolved NMR experiment that enables quantitative analysis of the physico-chemical composition of multi-component biomolecular condensates in equilibrium and label-free. Application of spatially-resolved NMR to condensates formed by the Alzheimer's disease-associated protein Tau demonstrates decreased water content, exclusion of the molecular crowding agent dextran, presence of a specific chemical environment of the small molecule DSS, and ≈150-fold increased concentration of Tau inside the condensate. The results suggest that spatially-resolved NMR can have a major impact in understanding the composition and physical chemistry of biomolecular condensates. 相似文献

Microalgae have been suggested as a potential source for new functional ingredients, making possible the development of new functional foods from natural origin. Among the natural ingredients, polyunsaturated fatty acids (PUFAs) have generally been identified as an interesting group of compounds with biological activity, mainly related to their anti-inflammatory properties. In this regard, the use of environmentally friendly extraction procedures (e.g. pressurized liquid extraction, PLE) to obtain such natural ingredients is also becoming necessary. In this work, an exhaustive characterization of the lipid fraction of a pressurized ethanolic extract of the microalga Spirulina platensis is carried out. To achieve this objective high-performance liquid chromatography (HPLC) coupled to quadrupole time-of-flight mass spectrometry (QTOF-MS) is employed. The use of the QTOF analyzer allows the selection and isolation of precursor ions as well as providing the high efficiency, sensitivity and mass accuracy required. By means of this powerful hyphenated technique, it was possible to identify several polar lipids in an extract of S. platensis (some of them, to our knowledge, described for the first time in this work), including four free fatty acids, four monogalactosyl monoacylglycerols, three phosphatidylglycerols and two sulfoquinovosyl diacylglycerols. 相似文献

A new, specially designed column has been developed for fractionation of supercritical fluid extract of rosemary by using a preparative supercritical fluid chromatography system (Prep-SFC). The column evaluated in this work was prepared using a new packing method consisting of a combination of slurry and supercritical CO2 with commercial silica particles coated with a stationary phase commonly used in gas chromatography, such as SE-54 (5% phenyl-, 95% methylsilicone). The new packing procedure provided columns with reasonable efficiencies, with high stability and useful at high-pressure range. A 25 cm x 10 mm i.d. column packed with silica particles coated with 3% of SE-54 was prepared, and its separation power was tested for isolating fractions with high antioxidant and/or antimicrobial activity from a supercritical rosemary extract. The SFC conditions were selected based on a previous work done with a commercial LC-Diol packed column (130 bar, 80 degrees C), and different percentages of modifier in the mobile phase were tested (5 and 10%). Two cyclones were employed to collect the fractions which were then characterized by HPLC-diode array detection (DAD), GC, and in vitro antioxidant and antimicrobial assays. The use of coated packed columns allowed the fractionation of a complex mixture of rosemary supercritical extract with a minimum amount of modifier in the mobile phase (5% ethanol). At the optimum conditions it was possible to obtain two very active fractions in terms of antioxidant and antimicrobial activity, with no residual rosemary aroma and with improved activities compared to the original supercritical extract. 相似文献
