Stability-indicating validated HPLC method for simultaneous determination of oral antidiabetic drugs from thiazolidinedione and sulfonylurea groups in combined dosage forms

For type 2 diabetes treatment, combinations of drugs from the thiazolidinedione and sulfonylurea groups are now available in the same tablet or capsule. Therefore, a stability-indicating and validated HPLC method was developed for simultaneous determination of pioglitazone, rosiglitazone, and glipizide in combined dosage forms. The examined drugs were subjected to different conditions such as acid and base, temperature, and UV light, and degradation of pioglitazone and glipizide was observed under thermal and acidic stress. However, selectivity of the presented method for pioglitazone, rosiglitazone, and glipizide assay against their degradation products was confirmed. It was also demonstrated to be robust, resisting small deliberate changes in pH of the buffer, flow rate, and percentage of acetonitrile in the mobile phase. The presented method utilizes a LiChrospher RP18 column (125 x 4.0 mm), acetonitrile in phosphate buffer at pH 4.3 (40 + 60, v/v) as the mobile phase, and UV detection at 225 nm for pioglitazonel glipizide or 245 nm for rosiglitazone/glipizide. The method was validated with respect to linearity, precision, and accuracy. Finally, the elaborated procedure was applied for the QC of pioglitazone/glipizide and rosiglitazone/glipizide mixtures.     

Structural diversity through ligand flexibility: two novel metal-organic nets via ligand-to-ligand cross-linking of "paddlewheels"

Solvothermal reaction of a partially flexible ligand, H(4)L, and Cu(NO(3))(2)·2.5H(2)O afforded two cross-linked Kagomé lattices of formula [Cu(2)(L)](n): an acs net sustained by novel trigonal prismatic supermolecular building blocks (SBBs) and the first example of a partially pillared Kagomé net.     

Influence of some crosslinking agents on thermal and mechanical properties of electron beam irradiated polylactide

The aim of this article was to determine and compare the influence of trimethylopropane trimethacylate (TMPTA) and trially isocyanurate (TAIC) crosslinking agents on thermal and mechanical properties of electron beam irradiated polylactide (PLA). The blends were made of PLA mixed with 3 wt% of TMPTA (PLA/TMPTA), and PLA mixed with 3 wt% of TAIC (PLA/TAIC). Injection moulded samples were irradiated with the use of high energy (10 MeV) electron beam at various radiation doses to crosslinking PLA macromolecules. Thermal and mechanical properties were investigated by means of differential scanning calorimetry (DSC), dynamic mechanical analysis (DMA), tensile strength, and impact strength measurements. The samples were also characterized by Fourier transform infrared spectroscopy (FTIR). It was found that under the influence of electron irradiation PLA/TMPTA samples underwent degradation while PLA/TAIC samples became crosslinked. Tensile and impact strengths of PLA/TMPTA samples decreased with increasing radiation dose while an enhancement of these properties for PLA/TAIC samples was observed.     

Polypeptide hybrid copolymers as selective micellar nanocarriers in nonaqueous media

The self-assembly of polystyrene-block-poly(l-lysine) (PS-PLLys·HCl) copolymers with different block lengths has been studied in toluene. The obtained spherical micelles exhibit size variations upon addition of acids or bases, as indicated by light and neutron scattering studies. It is shown that pyridine induces a shrinking of the polystyrene chains in the corona region of the micelles, decreasing the aggregate solvent interface. The addition of benzoic acid, on the other hand, leads to a swelling of the copolymer micelles proportional to the molar fraction of polypeptide. This behavior suggests a selective permeability of the PS-PLLys micelles and the possibility to encapsulate organic compounds in toluene depending on their chemical nature.
