We provide an ergodic theorem for certain Banach-space valued functions on structures over , which allow for existence of frequencies of finite patterns. As an application we obtain existence of the integrated density of states for associated discrete finite-range operators in the sense of convergence of the distributions with respect to the supremum norm. These results apply to various examples including periodic operators, percolation models and nearest-neighbour hopping on the set of visible points. Our method gives explicit bounds on the speed of convergence in terms of the speed of convergence of the underlying frequencies. It uses neither von Neumann algebras nor a framework of random operators on a probability space.   相似文献

We report the crystal structure of the DNA adenine-N6 methyltransferase, M.TaqI, complexed with DNA, showing the fluorescent adenine analog, 2-aminopurine, flipped out of the DNA helix and occupying virtually the same position in the active site as the natural target adenine. Time-resolved fluorescence spectroscopy of the crystalline complex faithfully reports this state: base flipping is accompanied by the loss of the very short ( approximately 50 ps) lifetime component associated with fully base-stacked 2-aminopurine in DNA, and 2-aminopurine is subject to considerable quenching by pi-stacking interactions with Tyr108 in the catalytic motif IV (NPPY). This proves 2-aminopurine to be an excellent probe for studying base flipping by M.TaqI and suggests similar quenching in the active sites of DNA and RNA adenine-N6 as well as DNA cytosine-N4 methyltransferases sharing the conserved motif IV. In solution, the same distinctive fluorescence response confirms complete destacking from DNA and is also observed when the proposed key residue for base flipping by M.TaqI, the target base partner thymine, is substituted by an abasic site analog. The corresponding cocrystal structure shows 2-aminopurine in the active site of M.TaqI, demonstrating that the partner thymine is not essential for base flipping. However, in this structure, a shift of the 3' neighbor of the target base into the vacancy left after base flipping is observed, apparently replicating a stabilizing role of the missing partner thymine. Time-resolved fluorescence and acrylamide quenching measurements of M.TaqI complexes in solution provide evidence for an alternative binding site for the extra-helical target base within M.TaqI and suggest that the partner thymine assists in delivering the target base into the active site.  相似文献

Protein-RNA interactions within ribonucleoprotein particles (RNPs) can be investigated by UV-induced crosslinking of proteins to their cognate RNAs and subsequent isolation and mass-spectrometric analysis of crosslinked peptide-RNA oligonucleotides. Because of the low crosslinking yield, a major challenge in protein-RNA UV crosslinking is the detection of the crosslinked species over the excess of non-crosslinked material, especially when complex systems (native RNPs) are investigated. Here, we applied a novel approach that uses on-line nanoLC-ESI-MS/MS to detect and subsequently sequence peptide-RNA oligonucleotide crosslinks from crude mixtures. To detect the crosslinks we made use of features shared by crosslinks and phosphopeptides, that is, the phosphate groups that both carry. A precursor ion scan for m/z 79 (negative-ion mode, -ve) is applied to selectively detect analytes bearing the phosphate-containing species (i.e., residual non-crosslinked RNA and peptide-RNA crosslinks) from crude mixtures and to determine their exact m/z values. On this basis, a multiple reaction monitoring (MRM) experiment monitors the expected decomposition from the different precursor charge states of the putative crosslinks to one of the four possible RNA nucleobases [m/z 112, 113, 136, 152 (positive-ion mode, +ve)]. On detection, a high-quality MS/MS is triggered to establish the structure of the crosslink. In a feasibility study, we detected and subsequently sequenced peptide-RNA crosslinks obtained by UV-irradiation of (1) native U1 snRNPs and (2) [15.5K-61K-U4atac] snRNPs prepared by reconstitution in vitro. MRM-triggered collision-induced dissociation (CID) MS/MS enabled us to obtain sequence information about the crosslinked peptide and RNA moiety.  相似文献

The instability of a system in which three stratified thin liquid films are confined in a channel with parallel walls and the interior film is subject to van der Waals-driven breakup is examined in this work. We derive a model based on lubrication theory and consisting of a pair of nonlinear partial differential equations describing the position of the two liquid interfaces. A linear stability analysis is carried out to show that the effects of varying the boundary film thicknesses can be understood in terms of several known limits, including a supported monolayer, confined bilayer, and supported bilayer. Variation of the boundary film viscosities is shown in many cases to eliminate the supported-bilayer limit. The parameter regimes in which squeezing and bending modes dominate the initial growth are determined, and nonlinear simulations are used to show that the mode always switches to squeezing near rupture. It is also found that a multi-modal dispersion relation may be created by asymmetries in thickness ratio, but not viscosity ratio, even in the absence of asymmetric interfacial tensions. The results of this study are expected to be relevant to multiphase microfluidic systems and the lithographic printing process.  相似文献

Inactivation efficiency of high hydrostatic pressure (HHP) processing of food is strongly affected by food matrix composition. We investigated effects of fat on HHP inactivation of spoilage-associated Lactobacillus (L.) plantarum strains using defined oil-in-water (O/W)-emulsion model systems. Since fat-mediated effects on HHP inactivation could be dependent on interactions between lipid phase and microbial cells, three major factors possibly influencing such interactions were considered, that is, cell surface hydrophobicity, presence and type of surfactants, and oil droplet size. Pressure tolerance varied noticeably among L. plantarum strains and was independent of cell surface hydrophobicity. We showed that HHP inactivation of all strains tended to be more effective in presence of fat. The observation in both, surfactant-stabilized and surfactant-free (O/W)-emulsion, indicates that cell surface hydrophobicity is no intrinsic pressure resistance factor. In contrast to the presence of fat per se, surfactant type and oil droplet size did not affect inactivation efficiency.  相似文献

We study uniquely ergodic dynamical systems over locally compact, sigma-compact Abelian groups. We characterize uniform convergence in Wiener/Wintner type ergodic theorems in terms of continuity of the limit. Our results generalize and unify earlier results of Robinson and Assani respectively. We then turn to diffraction of quasicrystals and show how the Bragg peaks can be calculated via a Wiener/Wintner type result. Combining these results we prove a version of what is sometimes known as the Bombieri/Taylor conjecture. Finally, we discuss various examples including deformed model sets, percolation models, random displacement models, and linearly repetitive systems.  相似文献
