We introduce active, probe-based microrheological techniques for measuring the flow and deformation of complex fluids. These techniques are ideal for mechanical characterization either when little sample is available, or when samples show significant spatial heterogeneity. The efficiency of Zn(2+), Cu(2+), Ni(2+), Co(2+), Fe(2+) or Mn(2+) labeling of the conformational and charge states of lysozyme was studied in H(2)O solvent at pH 2.5-6.8. Labeling of lysozyme was conducted with 50 M, 100 M and 500 M excess of the metal ion, resulting in the number of metal ions attached to lysozyme increasing two-fold over this range. At pH 6.2-6.8, Zn(2+), Cu(2+), Ni(2+), Co(2+) and Mn(2+) labeled the highly folded 7+ conformer and the 8+ and 9+ partially unfolded conformers of lysozyme with the same number of metal ion tags, with only Fe(2+) exhibiting no labeling. Lysozyme conserved its charge after metal ion labeling which shows at each charge state the divalent metal ion is replacing two protons. As the pH is lowered to 4.7-5.0 and 2.5-2.9, the labeling of lysozyme by Zn(2+), Cu(2+), Ni(2+), Co(2+) or Mn(2+) decreased in efficiency due to increased competition from protons for the aspartate and glutamate binding sites. The metal ions preferentially labeled the highly folded 7+ and partially unfolded 8+ conformers, but labeling decreased as the charge of lysozyme increased. In contrast to the other metal ions, Fe(2+) exhibited labeling of lysozyme only at the lowest pH of 2.8. At higher pH, the oxidation of Fe(2+) and formation of hydroxy-bridged complexes probably make the Fe(2+) unreactive towards lysozyme. 相似文献

The technique of speckle visibility spectroscopy has been employed for the measurement of dynamics using coherent X‐ray scattering. It is shown that the X‐ray contrast within a single exposure can be related to the relaxation time of the intermediate scattering function, and this methodology is applied to the diffusion of 72 nm‐radius latex spheres in glycerol. Data were collected with exposure times as short as 2 ms by employing a resonant shutter. The weak scattering present for short exposures necessitated an analysis formalism based on the spatial correlation function of individual photon charge droplets on an area detector, rather than the usual methods employed for intensity correlations. It is demonstrated that this method gives good agreement between theory and experiment and thus holds promise for extending area‐detector‐based coherent scattering methods to the study of faster dynamics than previously obtainable. 相似文献

In this survey we attempt to give a unified presentation of a variety of results on the lifting of valid inequalities, as
well as a standard procedure combining mixed integer rounding with lifting for the development of strong valid inequalities
for knapsack and single node flow sets. Our hope is that the latter can be used in practice to generate cutting planes for
mixed integer programs.
The survey contains essentially two parts. In the first we present lifting in a very general way, emphasizing superadditive
lifting which allows one to lift simultaneously different sets of variables. In the second, our procedure for generating strong
valid inequalities consists of reduction to a knapsack set with a single continuous variable, construction of a mixed integer
rounding inequality, and superadditive lifting. It is applied to several generalizations of the 0–1 single node flow set.

This paper reports on the use of a multi-goal linear programming model to establish the tuition and fee policy at a major state university over a three-year period. The process of establishing tuition and fee charges is characterized by multiple and conflicting goals. Further complicating the process is the fact that multiple parties are involved in or impact on the decision process, including the various academic and administrative units of the university, the state legislature, parents and students. The charges are established based on the institution's anticipated operating costs and fiscal needs, state-level policies and requirements, projected enrolment patterns, charges at peer institutions, and the university's philosophy concerning differential charges between various student categories. The modelling process employed provides university administration with the maximum possible flexibility in stating and prioritizing university goals and objectives regarding the tuition and fee structure. 相似文献

Chelating diiminophosphorane and tripodal iminophosphorane copper and palladium complexes are found to efficiently catalyze the cyclopropanation of activated monosubstituted olefins with ethyl diazoacetate. Cycloolefins, and linear α-olefins are somewhat less reactive. New routes to ladder‐type phenylene materials 1 and 2 are described. The oligomers 1 and 2 , which possess a “3π‐2spiro” architecture, have been synthesized by using extended diketone derivatives 3 and 10 as key intermediates. The physicochemical properties of the new blue‐light emitter 2 were studied in detail and compared with those of the less‐extended 1 . Owing to the recent development of fluorenone derivatives and their corresponding more conjugated analogues as potential electron‐transport materials in organic light‐emitting diodes (OLEDs) and as n‐type materials for photovoltaic applications, we also report herein the thermal, optical and electrochemical behavior of the key intermediates, diketones 3 and 10 .
