This paper reports examples of the relatively infrequent appearance of shake-up satellites in Hel photoelectron spectra, namely those of N2O4 and CH3NO. High-throughput metabolic screening has been requested routinely to keep pace with high-throughput organic synthesis. Liquid chromatography/tandem mass spectrometry (LC/MS/MS) with a fast gradient has become the method of choice for the task due to its sensitivity and selectivity. We have developed an automated system that consists of a robotic system for in vitro incubation and a commercially available software package for automatic MS/MS method development. A short, generic LC gradient and MS conditions that are applicable to most compounds have been developed to minimize the method development time and data analysis. This system has been used to support a number of in vitro screening assays in early drug discovery phase including microsomal stability and protein binding. 相似文献

A simple, efficient and accurate gas-liquid chromatography (GLC) method for the simultaneous determination of eight active ingredients in cough-cold syrups has been developed. The active ingredients under study were bromhexine, chlorpheniramine, codeine, dextromethorphan, diphenhydramine, ephedrine/pseudoephedrine, guaiphenesin and papaverine. Before injection, the active ingredients were first separated, from the excipients present in the cough-cold syrups, with chloroform, from alkaline medium. They were then separated by GLC on a glass column (5 ft x 2 mm i.d.) packed with 3% of OV-25 supported on Supelcoport (80-100 mesh). The column temperature was maintained at 170 degrees C for 1 min, then programmed to 265 degrees C at a rate of 10 degrees C min-1, and maintained at this temperature for 10 and 1 min, respectively, for samples with and without papaverine. The flow-rate of the nitrogen carrier gas was 30 ml min-1. A flame-ionisation detector was used for detection, and clomipramine hydrochloride was used as the internal standard. The recoveries of the drugs ranged from 96.0 to 99.7%, and the relative standard deviations for ten replicate determinations ranged from 0.49 to 4.7%. The effect of showerhead design, number of holes and geometry, in a parallel plate reactor was studied by measuring the concentration of silane reactant by coherent anti-Stokes Raman scattering (CARS) spectroscopy as a function of radio frequency (rf) pulse width and peak power during pulsed power plasma enhanced chemical vapor deposition (PECVD) of silicon nitride thin films. Film deposition rate, stress, SiH/NH ratio, and thickness and index of refraction homogeneity were correlated with the change in silane concentration for each of the three head geometries: radial, square, and asymmetrical. The asymmetrical head caused plasma quality problems which affected the films' qualities. The square pattern showed good mixing qualities, but produced a film with high compressive stress. The radial head provided the most homogenous film, with respect to index of refraction and film thickness. With a 10 ms pulse width, however, the radial head plasma acted as a continuous plasma for depletion and stress data. The showerhead geometry affects plasma qualities, like stability and intensity, and reactant gas velocities, which in turn affect the nitride film thickness, nitride composition, and stress. 相似文献

Photoelectron energy distribution curves from solid nitrogen were measured for excitation energies to 40 cV using synchrotron radiation. Partial cross sections for emission from 3σg, 1πu and 2σu derived valence bands show pronounced mauna 3, 4, 2.9 and 3.0 eV above the vacuum levels, interpreted as due to high density of conduction-band final states.
