G protein-coupled receptors (GPCRs) are core switches connecting excellular survival or death signals with cellular signaling pathways in a context-dependent manner. Opsin 3 (OPN3) belongs to the GPCR superfamily. However, whether OPN3 can control the survival or death of human melanocytes is not known. Here, we try to investigate the inherent function of OPN3 on the survival of melanocytes. Our results demonstrate that OPN3 knockdown by RNAi-OPN3 in human epidermal melanocytes leads to cell apoptosis. The downregulation of OPN3 markedly reduces intracellular calcium levels and decreases phosphorylation of BAD. Attenuated BAD phosphorylation and elevated BAD protein level alter mitochondria membrane permeability, which trigger activation of BAX and inhibition of BCL-2 and raf-1. Activated BAX results in the release of cytochrome c and the loss of mitochondrial membrane potential. Cytochrome c complexes associate with caspase 9, forming a postmitochondrial apoptosome that activate effector caspases including caspase 3 and caspase 7. The release of apoptotic molecules eventually promotes the occurrence of apoptosis. In conclusion, we hereby are the first to prove that OPN3 is a key signal responsible for cell survival through a calcium-dependent G protein-coupled signaling and mitochondrial pathway.  相似文献

Hericium erinaceus, a culinary and medicinal mushroom, is widely consumed in Asian countries. Chemical investigation on the fruiting bodies of Hericium erinaceus led to the isolation of one new ergostane-type sterol fatty acid ester, erinarol K (1); and eleven known compounds: 5α,8α -epidioxyergosta-6,22-dien-3β-yl linoleate (2); ethyl linoleate (3); linoleic acid (4); hericene A (5); hericene D (6); hericene E (7); ergosta-4,6,8(14),22-tetraen-3-one (8); hericenone F (9); ergosterol (10); ergosterol peroxide (11); 3β,5α,6α,22E-ergosta-7,22-diene-3,5,6-triol 6-oleate (12). The chemical structures of the compounds were determined by 1D and 2D NMR (nuclear magnetic resonance) spectroscopy, mass spectra, etc. Anti-inflammatory effects of the isolated aromatic compounds (5–7, 9) were evaluated in terms of inhibition of pro-inflammatory mediator (TNF-α, IL-6 and NO) production in lipopolysaccharide (LPS)-stimulated murine RAW 264.7 macrophage cells. The results showed that compounds 5 and 9 exhibited moderate activity against TNF-α (IC50: 78.50 μM and 62.46 μM), IL-6 (IC50: 56.33 μM and 48.50 μM) and NO (IC50: 87.31 μM and 76.16 μM) secretion. These results supply new information about the secondary metabolites of Hericium erinaceus and their anti-inflammatory effects.  相似文献

A simple, rapid, and low-cost coulometric method for direct detection of glyphosate using hydrophilic interaction chromatography is presented. The principle of detection is based on the enhancement of the anodic current of copper microelectrode in the presence of complexing agents, such as glyphosate, with the formation of a soluble Cu(II) complex. Under optimized conditions, the limit of detection (S/R = 3) for glyphosate was 0.1 mg L−1 (0.59 μM) without any preconcentration method. The calibration curve has been found linear in all concentration range tested (from limit of detection to 34 mg L−1) with an excellent correlation coefficient (0.9999). The present method was successfully applied for the determination of glyphosate in fruit juices without any kind of extraction, clean-up, or preconcentration step, with recoveries of 92 and 90% for apple and grape juice, respectively.  相似文献

In present times, barcode decoders on mobile phones can extract the data content of QR codes. However, this convenience raises concerns about security issues when using QR codes to transmit confidential information, such as e-tickets, coupons, and other private data. Moreover, current secret hiding techniques are unsuitable for QR code applications since QR codes are module-oriented, which is different from the pixel-oriented hiding manner. In this article, we propose an algorithm to conceal confidential information by changing the modules of the QR Code. This new scheme designs the triple module groups based on the concept of the error correction capability. Additionally, this manner can conceal two secret bits by changing only one module, and the amount of hidden confidential information can be twice the original amount. As a result, the ordinary data content (such as URL) can be extracted correctly from the generated QR code by any barcode decoders, which does not affect the readability of scanning. Furthermore, only authorized users with the secret key can further extract the concealed confidential information. This designed scheme can provide secure and reliable applications for the QR system.  相似文献

Nanoparticles have been known as the useful materials in working fluids for petroleum industry. But the stabilization of nano-scaled materials in water-based working fluids at high salinities is still a big challenge. In this study, we successfully prepared the anionic polymer/multi-walled carbon nanotubes (MWNTs) composites by covalently wrapping of MWNTs with poly (sodium 4-styrenesulfonate) (PSS) to improve the stability of MWNTs in high concentration electrolytes. The PSS/MWNTs composites can automatically disperse in salinity up to 15 wt% NaCl and API brines (8 wt% NaCl?+?2 wt% CaCl2). Hydrodynamic diameters of composites were measured as a function of ionic strength and API brines by dynamic light scattering (DLS). By varying the concentration of brines, hydrodynamic diameter of PSS/MWNTs composites in brines fluctuated between 545?±?110 nm for 14 days and 673?±?171 nm for 30 days. Above results showed that PSS/MWNTs could be well stable in high salts solutions for a long period of time. After wrapped with PSS, the diameters of nanotubes changed from 30?~?40 to ~?430 nm, the thickness of wrapped polymer is about ~?400 nm by analysis of morphologies. The zeta potentials of PSS/MWNTs composites in various salinity of brines kept at approximately ??41?~???52 mV. Therefore, the well dispersion of PSS/MWNTs in high salinity is due to large negative charges of poly (sodium 4-styrenesulfonate), which provide enough electrostatic repulsion and steric repulsion to hinder compression of electric double layer caused by high concentration electrolytes.  相似文献

A series of measurements on the high pressure (up to 18 bar) coaxial e-beam pumped Ar/Xe laser has been made at high power loading (up to 25 MW/cm~2) for various Xe mole fraction in Ar. We found that the optimum laser gas composition in our system was 0.4% Xe in Ar which was independent of the total gas pressure. At the optimum gas composition the output energy increased with increasing total gas pressure up to 16 bar, but the intrinsic efficiency reached a maximum at the total gas pressure of 8 bar. It was also found that at the optimum power deposition for both the 2.63μm and 2.65μm transitions the ratio of the integrated optical energy to that of the 1.73μm transition had a minimum in most cases at the optimum gas mixture for various gas pressures. This ratio increased with increasing gas pressure. The experimental results suggested that besides the 2-body Xe quenching at high Xe fraction and electron collision mixing at high power depositions the heavy particle quenching from Ar at high gas press  相似文献
