This study describes the development of a microfluidic device for the high-throughput screening of culture conditions, such as the optimum sodium acetate concentration for promoting rapid growth and high lipid accumulation of Chlamydomonas reinhardtii. An analysis of the microalgal growth on the microfluidic device revealed an optimum sodium acetate concentration of 5.72 g L?1. The lipid content, determined by the 4,4-Difluoro-1,3,5,7-tetramethyl-4-bora-3a,4a-diaza-s-indacene (BODIPY® 505/515) staining method, increased with the sodium acetate concentration. The results were found to be statistically reproducible with respect to cell growth and lipid production. Other nutrient conditions, including the nitrogen and phosphorus concentrations, can also be optimized on the same microfluidic platform. The microfluidic device performance results agreed well with the results obtained from the flask-scale experiments, validating that the culture conditions were scalable. Finally, we, for the first time, established a method for the absolute quantification of the microalgal lipid content in the picoliter culture volumes by comparing the on-chip and off-chip data. In conclusion, we successfully demonstrated the high-throughput screening of sodium acetate concentrations that induced high growth rates and high lipid contents in C. reinhardtii cells on the microfluidic device.

Cellulose diacetate (CDA) plasticized with triacetine was blended by melting extrusion with two different kind of elastomeric core–shell impact modifiers: methyl methacrylate (MMA, shell) grafted onto styrene–butadiene–rubber (SBR, core) (MSBR) and MMA (shell) grafted onto butyl acrylate rubber (BAR, core) (MBAR). The different CDA/MSBR and CDA/MBAR blends were characterized by mechanical properties and morphological observation with various impact modifier contents. The highest impact strength was observed in the case of the blend with 5 wt% of MSBR and 3 wt% of MBAR, respectively. The tensile strength and Young’s modulus of CDA blends were decreased with increasing both MSBR and MBAR. According to SEM observation, MBAR was dispersed more effectively in CDA matrix than that of MSBR, thus indicating improved impact strength.  相似文献

Cotton fabrics with antibacterial properties were prepared by the treatment with 3,3′4,4′-benzophenone tetracarboxylic dianhydride (BPTCD) in a combined process of shaking immersion in dyeing machine and pad-dry-cure. Environmentally-benign choline chloride (ChCl)-based deep eutectic solvents (DESs) were mainly examined as treatment media instead of using organic solvent. The results revealed that cotton fabrics treated with BPTCD in urea-ChCl DES showed a strong ester carbonyl peak in fourier transform infrared (FTIR) analysis, indicating fixation of BPTCD on cotton cellulose. Detailed characterizations of the BPTCD-treated cotton were carried out by FTIR, thermogravimetric analysis, scanning electron microscopy, dye staining, and evaluation of hydrophilicity and strength. The treated fabrics demonstrated a high level of antibacterial characteristics before and after UV irradiation. This indicated that addition of ChCl could enhance antibacterial activity of cotton before UV irradiation. Therefore, use of ChCl-based DES along with BPTCD incorporation provided environmentally-acceptable and economically-feasible treatment process for preparation of novel antibacterial cotton.  相似文献

In this study, the composite polymer electrolytes (CPEs) were prepared by solution casting technique. The CPEs consisted of PEO/PMMA blend as a host matrix doped with LiClO4. Propylene carbonate (PC) was used as plasticizer and a small amount of imidazolium salt-supported amorphous silica (IS-AS) as a filler was prepared by the sol–gel method. At room temperature, the highest conductivity was obtained for the composition having PEO–PMMA–LiClO4–PC–4wt. % IS-AS with a value of 1.15 × 10?4 S/cm. In particular, the CPE using the IS-AS filler showed a higher conductivity than any other sample (fumed silica, amorphous silica). Studies of differential scanning calorimetry and scanning electron microscopy indicated that the ionic conductivity increase was due to an expansion in the amorphous phase which enhances the flexibility of polymeric chains and the homogeneous structure of CPEs. It was found that the ionic conductivity and interfacial resistance stability of CPEs was significantly improved by the addition of IS-AS. In other words, the resistance stability and maximum ambient ionic conductivity of CPEs containing IS-AS filler were better than CPEs containing any other filler.  相似文献

Economic production of titanium dioxide (yield >98 %) from ilmenite has been achieved by use of a modified sulfate reduction process. A series of samples were prepared by varying the concentration of titanium dioxide nuclei (0.2, 0.3, and 0.6 %) and further impregnation with antimony and vanadia. The structural and acidic properties of the samples were comprehensively studied by X-ray diffraction (XRD), transmission electron microscopy, BJH pore size distribution, and temperature-programmed desorption of NH3. The XRD results revealed the presence of intense peaks from anatase titanium dioxide. Enhancement of surface area was observed for second-time filtered samples, possibly because of loss of iron from the bulk. As a result, formation of additional micropores was apparent from N2 adsorption and desorption isotherms. Among all the antimony and vanadia-doped samples, the first-time filtered sample with the low concentration of nuclei (0.2 %) had the highest catalytic activity at low temperatures, owing to its larger pore size and abundant acidic species.  相似文献

Abstract

A new Monte Carlo sampling scheme, namely High Directional Monte Carlo procedure, is used to obtain the global energy minimum conformations of polypeptides such as enkephalin and melittin. The resultant structures of enkephalin and melittin agree well with previous results of theoretical and experimental studies. Particularly, it is shown that some important parts in the conformation, such as the hinge region that principally determines the tertiary structure of proteins, are correctly described by the new method. The resultant structures are compared with those of other works and their stereoscopic views are shown.  相似文献

Abstract

Nitro-substituted poly[2,2′-(m-phenylene)-5,5′-bibenzimidazole]s (PBIs) were synthesized by the reaction of PBI with nitric acid in sulfuric acid under various conditions. The number of nitro groups substituted on the aromatic ring of PBI per polymeric unit varied from 1.44 to 3.55 according to the reaction conditions. An increase in reaction temperature and concentration of the nitric acid increased the degree of substitution. The inherent viscosity of the substituted polymer increased as the reaction temperature decreased. When the reaction temperature was 30°C, the inherent viscosity of the polymer increased as the concentration of nitric acid increased. The nitro-substituted PBI exhibited polyelectrolyte behavior in formic acid. The nitro groups substituted on PBI were dissociated when the polymer was heated to 450°C, displaying exothermic behavior, and the decomposition of polymer was proportional to its nitro group content. All nitro-substituted PBIs showed better solubilities in polar aprotic and acidic solvents, such as dimethylacetamide, dimethylsulfoxide, dimethylformamide, N-methylpyrrolidone, formic acid, sulfuric acid, and trifluoromethanesulfonic acid.  相似文献

In this paper, we demonstrated a highly discriminated and reliable molecular switch based on the interaction between the self-duplex of (Py)A-substituted oligodeoxyadenylate and graphene oxide in aqueous solution. This system showed a clear on/off state through the association and dissociation of (Py)A-modified oligodeoxynucleotide with graphene oxide in manipulated pH conditions, high amplitude efficiency for at least 50 cycles, and rapid response within seconds. Our molecular switch system has high reproducibility and simple operation by using pH stimulus.  相似文献

We have developed an improved method for constructing a full-length cDNA library using small quantity of material by modifying the original oligo-capping method. In our devised method, total RNAs are used in sequential oligo-capping steps directly without preliminary mRNA purification. Using this method, we constructed full- length cDNA libraries from 100 mg of total RNA. These libraries contained 8x10(5) to 8x10(6) independent clones with average insert sizes of 2.0 kb. Moreover, the number of full-length cDNAs containing the translation initiation codon ATG in the constructed libraries was estimated to 60-70%. In addition, 54% of the known cDNAs had a longer 5' end than the corresponding genes in the public database. Our results show that the method can be effectively used to construct full-length enriched cDNA libraries, especially, if starting material is limited.  相似文献
