The bengamides, sponge‐derived natural products that have been characterized as inhibitors of methionine aminopeptidases (MetAPs), have been intensively investigated as anticancer compounds. We embarked on a multidisciplinary project to supply bengamides by fermentation of the terrestrial myxobacterium M. virescens, decipher their biosynthesis, and optimize their properties as drug leads. The characterization of the biosynthetic pathway revealed that bacterial resistance to bengamides is conferred by Leu 154 of the myxobacterial MetAP protein, and enabled transfer of the entire gene cluster into the more suitable production host M. xanthus DK1622. A combination of semisynthesis of microbially derived bengamides and total synthesis resulted in an optimized derivative that combined high cellular potency in the nanomolar range with high metabolic stability, which translated to an improved half‐life in mice and antitumor efficacy in a melanoma mouse model.  相似文献

So far, the maximum size of a partial \((k-1)\)-spread in \({\text {PG}}(n-1,q)\) was known for the cases \(n\equiv 0\pmod k\), \(n\equiv 1\pmod k\), and \(n\equiv 2\pmod k\) with the additional requirements \(q=2\) and \(k=3\). The photorefractive effect of undoped LiNbO3 crystals of high purity is studied by means of two-photon excitation of picosecond light pulses. We show that two-photon photorefractive recording is accompanied by characteristic changes of the optical absorption and electron spin resonance spectra due to the formation of color centers. The diffraction of thermal neutrons by a plane Ni covered ruled grating was investigated at various angles of incidence. A 54 lines per mm grating was used in the reflection case. Science and technology are intimately related, and advances in science often become possible with the availability of new instrumentation. This has certainly been the case in mass spectrometry, which is used in so many scientific disciplines. Originally developed as an instrument for research in physics it was used in the discovery of isotopes, their recognition as the fundamental species comprising the elements, and the investigation of elemental isotopic composition. Isotope ratio mass spectrometry is a metrological technique of the highest order, and has been widely used in chemical, biochemical, cosmochemical, environmental, geological, physical, and nuclear research. Mass spectrometry presently plays a key role not only in scientific research, but also in industrial operations. This paper highlights the role that Alfred Otto Carl Nier played in bringing mass spectrometry into the mainstream of science. Nier's career spanned a remarkable period in science, and he made crucial contributions to atomic weights, geochronology, isotope geochemistry, nuclear physics, and space science. He is widely viewed as the 'father of modern mass spectrometry', because of his genius with instrumentation, his innovations, and the generosity with which he shared his ideas and designs. It is timely to remember his fundamental work in mass spectrometry, particularly the development of the sector field mass spectrometer, which is still the instrument of choice for many isotope scientists some 66 years after its first appearance in 1940.  相似文献

We consider a setting where one has to organize one or several group activities for a set of agents. Each agent will participate in at most one activity, and her preferences over activities depend on the number of participants in the activity. The goal is to assign agents to activities based on their preferences in a way that is socially optimal and/or stable. We put forward a general model for this setting, which is a natural generalization of anonymous hedonic games. We then focus on a special case of our model where agents’ preferences are binary, i.e., each agent classifies all pairs of the form ‘(activity, group size)’ into ones that are acceptable and ones that are not. We formulate several solution concepts for this scenario, and study them from the computational point of view, providing hardness results for the general case as well as efficient algorithms for settings where agents’ preferences satisfy certain natural constraints.  相似文献
