Here, we describe a synthetic approach for generating artificial proteins by the assemblage of naturally occurring peptide motifs. Two motifs respectively related to apoptosis induction and protein transduction were encrypted into different reading frames of an artificial gene (microgene), which was then polymerized; random frame shifts at the junctions between the microgene units yielded combinatorial polymers of three reading frames. Among the proteins created, #284 was found to penetrate through cell membranes and exert a strong apoptotic effect on several cancer cell lines. Because a simple linkage of these motifs was not sufficient to construct a bifunctional peptide, and the successful reconstitution was dependent on how they were joined together, the combinatorial strategy is important for reconstituting functions from mixtures of motifs. This microgene-based approach represents a novel system for creating proteins with desired functions.  相似文献

Ground state properties of a Kondo superconductor of low impurity concentration are calculated exactly within the Nagaoka approximation. The results for the location of the bound state motivate us to formulate the notion of intrinsic gaplessness. The order parameter, the ground state energy, and the thermodynamic critical field are computed. The thermal decomposition of ethane was studied behind reflected shock waves over the temperature range 1200–1700 K and over the pressure range 1.7?2.5 atm, by both tracing the time variation of absorption at 3.39 μm and analyzing the concentration of the reacted gas mixtures. The mechanism to interpret well not only the earlier stage of C2H6 decomposition, but also the later stage was determined. The rate constant of reactions, C2H6 → CH3 + CH3, C2H6 + C2H3 → C2H5 + C2H4, C2H5 → C2H4 + H were calculated. The rate constants of the other reactions were also discussed.  相似文献

The purpose of this study is to develop peptide-based platelet-derived growth factor receptor β (PDGFRβ) imaging probes and examine the effects of several linkers, namely un-natural amino acids (D-alanine and β-alanine) and ethylene-glycol (EG), on the properties of Ga-DOTA-(linker)-IPLPPPRRPFFK peptides. Seven radiotracers, 67Ga-DOTA-(linker)-IPLPPPRRPFFK peptides, were designed, synthesized, and evaluated. The stability and cell uptake in PDGFRβ positive peptide cells were evaluated in vitro. The biodistribution of [67Ga]Ga-DOTA-EG2-IPLPPPRRPFFK ([67Ga]27) and [67Ga]Ga-DOTA-EG4-IPLPPPRRPFFK ([67Ga]28), which were selected based on in vitro stability in murine plasma and cell uptake rates, were determined in BxPC3-luc-bearing nu/nu mice. Seven 67Ga-labeled peptides were successfully synthesized with high radiochemical yields (>85%) and purities (>99%). All evaluated radiotracers were stable in PBS (pH 7.4) at 37 °C. However, only [67Ga]27 and [67Ga]28 remained more than 75% after incubation in murine plasma at 37 °C for 1 h. [67Ga]27 exhibited the highest BxPC3-luc cell uptake among the prepared radiolabeled peptides. As regards the results of the biodistribution experiments, the tumor-to-blood ratios of [67Ga]27 and [67Ga]28 at 1 h post-injection were 2.61 ± 0.75 and 2.05 ± 0.77, respectively. Co-injection of [67Ga]27 and an excess amount of IPLPPPRRPFFK peptide as a blocking agent can significantly decrease this ratio. However, tumor accumulation was not considered sufficient. Therefore, further probe modification is required to assess tumor accumulation for in vivo imaging.  相似文献

We compute the bulk entanglement entropy of a massive scalar field in a Poincare AdS with the Dirichlet and Neumann boundary condition when we trace out a half space. Moreover, by taking into account the quantum back reaction to the minimal surface area, we calculate how much the entanglement entropy changes under a double‐trace deformation of a holographic CFT. In the AdS3/CFT2 setup, our result agrees with the known result in 2d CFTs. In higher dimensions, our results offer holographic predictions.  相似文献

2H‐Pyrone derivatives were synthesized through the reaction of aryl acetyl compounds with ketene dithioacetals in the presence of sodium hydroxide, and they showed very strong fluorescence in the solid state. The light‐emitting region of these 2H‐pyrones is 447‐630 nm in the solid states.  相似文献

在溶剂热条件下,合成了2个碱土金属配位聚合物[Ca(tdc)(DMF)2]n1)和[Ba(tdc)]n2)(H2tdc=2,5-噻吩二甲酸),分别用元素分析、红外光谱、X射线单晶衍射、粉末衍射、热重分析和荧光光谱对它们进行了表征。结构分析表明,配合物1具有4,4连接的二维层状结构,拓扑符号为(44·62),而配合物2呈现三维网络结构。固体荧光测试表明配合物1比配合物2具有更显著的荧光性能。  相似文献
