The Rhodobacter sphaeroides protein AppA has the unique quality of sensing and transmitting light and redox signals. By acting as antirepressor to the PpsR protein, it acts as a major regulator in photosynthesis gene expression. In this study, we show that by introducing amino acid exchanges into the AppA protein, the in vivo activity as an antirepressor can be greatly altered. The tryptophan 104 to phenylalanine (W104F) base exchange greatly diminished blue-light sensitivity of the BLUF domain. From the obtained in vivo data, the difference in thermal recovery rate of the signaling state of the BLUF domain between the wild type and mutated protein was calculated, predicting an about 10-fold faster recovery in the mutant, which is consistent with in vitro data. Introduction of a tyrosine 21 to phenylalanine (Y21F) or to cysteine (Y21C) mutation led to a complete loss of AppA antirepressor activity, while additionally leading to an increase of photosynthesis gene expression after illumination with high blue-light quantities. Interestingly, this effect is not visible in a W104F/Y21F double mutant that again shows a wild-type–like behavior of the BLUF domain after blue-light illumination, thus restoring the activity of AppA.  相似文献

Proton transfer from the photoacid 8‐hydroxy‐1,3,6‐pyrenetrisulfonic acid (HPTS) to water is studied in reverse micelles with ionic (AOT=sodium dioctyl sulfosuccinate) and non‐ionic (BRIJ‐30=polyoxyethylene(4)lauryl ether) surfactants. The dynamics are studied by probing the transient electronic absorption and transient vibrational absorption, both with sub‐picosecond resolution. The reverse micelle sizes range from approximately 1.6 to 5.5 nm in diameter. For both surfactants it is found that the rate of proton transfer decreases with decreasing reverse micelle size, regardless of surfactant. In addition, for AOT reverse micelles, a fraction of the photoacid molecules exhibit non‐radiative decay, preventing proton transfer.  相似文献

Knowledge of the presence of micropollutants such as pharmaceuticals, in coastal areas, is very limited; therefore, the main objective of this study was to optimize and validate a new analytical method for the quantitative analysis of 13 multiclass pharmaceuticals in seawater. Target compounds included antibiotics, non-steroidal anti-inflammatory drugs, β-blockers, lipid regulators and one psychiatric drug. A combination of solid-phase extraction and liquid chromatography coupled with multiple mass spectrometry enabled their detection at the low nanogram per litre level. The limits of quantification varied between 1 and 50 ng L-1, for most components the linearities were more than 0.99 and the recoveries obtained in seawater (95–108%) were satisfactory. This method was applied to seawater and estuarine water samples collected in the Belgian coastal zone, to assess the prevalence of common pharmaceuticals in this marine environment. Seven pharmaceuticals, including compounds of which the presence in marine environments had not been reported earlier, were detected, with salicylic acid and carbamazepine being the most abundant, in concentrations up to 855 ng L-1.  相似文献

A new concept for noncovalent immobilization of a ruthenium olefin metathesis catalyst is presented. The 2-isopropoxybenzylidene ligand of a Hoveyda-Grubbs carbene is further modified by an additional amino group (7) and immobilization is achieved by treatment with sulfonated polystyrene forming the corresponding ammonium salt. In this novel strategy for the immobilization of ruthenium-based metathesis catalysts, the amino group plays a two-fold role, being first an active anchor for immobilization and second, after protonation, activating the catalysts (electron donating to electron withdrawing activity switch). The polymeric support was prepared by precipitation polymerization which led to small bead sizes (0.2-2 microm) and large surface areas. Compared to commercial resins this tailor-made phase showed superior properties in immobilization of complex 7. This concept of immobilization was applied to glass-polymer composite megaporous Raschig rings. Ru catalyst 7 on Raschig rings was used under batch conditions in various metathesis reactions, including ring-closing (RCM), cross- (CM) and enyne metathesis, to give products of high chemical purity with very low ruthenium contamination levels (21-102 ppm). The same ring can be used for up to 6 cycles of metathesis.  相似文献

We analyze the photon-induced birefringence in semiconductors based on pump-probe setups, within the semiconductor Bloch equations formalism and the Luttinger-Kohn model for the band structure. When the pump and probe pulses are well separated in time, the anisotropic momentum space filling of the photo-excited electrons is the only mechanism causing the induced birefringence. The birefringence ratio is then for pump and probe having perpendicular vs. parallel linear polarizations. This ratio is for opposite vs. identical circular polarization. When the pump and probe pulses overlap in time, these birefringence ratios become for linear polarizations and in case of circular polarizations. These predictions differ markedly from those for optical fibers.  相似文献

This note is a somewhat personal account of a paper that L.E.J. Brouwer published in 1908 and that dealt with the possible cardinalities of subsets of the continuum. That paper is of interest because it represents the first time that Brouwer presented his ideas on foundations in an international forum.I found Brouwer’s notions and arguments at times hard to grasp if not occasionally perplexing. I hope that this note contributes to a further discussion of the definitions and reasonings as presented in Brouwer’s paper.  相似文献

To study carbohydrate digestion and glucose absorption, time-dependent (13)C enrichment in plasma glucose is measured after oral administration of naturally occurring (13)C-enriched carbohydrates. The isotope enrichment of the administered carbohydrate is low (APE <0.1%) and plasma (13)C glucose measurements are routinely determined with gas chromatography/combustion/isotope ratio mass spectrometry (GC/C/IRMS) or liquid chromatography/combustion/isotope ratio mass spectrometry (LC/C/IRMS). In this study, plasma glucose was converted into CO(2) by an in-tube reaction with yeast permitting direct measurement of (13)CO(2) in the headspace. Saccharomyces cerevisiae incubated under anaerobic conditions was able to convert sufficient glucose into CO(2) to produce a consistent CO(2) peak in IRMS with little variation in peak area and precise delta(13)C(PDB) values for corn glucose: -11.40 +/- 0.16 per thousand, potato glucose: -25.17 +/- 0.13 per thousand, and plasma glucose: -26.29 +/- 0.05 per thousand. The measurement showed high linearity (R(2) = 0.999) and selectivity and was not affected by the glucose concentration in the tested range of 5-15 mM. Comparison with GC/C/IRMS showed a good correlation of enrichment data: R(2) > 0.98 for both sources of glucose and plasma samples. Commercially available, instant dried baker's yeast was qualitatively and quantitatively comparable with freshly prepared yeast: R(2) > 0.96, slope 1.03 and 1.08 for glucose solutions and plasma, respectively. Thus, yeast conversion of plasma glucose into CO(2) and (13)C measurement applying a breath (13)CO(2) analyzer is an inexpensive, simple and equally accurate alternative to the more expensive and laborious GC/C/IRMS and LC/C/IRMS measurements.  相似文献

A key challenge for microbiology is to understand how evolution has shaped the wiring of regulatory networks. This is amplified by the paucity of information of power‐spectra of physicochemical stimuli to which microorganisms are exposed. Future studies of genome evolution, driven by altered stimulus regimes, will therefore require a versatile signal transduction system that allows accurate signal dosing. Here, we review the general stress response of Bacillus subtilis, and its upstream signal transduction network, as a candidate system. It can be activated by red and blue light, and by many additional stimuli. Signal integration therefore is an intricate function of this system. The blue‐light response is elicited via the photoreceptor YtvA, which forms an integral part of stressosomes, to activate expression of the stress regulon of B. subtilis. Signal transfer through this network can be assayed with reporter enzymes, while intermediate steps can be studied with live‐cell imaging of fluorescently tagged proteins. Different parts of this system have been studied in vitro, such that its computational modeling has made significant progress. One can directly relate the microscopic characteristics of YtvA with activation of the general stress regulon, making this system a very well‐suited system for network evolution studies.  相似文献
