The use of silica shells offers many advantages in surface-enhanced Raman scattering (SERS)-based biological sensing applications due to their optical transparency, remarkable stability in environmental media, and improved biocompatibility. Here, we report a novel layer-by-layer method for the preparation of silica-hollow gold nanosphere (HGN) SERS tags. Poly(acrylic acid) was used to stabilize Raman reporter-tagged HGNs prior to the adsorption of a coupling agent, after which a silica shell was deposited onto the particle surface using Sto?ber's method. Importantly, competitive adsorption of the Raman reporter molecules and coupling agents, which results in unbalanced loading of reporter molecules on individual nanoparticles, was avoided using this method. As a result, the loading density of reporter molecules could be maximized. In addition, HGNs exhibited strong enhancement effects from the individual particles because of their ability to localize the surface electromagnetic fields through pinholes in the hollow particle structures. The proposed layer-by-layer silica-encapsulated HGN tags showed strong SERS signals as well as excellent multiplexing capabilities.  相似文献

A single walled carbon nanotube (SWCNT)-based three-electrode system was fully integrated on glass substrates using a standard microfabrication process and electrochemically characterized using cyclic voltammetry. O(2) plasma functionalization of the SWCNT film working electrode for achieving high sensitivity was voltammetrically optimized with respect to the plasma power and treatment time. Chlorination of a Ag thin-film was done in an acidic solution for different dip times to form a thin-film Ag/AgCl reference electrode. The Nernstian behavior of as-prepared and seven-day-aged Ag/AgCl thin-film electrodes was investigated for seeking the optimum reference electrode with long-term stability and was compared to a commercial reference electrode. A quality control evaluation and a performance assessment of the fully integrated SWCNT-transferred sensing systems were performed using cyclic voltammetry. The proposed SWCNT-based three electrode device exhibited clear electrochemistry under voltammetric conditions, and is therefore a candidate for use in all electrochemical biosensors.  相似文献

In an oxygen-depleted environment, endothelial cells initiate an adaptive pattern of synthesis, which may enable them to survive hypoxic crises. Using high-resolution two-dimensional gel electrophoresis in conjunction with mass spectroscopy, we obtained a 24 differential display of proteins in the pancreatic endothelial cell line, MS-1, at four time points following induction of hypoxia. The induction of Wee1 under hypoxia was confirmed both at the mRNA and protein levels. The phosphorylation of cell division cycle 2, which is downstream of Wee1, was also increased after hypoxic exposure. In addition, pre-exposure to hypoxia attenuated a decrease in hydrogen peroxide-induced cell number. The induction of bax (a pro-apoptotic protein) and reduction of bcl (an anti-apoptotic protein) after hypoxia stimulus were also attenuated by hypoxic pre-exposure. Moreover, hydrogen peroxide-induced morphologic damage did not appear in the wild-type Wee1-expressing cells. Taken together, our results suggest that Wee1 may have important role in hypoxia- induced pathophysiological situations in endothelial cells.  相似文献

Osteoarthritis (OA) is an age-related joint disease that is characterized by degeneration of articular cartilage and chronic pain. Oxidative stress is considered one of the pathophysiological factors in the progression of OA. We investigated the effects of grape seed proanthocyanidin extract (GSPE), which is an antioxidant, on monosodium iodoacetate (MIA)-induced arthritis of the knee joint of rat, which is an animal model of human OA. GSPE (100 mg/kg or 300 mg/kg) or saline was given orally three times per week for 4 weeks after the MIA injection. Pain was measured using the paw withdrawal latency (PWL), the paw withdrawal threshold (PWT) and the hind limb weight bearing ability. Joint damage was assessed using histological and microscopic analysis and microcomputerized tomography. Matrix metalloproteinase-13 (MMP13) and nitrotyrosine were detected using immunohistochemistry. Administration of GSPE to the MIA-treated rats significantly increased the PWL and PWT and this resulted in recovery of hind paw weight distribution (P < 0.05). GSPE reduced the loss of chondrocytes and proteoglycan, the production of MMP13, nitrotyrosine and IL-1β and the formation of osteophytes, and it reduced the number of subchondral bone fractures in the MIA-treated rats. These results indicate that GSPE is antinociceptive and it is protective against joint damage in the MIA-treated rat model of OA. GSPE could open up novel avenues for the treatment of OA.  相似文献

B cells play an important role in the pathogenesis of rheumatoid arthritis (RA). High levels of B cell activating factor (BAFF) are detected in autoimmune diseases. BAFF and BAFF receptor (BAFF-R) are expressed in B and T cells of RA synovium. The study was undertaken to identify the NF-ΚB signal pathway involved in the induction of BAFF-R in human B cells. Immunohistochemical staining of NF-ΚB p65, NF-ΚB p50, BAFF, and BAFF-R was performed on sections of synovium from severe and mild RA and osteoarthritis (OA) patients. Peripheral blood mononuclear cells (PBMCs) were isolated from control and RA patients and B cells were isolated from controls. BAFF-R was analyzed by flow cytometry, realtime PCR and confocal staining after treatment with NF-ΚB inhibitors. NF-ΚB p65, NF-ΚB p50, BAFF, and BAFF-R were highly expressed in severe RA synovium relative to mild RA synovium or OA synovium. BAFF-R expression was reduced by NF-ΚB inhibitors in PBMCs and B cells from normal controls. We also showed reduction in expression of BAFF-R via inhibition of the NF-ΚB pathway in PBMCs of RA patients. BAFF/BAFF-R signaling is an important mechanism of pathogenesis in RA and that BAFF-R reduction by NF-ΚB blocking therapy is another choice for controlling B cells in autoimmune diseases such as RA.  相似文献

A novel method to prepare cadmium sulphide nanoparticles (CdS NPs) possessing nearly uniform size was adopted using eggshell membrane (ESM), under different pH conditions. Significant yield of CdS NPs with smallest possible size was obtained by increasing the pH of the reaction medium from acidic to alkaline. The above prepared CdS NPs have been characterized by UV-vis absorption as well as emission spectra, powder X-ray diffraction, scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The efficiency of the above prepared CdS NPs as a catalyst for the photodegradation of toluidine blue (TB) dye, as a function of pH as well as the ratio between the catalyst and the substrate was studied after irradiation with UV light. The results showed that an efficient interaction took place between the catalyst and the substrate to cause degradation of the selected dye. A maximum degradation of toluidine blue dye (90%) was observed at pH 8 which is higher than that of the efficiencies at pH 4 and pH 6.  相似文献

The direct intensity modulation of a three-guide coupled rectangular ring laser having bidirectional lasing characteristics is reported for the first time. The rectangular laser cavity consists of four low loss total internal reflection mirrors and an output coupler made out of three passive coupled waveguides. The laser is fabricated using a total cavity length of 580 μm with active section lengths of 250 μm. For both the clockwise and counterclockwise circulating directions, a lasing threshold of around 38 mA is obtained at room temperature under continuous wave operation. A maximum 3-dB modulation bandwidth of approximately 3.2 GHz is observed in both circulating directions.  相似文献

We investigated, via the classical MD simulation method based on Tersoff-Brenner potential, the fundamental resonance frequency changes of single-walled carbon nanotube (SWCNT) resonators originated from the purely mechanical coupling of the axial-strain-induced torsion (ASIT) response. The fundamental frequency changes were also negligible where the ASIT responses were negligible in achiral SWCNTs whereas those were explicitly found under both compression and tension for the chiral SWCNTs with the obvious ASIT responses. Specially, for SWCNT with the chiral angle of π/12, where the highest ASIT response can be found, the fundamental resonance frequency changes were highest. The fundamental resonance frequencies under the tensioning increased almost linearly with increasing the axial strain whereas they rapidly decreased under compression with increasing the compressive strain.  相似文献

By studying its subgraphs, it is argued that the lower critical percolation threshold of the enhanced binary tree (EBT) is bounded as pc1<0.355059, while the upper threshold is bounded both from above and below by 1/2 according to renormalization-group arguments. We also review a correlation analysis in an earlier work, which claimed a significantly higher estimate of pc2 than 1/2, to show that this analysis in fact gives a consistent result with this bound. Our result confirms that the duality relation between critical thresholds does not hold exactly for the EBT and its dual, possibly due to the lack of transitivity.  相似文献
