Luminescent 50-nm silicon dioxide nanoparticles containing both types of rhodamine 6G (R; particles denoted R-SiO2) were synthesized by the sol–gel method. In the presence of Pb(Ac)2 as a heavy atom perturber the particle can emit the intense and stable room-temperature phosphorescence (RTP) signal of R on a polyamide membrane, with exmax/emmax=470/635 nm for R. Our research indicates that the specific immune reaction between goat-anti-human IgG antibody labeled with R-SiO2 and human IgG can be carried out quantitatively on a polyamide membrane, and the phosphorescence intensity was enhanced after the immunoreaction. Thus a new method for solid-substrate room-temperature phosphorescence immunoassay (SS-RTP-IA) for determination of human IgG was established on the basis of antibody labeled with the nanoparticles containing binary luminescent molecules. The linear range of this method is 0.0624–20.0 pg spot–1 of human IgG (corresponding to a concentration range of 0.156–50.0 ng mL–1, sample volume 0.40 L spot–1). The regression equations of the working curves are Ip=71.27+7.208mIgG (pg spot–1) (r=0.9996). Detection limits calculated as 3Sb/k are 0.022 pg spot–1. Compared with the same IA using fluorescein isothiocyanate (FITC) as the marker the new method was more sensitive and had a wider linear range. After elevenfold replicate measurement RSD are 4.5 and 3.6% for samples containing 0.156 and 50.0 ng mL–1 IgG, respectively. This method is sensitive, accurate, and of high precision. 相似文献

Determination of organophosphate and carbamate pesticides based on enzyme inhibition using a pH-sensitive fluorescence probe 总被引:1,自引:0,他引:1
A flow injection system for the determination of organophosphate and carbamate pesticides is described. A sensitive fluorescence probe was synthesized and used as the pH indicator to detect the inhibition of the enzyme acetylcholinesterase (AChE). The percentage inhibition of enzyme activity is correlated to the pesticide concentration. Several parameters influencing the performance of the system are discussed. The detection limits of 3.5, 50, 12 and 25 μg/l for carbofuran, carbaryl, paraoxon and dichlorvos, in pure water, respectively were achieved with an incubation time of 10 min. A complete cycle of analysis, including incubation time, took 14 min. The detection system has been applied to the determination of carbofuran in spiked vegetable juices (Chinese cabbage and cole), achieving recovery values between 93.2 and 107% for Chinese cabbage juice and 108 and 118% for cole juice at the different concentration levels assayed. 相似文献

以聚合-β-环糊精作为毛细管区带电泳手性选择剂,成功分离了2-苯氧丙酸(PPA)、顺式-功夫菊酸(cis-PA)、1-苯基-2-(对甲苯基)乙胺(PTE)和1-苯基-2-(对甲氧基苯基)乙胺(PME)4种光学活性农药中间体的对映体。考察了不同手性选择剂及其浓度、背景电解质的pH等操作条件对分离的影响。结果表明,在12kV操作电压下、30mmol/Ltris-HCl背景电解质中,用14g/L聚合-β-CD作为手性选择剂,PPA和cis-PA对映体在pH6.0时,PIE和PME对映体分别在pH2.5和pH3.0时可以被较好分离。在优化的分离条件下,用聚合-β-CD作毛细管区带电泳手性选择剂分离PIE对映体的分离度为1.513,成功测定了两种旋光性PIE的对映体过量值。 相似文献

The interactions between Trichoderma reesei cellulase and an anionic surfactant, sodium dodecyl sulfate (SDS), at critical micelle concentration level have been investigated using isothermal titration calorimetry, fluorescence spectroscopy, and circular dichroism. SDS micelles have dual interactions with cellulase: electrostatic at first and then hydrophobic interactions. When the concentration of SDS is smaller than 45.0 mM, SDS micelles cause a partial loss in the hydrolytic activity together with a steep decrease in the -helical content of cellulase. With further increasing the concentration of SDS, however, a re-formation of the -helical structure and a partial recovery of the hydrolytic activity of cellulase induced by SDS micelles are observed. Taken together, these results indicate that SDS micelles exert dual effects on cellulase through binding as both a denaturant and a recovery reagent. 相似文献

Molar heat capacities
of acetaminophen were precisely measured with a small sample precision automated
adiabatic calorimeter over the temperature range from 80 to 330 K. A solid-solid
transition at 149.96 K was found from the Cp,m-T curve. The polynomial functions of Cp,.m(J
K-1 mol-1) vs. T were established
on the heat capacity measurements by means of the least square fitting method.

thermal decomposition kinetics parameters, such as activation energy E, pre-exponential factor A
and reaction order n, were calculated by
TG-DTG techniques with the Freeman-Carroll method, Kissinger method
and Ozawa method. Accordingly the thermal decomposition kinetics equation

The process of fusion has been investigated through
