The synthesis and the properties of 1,4-dibutoxy-2,3-dicyanotriptycene, of a metal-free tetradibenzobarrelenooctabutoxyphthalocyanine, and of the corresponding zinc phthalocyanine are described. The two phthalocyanines do not aggregate when dissolved in benzene at concentrations up to 450 microM. For the metal-free and the zinc compounds, the red band maxima are at 736 and 757 nm, the triplet maxima are at 590 and 605 nm, the triplet state lifetimes are 58 and 177 microseconds, and the protoporphyrin-IX dimethyl ester-to-compound bimolecular rate constants for triplet energy transfer are 2.61 x 10(8) and 1.47 x 10(8) M-1 s-1. Triplet energy transfer from the metal-free compound to O2 is endoergonic by 1.0 kcal mol-1. The potential of the zinc compound for photodynamic therapy is touched upon.  相似文献

Reversed-phase liquid chromatography of recombinant platelet-derived growth factor (PDGF) results in the appearance of at least four distinguishable peaks. The relative areas of these peaks are, in part, dependent upon the gradient time and the temperature. Isolation and reinjection of each peak gave chromatographic profiles comparable to that obtained from unfractionated PDGF. Increasing the temperature above 60 degrees C resulted in a single peak that, when isolated and reinjected at ambient temperature, produced a chromatogram comparable to PDGF which had not been exposed to elevated temperature. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed that all four peaks had the same molecular mass as PDGF and were active as determined by a PDGF mitogenic bioassay. These results indicate that multiple conformations of PDGF are present and we postulate that their appearance may be a result of isomeric structures arising from the presence of Pro-Pro bonds within the primary structure of the protein.  相似文献

Photodynamic therapy (PDT) is a cancer treatment modality utilizing a photosensitizer, light and oxygen. Photodynamic therapy with Photofrin has been approved by the U.S. Food and Drug Administration for treatment of advanced esophageal and early lung cancer. Because of certain drawbacks associated with the use of Photofrin, there is a need to identify new photosensitizers for human use. The photosensitizer Pc 4 (HOSiPc-OSi[CH3]2[CH2]3N[CH3]2) has yielded promising PDT effects in many in vitro and in vivo systems. The aim of this study was to assess the usefulness of Pc 4 as a PDT photosensitizer for a human tumor grown as a xenograft in athymic nude mice. The ovarian epithelial carcinoma (OVCAR-3) was heterotransplanted subcutaneously in athymic nude mice. Sixty mice bearing OVCAR-3 tumors (approximately 80-130 mm3) were divided into six groups of 10 animals each, three for controls and three for treatment. The Pc 4 was given by tail vein injection, and 48 h later a 1 cm area encompassing the tumor was irradiated with light from a diode laser coupled to a fiberoptic terminating in a microlens (lambda = 672 nm, 150 J/cm2, 150 mW/cm2). Tumors of control animals receiving no treatment, light alone or Pc 4 alone continued to grow. Of animals receiving 0.4 mg/kg Pc 4 and light, one (10%) had a complete response and was cured (no regrowth up to 90 days post-PDT), while all others (90%) had a partial response and were delayed in regrowth. Of animals receiving 0.6 mg/kg Pc 4 and light, eight (80%) had a complete response, and two of these were cured. Of animals receiving 1.0 mg/kg Pc 4 and light, six (60%) had a complete response, and two of these were cured. In additional experiments, tumors from animals treated with Pc 4 (1 mg/kg) and light were removed 15, 30, 60 and 180 min post-PDT, and from these tumors DNA and protein were extracted. Agarose gel electrophoresis revealed the presence of apoptotic DNA fragmentation as early as 15 min post-PDT. Western blotting showed the cleavage of the 116 kDa native poly(ADP-ribose) polymerase (PARP) into fragments of approximately 90 kDa, another indication of apoptosis, and the presence of p21/WAF1/CIP1 (p21) in all PDT-treated tumors. These changes did not occur in control tumors. Pc 4 appears to be an effective photosensitizer for PDT of human tumors grown as xenografts in nude mice. Early apoptosis, as revealed by PARP cleavage, DNA fragmentation and p21 overexpression, may be responsible for the excellent Pc 4-PDT response. Clinical trials of Pc 4-PDT are warranted.  相似文献

All-atom molecular dynamics simulations were used to predict water-cyclohexane distribution coefficients \(D_{cw}\) of a range of small molecules as part of the SAMPL5 blind prediction challenge. Molecules were parameterized with the transferable all-atom OPLS-AA force field, which required the derivation of new parameters for sulfamides and heterocycles and validation of cyclohexane parameters as a solvent. The distribution coefficient was calculated from the solvation free energies of the compound in water and cyclohexane. Absolute solvation free energies were computed by an established protocol using windowed alchemical free energy perturbation with thermodynamic integration. This protocol resulted in an overall root mean square error in \(\log D_{cw}\) of almost 4 log units and an overall signed error of ?3 compared to experimental data. There was no substantial overall difference in accuracy between simulating in NVT and NPT ensembles. The signed error suggests a systematic error but the experimental \(D_{cw}\) data on their own are insufficient to uncover the source of this error. Preliminary work suggests that the major source of error lies in the hydration free energy calculations.  相似文献

The nanocrystalline cubic Phase of zirconia was found to be thermally stabilized by the addition of 2.56 to 17.65 mol % Y2O3 (5.0 to 30.0 mol % Y, 95.0 to 70.0 mol % Zr cation content). The cubic phase of yttria stabilized zirconia was prepared by thermal decomposition of the hydroxides at 400°C for 1 hr. 2.56 mol % Y2O3‐ZrO2 was stable up to 800°C in an argon atmosphere. The samples with 4.17 to 17.65 mol % Y2O3 were stable to 1200°C and higher. All samples at temperatures between 1450°C to 1700°C were cubic except the sample with 2.56 mol % Y2O3 which was tetragonal. The crystallite sizes observed for the cubic phase ranged from 50 to 150 Å at temperatures below 900°C and varied from 600 to 800 nm between 1450°C and 1700°C. Control of furnace atmosphere is the main factor for obtaining the cubic phase of Y‐SZ at higher temperature. Nanocrystalline cubic Fe‐SZ (Iron Stabilized Zirconia) with crystallite sizes from 70 to 137 Å was also prepared at 400°C. It transformed isothermally at temperatures above 800°C to the tetragonal Fe‐SZ and ultimately to the monoclinic phase at 900°C. The addition of up to 30 mol % Fe(III) thermally stabilized the cubic phase above 800°C in argon. Higher mol % resulted in a separation of Fe2O3. The nanocrystalline cubic Fe‐SZ containing a minimum 20 mol % Fe (III) was found to have the greatest thermal stability. The particle size was a primary factor in determining cubic or tetragonal formation. The oxidation state of Fe in zirconia remained Fe3+. Fe‐SZ lattice parameters and rate of particle growth were observed to decrease with higher iron content. The thermal stability of Fe‐SZ is comparable with that of Ca‐SZ, Mg‐SZ and Mn‐SZ prepared by this method.  相似文献

Charged pion production in high energy π-π- interactions is studied in the pT region of 1–2 GeV/c. The characteristics of pion production in π-π- interactions are compared with those in π-p and pp interactions. The pT dependence of pion production in these reactions follows a systematic trend which indicates that high-pT pion production for ππ interactions as well as for πp and pp interactions proceeds via quark-quark scattering as predicted by QCD.  相似文献
