Synthesis of a novel ether-bridged GM3-lactone analogue as a target for an antibody-based cancer therapy

We describe herein the synthesis of a new analogue of the GM3-lactone containing a cyclic ether moiety. The ether moiety was chosen as a replacement for the regular lactone group since it shows high resemblance with the lactone and is completely stable under biological conditions. The cyclic ether moiety was formed by reduction of the corresponding lactone to give the lactol followed by formation of the S,O-hemiacetal and hydrogenation. In addition, we have prepared haptens with a hexanoic acid moiety, which can be used for the preparation of poly- and monoclonal antibodies after binding to BSA or KLH. This is the first example of an analogue of the GM3-lactone which is stable under hydrolytic conditions in vitro and probably also in vivo. Reaction of lactone 18 with a Red/Al derivative led to the lactol 19 which was transformed into the S,O-hemiacetal 20 using 2,2'-bis(pyridinium) disulfide in quantitative yield. Hydrogenation with Raney Nickel gave 21 from which after removal of the protecting group at C-1a the trichloroacetimidate 25 was prepared. Reaction with azidosphingosine to give 26 followed by reduction of the azido group with NHEt3+[(PhS)3Sn], acylation with stearic acid using EDC and removal of the protecting groups led to the desired ether analogue of GM3 lactone 4. In addition the trichloroacetimidate 25 was glycosidated with 6-hydroxyhexanoic acid methyl ester, which was deprotected to give 29. The compound will be used for the preparation of poly- and monoclonal antibodies after coupling with BSA and KLH.     

Enantiomer separation by gas chromatography on cyclodextrin chiral stationary phases

Fused silica capillary columns coated with several alkyl or acyl cyclodextrin derivatives, especially those of α- and β-cyclodextrins, are suitable for the enantiomer separation of a wide variety of volatile compounds of different molecular size and functionality. Positional isomers and more than 250 pairs of optical isomers have been resolved, including chiral hydrocarbons, acetals, ethers, epoxides, carbonates, lactones, esters, acids, ketones, aldehydes, alcohols, halocarbons, and also nitrogen-and sulfur-containing compounds. The physical properties of the cyclodextrin derivatives, even those obtained as viscous fluids, could be improved by dissolving them in polysiloxane liquid phases commonly used for GLC.     

Endomorphin analogues with balanced affinity for bothμ- andδ-opioid receptors

Analogues of endomorphin and tripeptidcs modified at positions 4 and 3,respectively,with various phenylalanine analogues were synthesized and their affinities for opioid receptors were evaluated.Most of the peptides exhibited potentμ-receptor affinity and selectivity,among them,compound 7(Dmt-Pro-Tmp-Tmp-NH_2) exhibited potent affinity for bothμ-andδ-receptors (K_iμ= 0.47 nmol/L,K_iδ= 1.63 nmol/L).     

Effective one-dimensional models from matrix product states

In this paper we present a method for deriving effective one-dimensional models based on the matrix product state formalism. It exploits translational invariance to work directly in the thermodynamic limit. We show, how a representation of the creation operator of single quasi-particles in both real and momentum space can be extracted from the dispersion calculation. The method is tested for the analytically solvable Ising model in a transverse magnetic field. Properties of the matrix product representation of the creation operator are discussed and validated by calculating the one-particle contribution to the spectral weight. Results are also given for the ground state energy and the dispersion.
