Raffinose [or O‐α‐D‐galactopyranosyl‐(1→6)‐α‐D‐glucopyranosyl‐(1→2)‐β‐D‐fructofuranoside] pentahydrate, C18H32O16·5H2O, (I), and three lower hydrates, namely the 4.433‐, (II), 4.289‐, (III), and 4.127‐hydrated, (IV), forms, obtained in the course of the dehydration of (I), have been studied. The unit cells in the space group P212121 are of similar dimensions for all the crystals. The conformation of the raffinose molecules remains almost the same across the four crystal structures. The raffinose molecules are linked into a three‐dimensional hydrogen‐bonded network involving all the –OH groups, the ring and glycosidic O atoms, and the water molecules. Six water sites were identified in the structures of (II), (III) and (IV), of which W1, W4 and W6 (W = water) are partially occupied with their populations coupled. W1, W4 and one of the –OH groups of the galactose ring form an infinite hydrogen‐bonding chain around a 21 axis parallel to the a axis (denoted chain A), and W6 and the same –OH group form a similar chain (chain A′) disordered with chain A. The occupancy ratio of chain A to chain A′ for N‐hydrates (N is a hydration number between 4 and 5) is (N− 4):(5 −N). The transformation of chain A to chain A′ as part of the dehydration process has little effect on the rest of the structure. We developed a highly reproducible method for planar lipid bilayer reconstitution using a microfluidic system made of a polymethyl methacrylate (PMMA) plastic substrate. Planar lipid bilayers are formed at apertures, 100 microm in diameter, by flowing lipid solution and buffer alternately into an integrated microfluidic channel. Since the amount and distribution of the lipid solution at the aperture determines the state of the lipid bilayer, controlling them precisely is crucial. We designed the geometry of the fluidic system so that a constant amount of lipid solution is distributed at the aperture. Then, the layer of lipid solution was thinned by applying an external pressure and finally became a bilayer when a pressure of 200-400 Pa was applied. The formation process can be simultaneously monitored with optical and electrical recordings. The maximum yield for bilayer formation was 90%. Using this technique, four lipid bilayers are formed simultaneously in a single chip. Finally, a channel current through gramicidin peptide ion channels was recorded to prove the compatibility of the chip with single molecule electrophysiology. 相似文献

A three-dimensional magnetic material [{CoII(pyrimidine)(H2O)}2{CoII(H2O)2}{WV(CN)8}2](pyrimidine)2. 2H2O is prepared. This compound exhibits a charge-transfer-induced spin transition with a large thermal hysteresis loop of 90 K. Irradiating with light causes the low-temperature phase to exhibit a spontaneous magnetization with a Curie temperature of 40 K and a magnetic hysteresis loop with a coercive field of 12 000 G, which is the highest value reported for a photomagnet. The observed photoinduced magnetization is due to the charge-transfer phase transition from the {CoIIhs(S = 3/2)}{CoIIIls(S = 0)}2-NC-{WIV(S = 0)}2 phase to the {CoIIhs(S = 3/2)}3-NC-{WV(S = 1/2)}2 phase by the irradiation. 相似文献

The synthesis of D- and L-selenomethionine labeled with ?2Se and three deuteriums at Se-methyl group (D- and L-[2H?, ?2Se]selenomethionine) was described. D- And L-[2H?, ?2Se]selenomethionine were prepared by condensation of (R)- and (S)-2-amino-4-bromobutylic acid with lithium [2H?, ?2Se]methaneselenolate, which was prepared from metal (82)Se and [2H?]methyl iodide. The optical purities of D- and L-[2H?, ?2Se]selenomethionine were determined by HPLC with a chiral stationary phase column and were found more than 99% ee. The chemical ionization mass spectra showed that the molecular related ion for N-isobutyloxycarbonyl ethyl ester derivatives of [2H?, ?2Se]selenomethionine did not overlap with the m/z values known from that of non-labeled selenomethionine. 相似文献
