The versatility of an on-chip graphene oxide (GO) aptasensor was successfully confirmed by the detection of three different proteins, namely, thrombin (TB), prostate specific antigen (PSA), and hemagglutinin (HA), simply by changing the aptamers but with the sensor composition remaining the same. The results indicate that both DNA and RNA aptamers immobilized on the GO surface are sufficiently active to realize an on-chip aptasensor. Molecular selectivity and concentration dependence were investigated in relation to TB and PSA detection by using a dual, triple, and quintuple microchannel configuration. The multiple target detection of TB and PSA on a single chip was also demonstrated by using a 2 × 3 linear-array GO aptasensor. This work enables us to apply this sensor to the development of a multicomponent analysis system for a wide variety of targets by choosing appropriate aptamers.  相似文献

Due to the characteristic absorption peaks in the IR region, various molecules can be used as a matrix for infrared matrix‐assisted laser desorption/ionization (IR‐MALDI). Especially in the 6–7 µm‐band IR region, solvents used as the mobile phase for liquid chromatography have absorption peaks that correspond to their functional groups, such as O–H, CO, and CH3. Additionally, atmospheric pressure (AP) IR‐MALDI, which is applicable to liquid‐state samples, is a promising technique to directly analyze untreated samples. Herein we perform AP‐IR‐MALDI mass spectrometry of a peptide, angiotensin II, using a mid‐IR tunable laser with a tunable wavelength range of 5.50–10.00 µm and several different matrices. The wavelength dependences of the ion signal intensity of [M + H]+ of the peptide are measured using a conventional solid matrix, α‐cyano‐4‐hydroxycinnamic acid (CHCA) and a liquid matrix composed of CHCA and 3‐aminoquinoline. Other than the O–H stretching and bending vibration modes, the characteristic absorption peaks are useful for AP‐IR‐MALDI. Peptide ions are also observed from an aqueous solution of the peptide without an additional matrix, and the highest peak intensity of [M + H]+ is at 6.00 µm, which is somewhat shorter than the absorption peak wavelength of liquid water corresponding to the O–H bending vibration mode. Moreover, long‐lasting and stable ion signals are obtained from the aqueous solution. AP‐IR‐MALDI using a 6–7 µm‐band IR tunable laser and solvents as the matrix may provide a novel on‐line interface between liquid chromatography and mass spectrometry. Temperature-dependent electron transport in a metallic surface superstructure, Si(111)sqrt[3] x sqrt[3]-Ag, was studied by a micro-four-point probe method and photoemission spectroscopy. The surface-state conductivity exhibits a sharp transition from metallic conduction to strong localization at approximately 150 K. The metallic regime is due to electron-phonon interaction while the localization seemingly originates from coherency of electron waves. Random potential variations, caused by Friedel oscillations of surface electrons around defects, likely induce strong carrier localization.  相似文献

PURPOSE: The aim of this study was to evaluate the frequency and magnetic resonance imaging (MRI) features of clinically benign, small (<2 cm) hyperintense hepatic lesions in the cirrhotic liver on T1-weighted MR images seen at serial MRI. MATERIALS AND METHODS: This study included 189 patients with cirrhosis, who underwent hepatic MRI more than twice with an interval of at least 12 months. The initial MR images were reviewed for the presence of small hyperintense lesions on T1-weighted images. The size, location and signal intensity on T2-weighted images as well as enhancement patterns of the corresponding lesions were recorded. RESULTS: On the initial T1-weighted MR images, 43 small hyperintense hepatic lesions were detected in 23 (12%) of 189 patients. Twelve (28%) of 43 lesions showed early enhancement and were pathologically diagnosed as hepatocellular carcinoma (HCC) during the follow-up period. Thirty-one (72%) of 43 lesions showed no early enhancement with various signal intensity on T2-weighted images (hyperintensity=4, isointensity=20, hypointensity=7). Among these 31 lesions, 12 showed no interval change, while 11 disappeared (n=10) or decreased in size (n=1). In the remaining eight lesions, seven were diagnosed as HCC on the basis of pathologic confirmation or the interval growth. CONCLUSION: Small hyperintense hepatic lesions on T1-weighted magnetic resonance (MR) images without early enhancement on the arterial-phase contrast-enhanced dynamic studies in patients with cirrhosis usually showed no interval growth or disappeared during the serial MRI. These lesions with additional findings of iso- or hypointensity on the T2-weighted MR images without "washout effect" on the contrast-enhanced equilibrium-phase images may more frequently be clinically benign or hyperplastic nodules than HCCs.  相似文献

The cytotoxicity of a trivalent arsenic derivative (arsenite, AsIII) combined with arenobufagin or gamabufotalin was evaluated in human U-87 glioblastoma cells. Synergistic cytotoxicity with upregulated intracellular arsenic levels was observed, when treated with AsIII combined with arenobufagin instead of gamabufotalin. Apoptosis and the activation of caspase-9/-8/-3 were induced by AsIII and further strengthened by arenobufagin. The magnitude of increase in the activities of caspase-9/-3 was much greater than that of caspase-8, suggesting that the intrinsic pathway played a much more important role in the apoptosis. An increase in the number of necrotic cells, enhanced LDH leakage, and intensified G2/M phase arrest were observed. A remarkable increase in the expression level of γH2AX, a DNA damage marker, was induced by AsIII+arenobufagin. Concomitantly, the activation of autophagy was observed, suggesting that autophagic cell death associated with DNA damage was partially attributed to the cytotoxicity of AsIII+arenobufagin. Suppression of Notch signaling was confirmed in the combined regimen-treated cells, suggesting that inactivation of Jagged1/Notch signaling would probably contribute to the synergistic cytotoxic effect of AsIII+arenobufagin. Given that both AsIII and arenobufagin are capable of penetrating into the blood–brain barrier, our findings may provide fundamental insight into the clinical application of the combined regimen for glioblastoma.  相似文献

Gel-sol transition of sodium type gellan solutions with and without salts is studied by dynamic viscoelastic measurements and differential scanning calorimetry (DSC). Mechanical spectra show that gellan aqueous solutions behave as an entangled polymer solution in the concentration range around 2 wt.-% at temperatures >15°C and as a weak gel below this temperature. Concentrated solutions (> 3 wt.-%) show a true gel behavior below 30°C. The two step transition is observed for 2∼3 wt.-% gellan aqueous solutions in thermal scanning rheological (TSR) measurements; the transition at a higher temperature is attributed to a coil-helix transition whilst the transition at a lower temperature is attributed to sol-gel transition. The transition observed in dilute solutions of gellan is attributed to the coil-helix transition whilst the sol-gel transition occurs simultaneously with coil-helix transition in more concentrated solutions (>3 wt.-%). The sol-gel transition temperature shifts to higher temperatures with increasing concentration of the added salts. Junction zones formed in the presence of divalent cations are far more heat resistant than those with monovalent cations judging from both DSC and TSR, however, the possibility of the formation of junction zones by covalent bonds or by ionic bonds is excluded.  相似文献

Objective: Social participation is an essential component of active aging. Physical dysfunction is restriction of social participation, but it is inconclusive that improvement of physical function contributes to promote social participation. Therefore, understanding the other factor that moderates the association between physical dysfunction and social participation is important, and social network (i.e., ties with family and friends) may be a key factor. The aims of this study were to investigate the association between physical function and frequency of social participation, with social network as a moderator, and to examine the gender differences on the relationships. Methods: We conducted a cross-sectional study among 287 community-dwelling older adults. We asked how often they participated in social groups in a week to measure frequency of social participation. Physical function and social network were measured by using the modified version of Short Physical Performance Battery for community-dwelling older adults and the abbreviated Lubben Social Network Scale, respectively. To investigate the association, we performed a linear regression analysis. Results: After adjustment, a linear regression analysis showed interactions between physical function and social network on frequent social participation (β: −0.20, 95% confidence interval [CI]: −0.40 to −0.01). Furthermore, the same association was observed only in women (adjusted β: −0.33, 95% CI: −0.65 to −0.02). Conclusion: Our results suggested that social network moderates the association between physical function and social participation, and observed gender differences on the relationships. The findings of this study indicated the importance of multidimensional assessment and measures for improving social participation, not only physical function but also social network.  相似文献
