The gastrointestinal peptide glucose-dependent insulinotropic polypeptide (GIP1-42) is one of the incretin hormones regulating glucose-induced insulin secretion from the endocrine pancreas. GIP1-42 is a substrate of the circulating enzyme dipeptidyl peptidase IV, which removes the N-terminal peptide Tyr-Ala resulting in the inactive polypeptide GIP3-42. Hither to existing immunoassays do not enable a separate quantification of active and inactive forms, respectively. Therefore, we developed a highly specific and sensitive LC-MS assay for the identification and quantification of GIP1-42 and GIP3-42. Total GIP was immunoprecipitated from crude plasma samples using a C-terminally directed antibody. Thus, peptides were purified and concentrated prior to LC-MS analysis. The present immunoprecipitation-LC-MS assay enables the quantification of active and inactive GIP over a concentration range from 5 to 350 pmol/l in human plasma samples. Since this range covers the basal and postprandial levels of GIP the method is applicable to the determination of concentration changes and changes in the ratio of active and inactive forms of GIP in human plasma.  相似文献

Background

Previous work suggested that macrophage migration inhibitory factor (MIF) may be involved in bladder inflammation. Therefore, the location of MIF was determined immunohistochemically in the bladder, prostate, major pelvic ganglia, sympathetic chain, the L6-S1 dorsal root ganglia (DRG) and the lumbosacral spinal cord of the rat.

Results

In the pelvic organs, MIF immunostaining was prominent in the epithelia. MIF was widely present in neurons in the MPG and the sympathetic chain. Some of those neurons also co-localized tyrosine hydroxylase (TH). In the DRGs, some of the neurons that stained for MIF also stained for Substance P. In the lumbosacral spinal cord, MIF immunostaining was observed in the white mater, the dorsal horn, the intermediolateral region and in the area around the central canal. Many cells were intensely stained for MIF and glial fibrillary acidic protein (GFAP) suggesting they were glial cells. However, some cells in the lumbosacral dorsal horn were MIF positive, GFAP negative cells suggestive of neurons.

Conclusions

Therefore, MIF, a pro-inflammatory cytokine, is localized to pelvic organs and also in neurons of the peripheral and central nervous tissues that innervate those organs. Changes in MIF's expression at the end organ and at peripheral and central nervous system sites suggest that MIF is involved in pelvic viscera inflammation and may act at several levels to promote inflammatory changes.

The isomerizations preceding the metastable decompositions in the mass spectrometer of a number of [C6H12O]+˙ ions with the oxygen on the third carbon are characterized utilizing deuterium labeling. Hydrogens are transferred in these ions by three-, five- and six-membered ring rearrangements, with propensities determined by features of the individual reactions. Three-membered ring hydrogen transfers between α and β-carbons are preferred to all five-membered ring hydrogen transfers. However, six-membered ring hydrogen transfers take place to the apparent exclusion of three-membered ring hydrogen transfers to enol carbons when the products are of comparable stability. The low-energy [C6H12O]+˙ isomerizations characterized are predictable from the behavior of their lower homologs. It is concluded that the determinants of these reactions are the same as those of other highly reactive organic intermediates.  相似文献

In this paper, the origin of the conductivity of GaN films grown on ferroelectric materials was investigated using XPS, AES, and XRD analysis tools. Depth profiles confirmed the existence of impurities in the GaN film originating from the substrates. Bonding energy analysis from XPS and AES verified that oxygen impurities from the substrates were the dominant origin of the conductivity of the GaN film. Furthermore, Ga-rich GaN films have a greater chance of enhancing diffusion of lithium oxide from the substrates, resulting in more substrate phase separation and a wider inter-mixed region confirmed by XRD. Therefore, the direct GaN film growth on ferroelectric materials causes impurity diffusion from the substrates, resulting in highly conductive GaN films. Future work needs to develop non-conductive buffer layers for impurity suppression in order to obtain highly resistive GaN films.  相似文献

Onion soluble non-structural carbohydrates consist of fructose, glucose and sucrose plus fructooligosaccharides (FOS) with degrees of polymerisation (DP) in the range of 3-19. In onion, sugars and FOS are typically separated using liquid chromatography (LC) with acetonitrile (ACN) as a mobile phase. In recent times, however, the production of ACN has diminished due, in part, to the current worldwide economic recession. A study was therefore undertaken, to find an alternative LC method to quantify sugars and FOS from onion without the need for ACN. Two mobile phases were compared; the first taken from a paper by Vågen and Slimestad (2008) [3] using ACN mobile phase, the second, a newly reported method using ethanol (EtOH). The EtOH mobile phase eluted similar concentrations of all FOS compared to the ACN mobile phase. In addition, limit of detection, limit of quantification and relative standard deviation values were sufficiently and consistently lower for all FOS using the EtOH mobile phase. The drawback of the EtOH mobile phase was mainly the inability to separate all individual sugar peaks, yet FOS could be successfully separated. However, using the same onion extract, a previously established LC method based on an isocratic water mobile phase could be used in a second run to separate sugars. Although the ACN mobile phase method is more convenient, in the current economic climate a method based on inexpensive and plentiful ethanol is a valid alternative and could potentially be applied to other fresh produce types.In addition to the mobile phase solvent, the effect of extraction solvents on sugar and FOS concentration was also investigated. EtOH is still widely used to extract sugars from onion although previous literature has concluded that MeOH is a superior solvent. For this reason, an EtOH-based extraction method was compared with a MeOH-based method to extract both sugars and FOS. The MeOH-based extraction method was more efficacious at extracting sugars and FOS from onion flesh, eluting significantly higher concentrations of glucose, kestose, nystose and DP5-DP8.  相似文献

Catechol derivatives directly bonded to all-carbon quaternary stereocentres are prevalent in nature. An oxidative coupling strategy for the synthesis of this motif is described. Pivoting on the base-catalysed Michael addition of carbon-centred pro-nucleophiles to in situ generated ortho-benzoquinones, the method is broad in scope, high yielding and provides remarkably simple access to this challenging motif. The application of this methodology in the total synthesis of the crinane-type amaryllidaceae alkaloids (±)-powelline and (±)-buphanidrine is demonstrated and our efforts towards an enantioselective synthesis described.  相似文献

The photodynamic action of a novel photoactive polymer comprising covalently bound anthraquinone (AQ) moieties was evaluated after developing a methodology to reliably immobilize viable micro-organisms onto polymer film surfaces. The survival of Escherichia coli, Bacillus cereus (vegetative cells and spores), Fusarium oxysporum and Saccharomyces cerevisiae microbes inoculated on the surface of inert polymeric substrates was assessed to determine the effect of inoculum composition, drying rate and exposure to ultraviolet (UV-A) radiation. Their survival was highly dependent on microbial genus, with E. coli consistently displaying markedly shorter survival times than the other microbes, and B. cereus spores being the most resistant. Inoculation of the microbes onto the surface of the photoactive polymer films, followed by exposure to UV-A radiation, dramatically accelerated the inactivation of all microbial types studied compared with their survival on the surface of inert polymer substrates. Simultaneous exposure to both oxygen and UV-A radiation is required to affect cell survival, which is consistent with this effect most likely originating from the photoinduced production of singlet oxygen by the photoactive polymer. These results provide further compelling evidence that singlet oxygen produced exogenously by this photoactive polymeric substrate can successfully inactivate a broad spectrum of microbes on the substrate’s surface.  相似文献
