In heat exchange calorimetry, sample and reference vessels are fixed differentially in a water bath. The heat evolved in the sample vessel is exchanged freely with the ambient water. The accuracy and precision of the calorimetry depend on the temperature range of the drift and fluctuation in the bath water. In the present report, a simple temperature-control system with no empirical factors is proposed for the precision water bath. The heat exchange between the inside of the vessels and the ambient water is expressed by a differential equation or a Newtonian cooling equation. Similarly, the thermal behaviour of a water bath placed in a non-air-conditioned laboratory was also considered to be expressed by the same equation. The heat exchanged with the surroundings, the non-stationary heat flow from warm stirrer motors, and other thermal factors were considered to be part of the heat sources. Cooling water about 2°C lower than the set temperature was circulated in the water bath. The difference between the set temperature and the observed temperature was fed into a microcomputer every second. The latest four data points were used to fit a modified Newtonian equation by the common method of least-squares. The control input to the bath was calculated and given to the heater circuit after power amplification. The program written in basic was about 3.3 kbyte. The controlled range of drift and fluctuation was almost the same, or better, as that previously reported.  相似文献

The stability and replication of DNA containing self-pairs formed between unnatural nucleotides bearing benzofuran, benzothiophene, indole, and benzotriazole nucleobases are reported. These nucleobase analogues are based on a similar scaffold but have different hydrogen-bond donor/acceptor groups that are expected to be oriented in the duplex minor groove. The unnatural base pairs do not appear to induce major structural distortions and are accommodated within the constraints of a B-form duplex. The differences between these unnatural base pairs are manifest only in the polymerase-mediated extension step, not in base-pair stability or synthesis. The benzotriazole self-pair is extended with an efficiency that is only 200-fold less than a correct natural base pair. The data are discussed in terms of available polymerase crystal structures and imply that further modifications may result in unnatural base pairs that can be both efficiently synthesized and extended, resulting in an expanded genetic alphabet.  相似文献

A Ti-based oxysulfide, Sm(2)Ti(2)S(2)O(5), was studied as a visible light-driven photocatalyst. Under visible light (440 nm < or = lambda < or = 650 nm) irradiation, Sm(2)Ti(2)S(2)O(5) with a band gap of approximately 2 eV evolved H(2) or O(2) from aqueous solutions containing a sacrificial electron donor (Na(2)S-Na(2)SO(3) or methanol) or acceptor (Ag(+)) without any noticeable degradation. This oxysulfide is, therefore, a stable photocatalyst with strong reduction and oxidation abilities under visible-light irradiation. The electronic band structure of Sm(2)Ti(2)S(2)O(5) was calculated using the plane-wave-based density functional theory (DFT) program. It was elucidated that the S3p orbitals constitute the upper part of the valence band and these orbitals make an essential contribution to the small band gap energy. The conduction and valence bands' positions of Sm(2)Ti(2)S(2)O(5) were also determined by electrochemical measurements. It indicated that conduction and valence bands were found to have satisfactory potentials for the reduction of H(+) to H(2) and the oxidation of H(2)O to O(2) at pH = 8. This is consistent with the results of the photocatalytic reactions.  相似文献

With the increasing demand for novel devices with optical applications the search for new materials to data store and process becomes a priority. By introducing blends, tailor made properties and low cost give added advantage. Miscibility is an essential requirement for a new material, this research thus involves miscibility studies of poly(4‐(N‐(2‐methacryloyloxyethyl)‐N‐ethylamino)‐4′‐nitroazobenzene)90‐co‐(methyl methacrylate)10, (azobenzene derivative) with polymethyl‐methacrylate (PMMA), polyvinylacetate (PVAc) and polyvinylchloride (PVC) prepared in tetrahydrofuran (THF), and/or dimethylformamide (DMF) and/or dichloromethane (CH2Cl2). The glass transitions, solvent and varying molecular weight effect were investigated, since these all primarily influence the miscibility. THF was found to encourage miscibility at specific compositions of PVAc and PVC blends. However, with CH2Cl2 and DMF immiscibility is encouraged. The Fox–Flory equation was applied to the blends analyzing the PVC blends in DMF as deviations from ideality. Different molecular weights of PMMA were identified as immiscible regardless of solvent. PMMA's lower solubility in THF and CH2Cl2 compared to the azobenzene derivative, displayed the existence of PMMA islands. In all blends the favorable and unfavorable interactions between polymer–solvent–polymer systems are considered. Kynurenic acid (KYNA), an endogenous antagonist of ionotropic glutamate and α7 nicotinic receptors, was fluorometrically determined by column-switching high-performance liquid chromatography (HPLC) with fluorescence detection. The HPLC system consists of two octadecyl silica (ODS) columns, both of which are connected with an anion-exchange column (trapping column). Following sample injection onto the HPLC column, KYNA was separated on the first ODS column with a mobile phase of H2O/acetonitrile (95/5) containing 0.1% acetic acid. The peak fraction of KYNA was trapped on the anion-exchange column by changing the position of a six-port valve and then introduced into the second ODS column. Subsequently, KYNA was detected fluorometrically as a fluorescence complex formed with zinc ion which was pumped constantly. Instrumental limit of detection was approximately 0.16 nM, which corresponded to 8.0 fmol (per 50 μl injection, signal to noise ratio 3), and the limit of quantification was 0.53 nM (signal to noise ratio 10). Intra- and inter-day relative standard deviations were 1.1-3.9% (n = 3) and 3.0-5.3% (n = 3), respectively. The peak of KYNA in rat plasma was clearly detected by the proposed column-switching HPLC system after a facile pretreatment procedure. Intra- and inter-day relative mean errors were −1.6-1.4% (n = 3) and −2.4 to −0.4% (n = 3), respectively, with a satisfactory precision (within 5.0%). A calibration curve for the determination of KYNA showed a good linearity (r2 > 0.999) in the range of 25-200 nM. The KYNA concentrations in the plasma of male Sprague-Dawley rats (8-week-old) were 44 ± 5.5 nM (mean ± S.E., n = 5). In ketamine-treated rats, which are animal models of schizophrenia, the plasma KYNA concentrations were significantly increased compared with those in the control rats (p < 0.05).  相似文献
