The accurate three-dimensional (3D) velocity field measurement technique has been receiving large attention in the study of microfluidics. DHM-PTV technique was developed by combining the digital holographic microscopy and particle tracking velocimetry technique. DHM-PTV is an ideal method for measuring three-component-three-dimensional (3C-3D) velocity field in a microscale flow with a fairly good spatial resolution. The advances in the DHM-PTV technique enable the measurement of various microscale flows, such as transport of red blood cells in a microtube and 3D flows in microfluidic devices. DHM-PTV is also applied in studying the motile behavior of swimming microorganisms. DHM-PTV would play an important role in ascertaining the undiscovered basic physics in various microscale and biofluid flow phenomena. In the current study, the basic principle of the DHM-PTV technique and its typical applications to microscale flows are introduced and discussed.  相似文献

The interactions between femtosecond (fs) laser pulses and a thin Au film deposited on a silica glass substrate were systematically investigated based on experimental data. Different structures, including microholes, nanoholes, and nanobumps, are obtained when pulses with different energies are incident on the surface of a gold film. The experimental results are discussed according to specific experimental parameters. Two physical models were constructed in order to explain the experimental results. The formation of nanoholes in a silica substrate is attributed to etching by higher order harmonic generations (HHG) when the femtosecond laser pulse interacts with the generated plasma layer, while the formation of nanobumps on the surface of an Au film is attributed to the elastic and plastic characteristics of the metal film under laser pulse irradiation.  相似文献

Multichannel and omnidirectional transmission can be realized in one-dimensional periodic metamaterial layers, where double-negative materials and single-negative materials are alternately stacked. The proposed structures can produce as many resonance transmission modes as desired by adjusting the number of the periods. Such transmission modes with perfect transmission are robust against incident angles and polarizations. The frequencies and frequency intervals of these modes can be tuned by changing the thicknesses of the metamaterial layers. Composite right/left-handed transmission lines with lumped elements (capacitors and inductors) were used to verify the characteristics of the considered system. Our findings provide an efficient way to select useful multiple channel signals from all directions, and it is useful in optoelectronic device applications.  相似文献

In our study, the physical properties of (TiO2) x (SiO2)1?x , including band-gap, band-offset, and thermal stability and the electrical properties of band-engineered SiO2/(TiO2) x (SiO2)1?x tunnel barrier stacks, including the tunneling current and charge-trapping characteristics for applications to nonvolatile memory devices were investigated. It was observed that the band-gap and band-offset of (TiO2) x (SiO2)1?x can be controlled by adjustment in the composition of the (TiO2) x (SiO2)1?x films. Ti-silicate film with TiO2:SiO2 cycle ratio of 1:5 was maintained in an amorphous phase, even after annealing at 950 °C. The tunneling current of the band-engineered SiO2/(TiO2) x (SiO2)1?x stacked tunnel barrier was larger than that of a single SiO2 barrier under a higher external bias, while the tunneling current of a SiO2/(TiO2) x (SiO2)1?x stacked tunnel barrier under a lower external bias was smaller. Charge-trapping tests showed that the voltage shift for SiO2/(TiO2) x (SiO2)1?x is slightly larger than that for single SiO2.  相似文献

Wild ginseng has better pharmacological effects than cultivated ginseng. However, its industrialization is limited by the inability to grow wild ginseng on a large scale. Herein, we demonstrate how to optimize ginseng production through cultivation, and how to enhance the concentrations of specific ginsenosides through fermentation. In the study, we also evaluated the ability of fermented cultured wild ginseng root extract (HLJG0701-β) to inhibit acetylcholinesterase (AChE), as well as its neuroprotective effects and antioxidant activity. In in vitro tests, HLJG0701-β inhibited AChE activity and exerted neuroprotective and antioxidant effects (showing increased catalyst activity but decreased reactive oxygen species concentration). In in vivo tests, after HLJG0701-β was orally administered at doses of 0, 125, 250, and 500 mg/kg in an animal model of memory impairment, behavioral evaluation (Morris water maze test and Y-maze task test) was performed. The levels of AChE, acetylcholine (ACh), blood catalase (CAT), and malondialdehyde (MDA) in brain tissues were measured. The results showed that HLJG0701-β produced the best results at a dose of 250 mg/kg or more. The neuroprotective mechanism of HLJG0701-β was determined to involve the inhibition of AChE activity and a decrease in oxidative stress. In summary, both in vitro and in vivo tests confirmed that HJG0701-β administration can lead to memory improvement.  相似文献

We have produced nanophase metal clusters, (Fe)n, (Cr)n, (Mo)n and (W)n, by multiphoton decomposition of the corresponding metal carbonyls with a 10.6 μm CO2 laser in the presence of Ar and SF6. The size distribution was narrow and the average diameter was 6, 3.5, 2 and 1 nm for Fe, Cr, Mo and W clusters, respectively. The structure was found to be bcc for both Fe and Cr clusters, fcc for Mo clusters, and amorphous for W clusters (note that all the bulk metals have bcc structure). Considering the cluster sizes (9630, 1870, 230 and 30 for Fe, Cr, Mo and W clusters, respectively) estimated from their average diameters, it is likely that there exists a structural transition from fcc to bulk bcc with increasing cluster size in these metal clusters.  相似文献

Abnormally increased resorption contributes to bone degenerative diseases such as Paget’s disease of bone (PDB) through unclear mechanisms. Recently, the optineurin (OPTN) gene has been implicated in PDB, and global OPTN knockout mice (Optn−/−) were shown to exhibit increased formation of osteoclasts (osteoclastogenesis). Growing evidence, including our own, has demonstrated that intracellular reactive oxygen species (ROS) stimulated by receptor activator of nuclear factor kappa-B ligand (RANKL) can act as signaling molecules to promote osteoclastogenesis. Here, we report that OPTN interacts with nuclear factor erythroid-derived factor 2-related factor 2 (NRF2), the master regulator of the antioxidant response, defining a pathway through which RANKL-induced ROS could be regulated for osteoclastogenesis. In this study, monocytes from Optn−/− and wild-type (Optn+/+) mice were utilized to differentiate into osteoclasts, and both qRT-PCR and tartrate-resistant acid phosphatase (TRAP) staining showed that the Optn−/− monocytes exhibited enhanced osteoclastogenesis compared to the Optn+/+ cells. CellROX® staining, qRT-PCR, and Western blotting indicated that OPTN deficiency reduced the basal expression of Nrf2, inhibited the expression of NRF2-responsive antioxidants, and increased basal and RANKL-induced intracellular ROS levels, leading to enhanced osteoclastogenesis. Coimmunoprecipitation (co-IP) showed direct interaction, and immunofluorescence staining showed perinuclear colocalization of the OPTN-NRF2 granular structures during differentiation. Finally, curcumin and the other NRF2 activators attenuated the hyperactive osteoclastogenesis induced by OPTN deficiency. Collectively, our findings reveal a novel OPTN-mediated mechanism for regulating the NRF2-mediated antioxidant response in osteoclasts and extend the therapeutic potential of OPTN in the aging process resulting from ROS-triggered oxidative stress, which is associated with PDB and many other degenerative diseases.Subject terms: Mechanisms of disease, Stress signalling  相似文献

In the pea plant ( Pisum sativum ), compounds that intercalate into DNA induce the production of ∼20 major proteins similar to the pattern induced during nonhost disease resistance to the bean fungal pathogen, Fusarium solani f.sp. phaseoli . The pea phytoalexin, pisatin, as well as RNA homologous to several disease-resistance response (DRR) genes accumulate following treatment with these compounds. Psoralen was chosen to characterize this interaction further because it intercalates into DNA and, following irradiation with 365 nm UV light (UV365), forms covalent bonds with pyrimidines on either or both strands of DNA. This produces monoadducts or cross-links, respectively. Dose experiments showed that 60 μg/mL 4'-aminomethyl-4,5',8-trimethylpsoralen followed by 18 J/cm2 UV365 was sufficient to produce an accumulation of pisatin similar to that produced in response to the fungus. Under these inducing conditions, there was an average of 0.19 adducts per kb of pea genomic DNA. The accumulation of pisatin and the RNA of several DRR genes by psoralen required photoactivation, which suggests that covalent binding to DNA was necessary for induction. As the promoters of several putative fungal-induced pea genes contain long stretches of d(AT)n, which is the preferred psoralen photobinding site, restriction fragments spanning DRR genes were examined after in vivo psoralen treatment. The rate of crosslinking was compared between fungal-induced and noninduced genes using a modified Southern blot analysis. Implications of the induction of the DRR due to psoralen binding are discussed.  相似文献
