Mechanistic aspects of aldehyde and imine electro-reduction in a liquid–liquid carbon nanofiber membrane microreactor

A simple and electrolyte-free ion-transfer electrosynthesis micro-reactor system (volume 100 μL, up to 10 mg batches) for processes at liquid–liquid interfaces is developed and demonstrated for the reduction of aldehydes and imines. These cathodic reactions occur at an amphiphilic carbon nanofiber membrane accompanied by proton cation transfer from an aqueous phase into an organic phase.     

Association and Dissociation of Grignard Reagents RMgCl and Their Turbo Variant RMgCl⋅LiCl

Grignard reagents RMgCl and their so‐called turbo variant, the highly reactive RMgCl?LiCl, are of exceptional synthetic utility. Nevertheless, it is still not fully understood which species these compounds form in solution and, in particular, in which way LiCl exerts its reactivity‐enhancing effect. A combination of electrospray‐ionization mass spectrometry, electrical conductivity measurements, NMR spectroscopy (including diffusion‐ordered spectroscopy), and quantum chemical calculations is used to analyze solutions of RMgCl (R=Me, Et, Bu, Hex, Oct, Dec, iPr, tBu, Ph) in tetrahydrofuran and other ethereal solvents in the absence and presence of stoichiometric amounts of LiCl. In tetrahydrofuran, RMgCl forms mononuclear species, which are converted into trinuclear anions as a result of the concentration increase experienced during the electrospray process. These trinuclear anions are theoretically predicted to adopt open cubic geometries, which remarkably resemble structural motifs previously found in the solid state. The molecular constituents of RMgCl and RMgCl?LiCl are interrelated via Schlenk equilibria and fast intermolecular exchange processes. A small portion of the Grignard reagent also forms anionic ate complexes in solution. The abundance of these more electron‐rich and hence supposedly more nucleophilic ate complexes strongly increases upon the addition of LiCl, thus rationalizing its beneficial effect on the reactivity of Grignard reagents.     

Stark Hole-Burning Spectroscopy of Cresylviolet Perchlorate in Amorphous Hosts

Stark hole-burning spectroscopy is used to investigate the effective dipole moment change of cresylviolet perchlorate (CVP) in various glass and polymer hosts such as ethanol:methanol (EM), polyvinyl alcohol (PVA), poly (2-hydroxyethyl) methacrylate (PHEMA), polyvinylbutyral (PVB), and formamide. The strong correlation between effective dipole moment change of the guest molecule and the holeburning efficiencies of the host matrices illustrates the sensitivity of the dipole moment change as a direct measure of guest-host interactions. Hole-burning is found to be more efficient as the dipole induced reaction field increases. This relationship is discussed in terms of the unusual hole-burning mechanism suggested for this molecule. The effective dipole moment change of cresylviolet perchlorate ranges from 0.14 to 0.59 Debye.     

Plate-based diversity subset screening: an efficient paradigm for high throughput screening of a large screening file

The screening files of many large companies, including Pfizer, have grown considerably due to internal chemistry efforts, company mergers and acquisitions, external contracted synthesis, or compound purchase schemes. In order to screen the targets of interest in a cost-effective fashion, we devised an easy-to-assemble, plate-based diversity subset (PBDS) that represents almost the entire computed chemical space of the screening file whilst comprising only a fraction of the plates in the collection. In order to create this file, we developed new design principles for the quality assessment of screening plates: the Rule of 40 (Ro40) and a plate selection process that insured excellent coverage of both library chemistry and legacy chemistry space. This paper describes the rationale, design, construction, and performance of the PBDS, that has evolved into the standard paradigm for singleton (one compound per well) high-throughput screening in Pfizer since its introduction in 2006.     

Tetrathiafulvalene-annulated dipyrrolylquinoxaline: the effect of fluoride on its optical and electrochemical behaviors

A tetrathiafulvalene donor has been annulated to 2,3-di(1H-2-pyrrolyl)quinoxaline affording a new chemosensor 1, which shows a unique optical selectivity and reactivity for the fluoride ion over other anions in CH₂Cl₂ leading to a colorimetric response. Electrochemical polymerization of 1 occurred in the presence of fluoride.     

Regiospecific synthesis of cepanolide,a cancer chemoprotective micronutrient found in green onions

The naturally occurring γ-hydroxy-β-sulfanylbutenolide cepanolide and a range of new analogues were synthesized in concise, regiospecific manner through the combined use of 2-silyloxyfuran oxyfunctionalization and tandem thio-Michael addition/elimination.
