With the objective of developing small molecule based probes for proteins, interactions of polyhydroxyl-substituted squaraine dye (SQ) with bovine serum albumin (BSA) have been investigated by absorption, steady-state and time-resolved fluorescence, circular dichroism (CD), cyclic voltammetry (CV), 1H NMR, scanning electron, and tapping mode atomic force microscopic techniques. Increase in addition of BSA resulted in increase in absorbance and fluorescence quantum yields (80-fold) of SQ, along with significant bathochromic shifts in the absorption and fluorescence maxima. Half-reciprocal analysis of the absorption data gave a 1:1 stoichiometry for the complex between BSA and SQ with high association (Kass) constant of (1.4 +/- 0.1) x 106 M-1 and change in free energy of -35 kJ/mol. The complex formation was further confirmed by observation of induced CD signal corresponding to the SQ chromophore at 610 nm, upfield shift (about Deltadelta 0.1 ppm) of aromatic protons of SQ in 1H NMR spectra, and decrease in current intensity (CV) of SQ when bound to BSA. The picosecond time-resolved fluorescence studies indicated that the BSA-SQ complex exhibits biexponential decay with significantly enhanced lifetimes of 0.5 and 1.5 ns when compared to the lifetime of SQ (tau = 121 ps) in the absence of BSA. Employing displacement cum fluorimetry using site-specific binding ligands, such as dansylproline and dansylamide, indicated that SQ binds with protein selectively at site II involving hydrophobic, hydrogen bonding, and electrostatic interactions. The uniqueness of this molecular system is that it interacts with BSA selectively at site II and signals the binding event through dual mode recognition of "visual color" change and "turn on" fluorescence mechanism.  相似文献

A seemingly ipso-like nucleophilic substitution of the upper rim of p-tert-butylcalix[4]arene is accomplished by an indirect method involving calix[4]arene derived bis(spirodienone). This method not only provides both mono and 1,3-diaryloxy calixarenes but also enables the synthesis of upper rim monothio substituted calix[4]arenes. A modification of the methodology can be successfully extended for the selective synthesis of mono- and 1,3-diquinone calix[4]arenes having free hydroxyl groups at the lower rim, in fewer steps.  相似文献

We have reported earlier the observation of a self-written waveguide inside a bulk methylene blue sensitized poly (Vinyl Alcohol) /Acrylamide photopolymer material [1]. In this work we develop a theoretical model to explain the experimental observation. We reduce the governing equations for the propagation of a low power laser beam through such a medium into the form of nonlinear Schrodinger equation with a cubic-quintic nonlinearity and solve this equation using variational method. It is found that there is agreement between experimental and theoretical results.  相似文献

We theoretically and experimentally generate stationary crescent surface solitons pinged to the boundary of a microstructured vertical cavity surface emission laser by triggering the intrinsic cavity mode as a background potential. Instead of a direct transition from linear to nonlinear cavity modes, we demonstrate the existence of symmetry-breaking crescent waves without any analogs in the linear limit. Our results provide an alternative and general method to control lasing characteristics as well as to study optical surface waves.  相似文献

An automatic and efficient technique to analyze the uniformity of nanoscale images using wavelets, feature similarity index measure (FSIM) and fuzzy inference system is reported. It has been successfully tested on scanning electron micrographs of nanocrystalline silver thin films. Thin films are prepared using on‐axis and off‐axis pulse laser deposition (PLD) technique. It is found that the film prepared using on‐axis PLD is more uniformly distributed and has smoother texture compared with that of the off‐axis technique. In order to analyze the images quantitatively, they are transformed to the wavelet domain to extract the localized frequency variations and a uniformity measure is derived using a fuzzy inference system for quantitatively analyzing the uniformity of each image. The surface plot of the FSIM values of the image is found to be an efficient tool for nanoscientists to evaluate the smoothness of the thin film surfaces. The chemical nature of the polyphenols of cashew kernel testa has been determined. Testa contains tannins, which present large molecular complexity and has an ancient use as tanning agents. The use of tannins extracted from cashew testa, considered in many places as a waste, grants an extra value to the cashew. In this work we have analysed through high performance liquid chromatography, infrared spectroscopy (FT-IR) and thermo gravimetric analysis the average molecular weight, main functional groups and thermal properties of tannins extracted from Anacardium occidentale L. The results of these analyses are compared with the commercial grade tannic acid. The FT-IR spectra showed bands characteristic of C = C, C–C and OH bonds. This important bioactive compound present in the cashew nut kernel testa was suggested as an interesting economical source of antioxidants for use in the food and nutraceutical industry.  相似文献

The growth kinetics of an aerial bacterial colony on solid agar media was studied using laser induced fluorescence technique. Fluorescence quenching of Rhodamin B by the bacterial colony was utilized for the study. It is known that symmetric orbits in g* for any simple Lie algebra g are equipped with a Poisson pencil generated by the Kirillov-Kostant-Souriau bracket and the reduced Sklyanin bracket associated to the “canonical” R-matrix.
