Aminophenylboronic acid (APBA) has been immobilized on magnetic beads for the direct determination of glycoprotein by matrix assisted laser desorption/ionizaton time of flight mass spectrometry (MALDI-TOF-MS). An APBA layer was formed on the surface of carboxylic acid terminated magnetic beads by coupling with carbodiimide and subsequently reacted with an N-hydroxysuccinimide moiety. The immobilized APBA was identified by MALDI-TOF-MS without a matrix. Glycoproteins, such as HbA1c, fibrinogen, or RNase B were separated and desalted using APBA magnetic beads by simply washing the magnetic beads and then separating them by external magnet. Proteins can be identified by direct determination of proteins on beads on MALDI plate and confirmed again by peptide mass finger printing after digestion of proteins on magnetic beads by trypsin. Fluorescence image with a FITC tagging protein using confocal laser microscopy showed the difference of immobilization efficiency between glycoproteins and nonglycoproteins. The methods developed within this work allow the simple treatment and enrichment of glycoproteins as well as direct determination of proteins on beads by MALDI-TOF-MS. 相似文献

We report a novel vesicle formed by an amphiphilic CB[6] derivative, the surface of which can be easily modified via host-guest interactions by taking advantage of molecular cavities, readily accessible at the vesicle surface, and their strong affinity toward polyamines. Amphiphilic CB[6] derivative 1 synthesized by reaction between (allyloxy)12CB[6] and 2-[2-(2-methoxyethoxy)ethoxy]ethanethiol affords a vesicle that has been characterized by TEM, light scattering, and fluorescent dye entrapment experiments. Treatment of vesicle 1 with FITC (fluorescein isothiocyanate)-spermine conjugate ligand 2, in which spermine serves as a binding motif to CB[6] and FITC as a fluorescent tag, produced a surface-modified vesicle, which can be easily visualized by a confocal microscope. This result provides us with a new noncovalent, modular approach to the modification of vesicle surfaces. By treating the vesicle derived from the amphiphilic CB[6] with a tag-attached polyamine, we can easily decorate the surface of the vesicle with the tag. Sugar-decorated vesicles were prepared by this noncovalent method, and their interactions with concanavalin A (ConA) were studied. The binding constant of the vesicle decorated with mannose-spermidine conjugate 3 to ConA was measured to be approximately 3 x 104 M-1, which is almost 3 orders of magnitude higher than that of free ligand 3 to ConA (K = approximately 50 M-1). On the other hand, the binding constant of the vesicle coated with galactose-spermidine conjugate 4 to ConA was too small to be measured. These results illustrate the specific and multivalent interactions between the mannose-decorated vesicle and ConA. The ability for facile surface modification suggests many practical applications, including its use in targeted drug delivery and immunization. 相似文献

Electromagnetic interference (EMI) shielding materials of complex type of conductive polypyrrole (PPy) as an intrinsically conducting polymer and silver‐palladium (AgPd) metal compound coated on woven or non‐woven fabrics are synthesized. From dc conductivity and SEM photographs of PPy/fabric complexes, we discuss charge transport mechanism and the homogeneity of coating on the fabrics. The EMI shielding efficiency of PPy/fabric and AgPd/fabric complexes is in the range of 8 ~ 80 dB depending on the conductivity and the additional Ag vacuum evaporation. The highest EMI shielding efficiency of PPy/fabric complexes vacuum‐evaporated by Ag is ~80 dB, indicating potential materials for military uses. We propose that PPy/fabrics are excellent RF and microwave absorber because of the relatively high absorbance and low reflectance of the materials. Copyright © 2002 John Wiley & Sons, Ltd. 相似文献

and four disubstituted derivatives, (6,7-dihydroxycoumarin, 7-hydroxycoumarin-4-acetic acid, 7-methoxycoumarin-4-acetic acid,
and 7-hydroxy-4-methylcoumarin) were measured in the temperature range 35–50 °C and the pressure range 8.5–25 MPa. In general,
the substituted coumarin derivatives were less soluble than simple coumarin. It was also found that substitution at the C-4
position of coumarin tended to reduce the solubility more than substitution at the C-7 position. These solubility data are

Repetitive low dose thioacetamide (TA) treatment of hepatocytes was found to induce cells in G2 arrest. In the present study, an attempt was made to investigate alterations in expression of cell cycle regulators after G1 progression in the same repetitive low dose TA treated hepatocytes system and to define the determinators involved in G2 arrest. TA was daily administered intraperitoneally, with a dose of 50 mg/kg for 7 days. Expression levels of cyclin E and CDK2 were similar, increased at day 1 and reached a peak at day 2. And they recycled from day 3 reaching a second peak at day 5. Expression level of cyclin A was similar to p27(Kip1) and p57(Kip2) but not to CDK2 and increased to a peak level at day 2. Expression levels of cyclin B1 and cdc2 were similar although the cyclin B1 level was generally low, decreased from day 1 to basal levels at day 3 and persisted at a low level till day 7. The expression level of cyclin G1 was similar to p53 that peaked at day 3 and again at day 6 elevated over basal level. BrdU-labeled hepatocytic nuclei increased from 12 h, reached a peak at day 2, then decreased, and were not detectable from day 6. The number of PCNA-labeled nuclei increased immediately, peaked at day 2, and maintained till day 7. These results suggest that G2 arrest induced by repeated TA treatment might be p53-dependent, via activation of cyclin G1, rather than inhibition of cyclin B1- cdc2 complex, and inhibitors holding S phase progression might be p27(Kip1) and p57(Kip2). 相似文献

Following incubation of rutaecarpine, a new cyclooxygenase-2 inhibitor, with rat liver microsomes, the structures of the metabolites were characterized by liquid chromatography with tandem mass spectrometry. Nine metabolites corresponding to mono- or dihydroxylated rutaecarpine were formed. Characteristic product ions for the identification of rutaecarpine metabolites were observed at m/z 136, 158 and 286. The loss of water led to the fragment ion at m/z 286, indicating the hydroxylation of the aliphatic ring. The fragment ion at m/z 136 indicated the hydroxylated form of the phenyl group of the quinazolinone moiety, while that at m/z 158 indicated the hydroxylated form of the aromatic ring of the indole moiety. 相似文献

Efficient and reversible fluorescence modulation with excellent photo-stability was achieved from a sulfone form of diacetyl diarylethene, 1,2-bis(6-acetyl-2-methyl-1-benzothiophene-1,1-dioxide-3-yl)perfluorocyclopentene (DABTFO4). The DABTFO4 emits strong fluorescence in the closed-ring isomer even in the absence of extra fluorophores. The fluorescence quantum yield, fatigue resistance, and photo-cyclization yield of DABTFO4 were significantly improved compared with the unsubstituted 1,2-bis(2-methyl-1-benzothiophene-1,1-dioxide-3-yl)perfluorocyclopentene (BTFO4) and the sulfide analogue, 1,2-bis(6-acetyl-2-methyl-1-benzothiophene-3-yl)perfluorocyclopentene (DABTF6). 相似文献

The gelation behavior of aqueous solutions of poly(ethylene oxide-b-(DL-lactic acid-co-glycolic acid)-b-ethylene oxide) (PEO-PLGA-PEO) triblock copolymer containing short hydrophilic PEO end blocks is investigated using dynamic light scattering, rheology, small-angle neutron scattering (SANS), and differential scanning calorimetry (DSC). For polymer concentrations between 5 and 35 wt %, four distinct regions of the turbidity change depending on temperature were observed. Interestingly, in the turbid solution region, gel phase is formed for polymer concentrations above 14 wt % and an extremely slow relaxation was detected. In fact, a power law, which takes into account the dynamics of percolation clusters, dominates the correlation function. In rheological measurements, the local maximum in G' is observed at around the temperature of maximum turbidity. We further found that G" > G' and G' is highly dependent on frequency at the gel state implying viscoelastic characteristics, which is quite different from general concepts of gels, typically formed by the micellar packing. SANS profiles showing multiple peaks in the sol state rather than in the gel state as well as a DSC exotherm at the temperature of gels can also serve as the evidence of different gel states. Based upon the experimental data obtained in the present study, a new gelation mechanism induced by the macroscopic phase separation of triblock copolymers containing short hydrophilic PEO end blocks such as PEO-PLGA-PEO is proposed. The effect of the type ofhydrophobic middle blocks on the gelation is also discussed. 相似文献
